Aspects on gametogenesis, fertilization and embryogenesis of two deep-sea polychaetes from Eastern Atlantic cold seeps ArchiMer
Gaudron, S. M.; Hourdez, S.; Olu, Karine.
We investigated two gonochoristic species of annelid polychaetes (one siboglinid and one polynoid) from cold seeps that ranged from 525 m to 3300 m in depth (Guiness, Worm Hole and Regab pockmarks) on the Gabon and Congo continental margins (Gulf of Guinea). Different aspects of gametogenesis (oocyte diameter, presence of ovisac, spermatozoa shape, and fecundity), fertilization (in vitro fertilization experiments: IVF) and embryogenesis (cleavage rate) were studied. The sampled siboglinid was a new species of Lamellibrachia and the second population of this genus in the Eastern Atlantic. Mean oocyte diameter was about 100 µm and fully-grown primary oocytes were stored in an ovisac, as in other studied siboglinids. The...

Bovine conceptus of Bos indicus produced by somatic cell nuclear transfer and parthenogenesis present morphological variations since the blastocyst stage Arq. Bras. Med. Vet. Zootec.
Oliveira,F.D.; Sangalli,J.R.; Meirelles,F.V.; Perecin,F.; Silva Filho,P.P.C.; Watanabe,Y.F.; Miglino,M.A.; Assis Neto,A.C..
In cattle, embryo development is characterized by the appearance of two distinct cell layers, the trophectoderm and the inner cell mass. The latter will undergo differentiation to form the embryonic disc consisting of the epiblast and hypoblast. The aim of this study was to ultrastructurally characterize the bovine embryo from different in vitro production techniques, with emphasis on trophectoderm and inner cell mass cells. Bovine embryos on day 7 (conception = D1) of pregnancy, derived via in vitro production techniques, were fixed for light and transmission electron microscopy processing. Results suggested that embryos produced by nuclear transfer of somatic cells and parthenogenesis showed significant changes in macroscopic and microscopic structure....

Bovine embryo induces an anti-inflammatory response in uterine epithelial cells and immune cells in vitro: Possible involvement of interferon tau as an intermediator OAK
Talukder, Anup K.; Yousef, Mohamed S.; Rashid, Mohammad B.; Awai, Kensuke; Acosta, Tomas J.; Shimizu, Takashi; Okuda, Kiyoshi; Shimada, Masayuki; Imakawa, Kazuhiko; Miyamoto, Akio.
Recent observations suggest that the bovine uterus starts to react to the early embryo immediately after its arrival from the oviduct. The present study aimed to investigate the effect of the early developing embryo on the immune-related gene profile in bovine uterine epithelial cells (BUECs) in vitro, and to further examine the impact of conditioned media (CM), either from embryo-BUEC co-culture or embryo culture alone, on gene expression in peripheral blood mononuclear cells (PBMCs). First, BUECs were co-cultured with morulae (n = 10) for D5-D9 (D0 = IVF), and gene expression in BUECs was analyzed. Subsequently, PBMCs were cultured in CM from embryo-BUEC co-culture or D5-D9 embryo culture, and gene expression was...

The objective of this work was to evaluate the relationship between sperm head morphometry and chromatin packaging with the cleavage ratio and blastocyst formation during in vitro fertilization programs (IVF). Data of 38 IVFs using semen of zebu bulls with normal spermogram, carried out at two laboratories in Uberaba, Minas Gerais, Brazil, were used in this study. Sperm smears stained with toluidine blue and analyzed by computer image analysis were used to evaluate the morphometry and chromatin packaging. The packaging intensity and homogeneity of sperm chromatin were evaluated. The results made it possible to conclude that light variations in the head morphometry and chromatin packaging had little influence on the...

Effect of conditioned medium of mesenchymal stem cells on the in vitro maturation and subsequent development of mouse oocyte BJMBR
Ling,B.; Feng,D.Q.; Zhou,Y.; Gao,T.; Wei,H.M.; Tian,Z.G..
Mesenchymal stem cells (MSCs) secrete a variety of cytokines and growth factors in addition to self-renewal and multiple forms of differentiation. Some of these secreted bioactive factors could improve meiotic maturation in vitro and subsequent embryo developmental potential. The aim of the present study was to determine whether in vitro maturation (IVM) of mouse oocyte with or without cumulus cells could be improved by contact with conditioned medium (CM) of MSCs as well as the efficiency of CM to support follicular growth and oocyte maturation in the ovarian organ of mice cultured on soft agar. The developmental potential of matured oocyte was assessed by blastocyst formation after in vitro fertilization (IVF). Germinal...

Effect of Zona Incision by Piezo-Micromanipulator (ZIP) on In Vitro Fertilization in 21 Transgenic Mice Lines OAK
KAWASE, Yosuke; TACHIBE, Takanori; HANI, Toshio; TATEISHI, Hiromi; JISHAGE, Kou-ichi; SUZUKI, Hiroshi; 鈴木, 宏志.
Zona incision using a piezo-micromanipulator (ZIP) has been demonstrated to be effective for in vitro fertilization (IVF) using cryopreserved C57BL/6 spermatozoa. In this study, ZIP oocytes inseminated with frozen-thawed genetically modified C57BL/6J or FVB mice spermatozoa (21 lines) showed fertilization rates of 22-75% and live fetus rates of 8-49%. In 6 of the lines, the fertilization rates for oocytes compared with ZIP (42-75%) were significantly higher than that of nontreated oocytes (0-50%). Using only 90 oocytes for IVF with ZIP, 5 breeding pairs were produced from cryopreserved genetically modified mice spermatozoa. Our results indicate that application of the ZIP technique is...

El embrión extra : ética de vida, ética de parentesco y cryopreservación en las clínicas ecuatorianas de fertilización in-vitro (Dossier) Buscador Latinoamericano
Roberts, Elizabeth.
In this paper I trace two divergent understandings of personhood that manifest through the technology of embryo cryopreservation in Ecuadorian IVF clinics. Those who wish to cryopreserve embryos participate in what I call “life ethics”, and following the edicts of the Catholic Church, view the embryo as sacred human life that should not be destroyed. Those who wish to dispose of embryos participate in what I call “kin ethics” and view the embryo as a member of a larger family. Within this model freezing leaves embryos open to the possibility of their future circulation out side the bounds of the natal family. These findings have implications for the debates about when life begins and also demonstrates that the Catholic...

Extended culture up to the blastocyst stage: a strategy to avoid multiple pregnancies in assisted reproductive technologies Biol. Res.
Sepúlveda,Soledad J; Portella,Jimmy R; Noriega,Luis P; Escudero,Ernesto L; Noriega,Luis H.
The aim of this study was to review the experience and outcomes of assisted reproduction cycles with embryos grown up to day 5 of development, comparing different parameters according to the ages of the patients. We retrospectively studied 1,874 assisted reproduction cycles where embryo culture was extended up to the fifth or sixth day of development. All IVF and ICSI cycles were included, comparing, according to patient age, the following rates: blastocyst formation, pregnancy, implantation and abortion. As control, we analyzed cycles with donated oocytes from young donors (OD). The number of embryos reaching the blastocyst stage is similar in all groups of patients. Only the OD group was different in terms of blastocyst...

Early embryogenesis involves a series of dynamic processes, many of which are currently not well described or understood. Aneuploidy and aneuploid mosaicism, a mixture of aneuploid and euploid cells within one embryo, in early embryonic development are principal causes of developmental failure.^1,2^ Here we show that human embryos demonstrate a significant rate of genetic correction of aneuploidy, or "genetic normalization" when cultured from the cleavage stage on day 3 (Cleavage) to the blastocyst stage on day 5 (Blastocyst) using routine in vitro fertilization (IVF) laboratory conditions. One hundred and twenty-six human Cleavage stage embryos were evaluated for clinically indicated preimplantation...

In vitro fertilization (IVF) in mammals: epigenetic and developmental alterations. Scientific and bioethical implications for IVF in humans Biol. Res.
Ventura-Juncá,Patricio; Irarrázaval,Isabel; Rolle,Augusto J; Gutiérrez,Juan I; Moreno,Ricardo D; Santos,Manuel J.
The advent of in vitro fertilization (IVF) in animals and humans implies an extraordinary change in the environment where the beginning of a new organism takes place. In mammals fertilization occurs in the maternal oviduct, where there are unique conditions for guaranteeing the encounter of the gametes and the first stages of development of the embryo and thus its future. During this period a major epigenetic reprogramming takes place that is crucial for the normal fate of the embryo. This epigenetic reprogramming is very vulnerable to changes in environmental conditions such as the ones implied in IVF, including in vitro culture, nutrition, light, temperature, oxygen tension,...

Late Cretaceous-Early Palaeogene echinoderms and the K/T boundary in the southeast Netherlands and northeast Belgium — Part 6: Conclusions Naturalis
Jagt, J.W.M..
The palaeobiology of echinoderms occurring in the Meerssen and Geulhem members is discussed and changes in diversity across the K/T boundary are documented. Using literature data on the ecology of extant faunas, the various echinoderm groups are considered. Naturally, such data can only be applied with due caution to fossil forms, whose skeletal morphology is often incompletely known. This holds especially true for asteroids, ophiuroids, and crinoids, which, upon death, rapidly disintegrate into jumbles of dissociated ossicles. Bioturbation, scavenging, and current winnowing all contribute to blurring the picture still further. However, data on extant forms do allow a preliminary subdivision of fossil species into various ecological groups, which are...

Pre-incubation of porcine semen reduces the incidence of polyspermy on embryos derived from low quality oocytes Ciência Rural
Brogni,Cláudio Francisco; Ohlweiler,Lain Uriel; Klein,Norton; Mezzalira,Joana Claudia; Cristani,Jose; Mezzalira,Alceu.
ABSTRACT: The main cause of low efficiency of in vitro produced porcine embryos is the high polyspermic penetration rates at fertilization, which is aggravated in low quality oocytes. Experiment 1 evaluated the embryo development in high and low quality oocytes. Experiment 2 evaluated the embryo development and quality of low quality oocytes fertilized with sperm pre-incubated during 0h (control), 0.5h, 1h and 1.5h. Experiment 3 investigated fertilization and monospermic rates of the same groups of Experiment 2. Experiment 4 evaluated embryo development, cell density, fertilization and monospermic rates of high quality oocytes using semen pre incubated during the best time observed in the previous experiments. Cleavage and blastocyst rates were analyzed by...

Reactive oxygen species in bovine embryo in vitro production Biocell
Dalvit,G.C.; Cetica,P.D.; Pintos,L.N.; Beconi,M.T..
Oxidative modifications of cell components due to the action of reactive oxygen species (ROS) is one of the most potentially damaging processes for proper cell function. However, in the last few years it has been observed that ROS participate in physiological processes. The aim of this work was to determine ROS generation during in vitro production of bovine embryos. Cumulus-oocyte complexes were recovered by aspiration of antral follicles from ovaries obtained from slaughtered cows and cultured in medium 199 for 22 h at 39ºC in 5% CO2: 95% humidified air. In vitro fertilization was carried out in IVF-mSOF with frozenthawed semen in the same culture conditions and embryo in vitro culture in IVC-mSOF at 90% N2: 5% CO2: 5%...
