The palaeobiology of echinoderms occurring in the Meerssen and Geulhem members is discussed and changes in diversity across the K/T boundary are documented. Using literature data on the ecology of extant faunas, the various echinoderm groups are considered. Naturally, such data can only be applied with due caution to fossil forms, whose skeletal morphology is often incompletely known. This holds especially true for asteroids, ophiuroids, and crinoids, which, upon death, rapidly disintegrate into jumbles of dissociated ossicles. Bioturbation, scavenging, and current winnowing all contribute to blurring the picture still further. However, data on extant forms do allow a preliminary subdivision of fossil species into various ecological groups, which are...

The objective of this work was to evaluate the relationship between sperm head morphometry and chromatin packaging with the cleavage ratio and blastocyst formation during in vitro fertilization programs (IVF). Data of 38 IVFs using semen of zebu bulls with normal spermogram, carried out at two laboratories in Uberaba, Minas Gerais, Brazil, were used in this study. Sperm smears stained with toluidine blue and analyzed by computer image analysis were used to evaluate the morphometry and chromatin packaging. The packaging intensity and homogeneity of sperm chromatin were evaluated. The results made it possible to conclude that light variations in the head morphometry and chromatin packaging had little influence on the...

Zona incision using a piezo-micromanipulator (ZIP) has been demonstrated to be effective for in vitro fertilization (IVF) using cryopreserved C57BL/6 spermatozoa. In this study, ZIP oocytes inseminated with frozen-thawed genetically modified C57BL/6J or FVB mice spermatozoa (21 lines) showed fertilization rates of 22-75% and live fetus rates of 8-49%. In 6 of the lines, the fertilization rates for oocytes compared with ZIP (42-75%) were significantly higher than that of nontreated oocytes (0-50%). Using only 90 oocytes for IVF with ZIP, 5 breeding pairs were produced from cryopreserved genetically modified mice spermatozoa. Our results indicate that application of the ZIP technique is...

The aim of this study was to review the experience and outcomes of assisted reproduction cycles with embryos grown up to day 5 of development, comparing different parameters according to the ages of the patients. We retrospectively studied 1,874 assisted reproduction cycles where embryo culture was extended up to the fifth or sixth day of development. All IVF and ICSI cycles were included, comparing, according to patient age, the following rates: blastocyst formation, pregnancy, implantation and abortion. As control, we analyzed cycles with donated oocytes from young donors (OD). The number of embryos reaching the blastocyst stage is similar in all groups of patients. Only the OD group was different in terms of blastocyst...

Numerous studies have repeatedly shown that women who smoke experience problems establishing and maintaining pregnancies, and recent work has further demonstrated that the in utero effects of smoke may not be manifested until months or even years after birth. The purpose of this review is to examine the recent literature dealing with the effects of cigarette smoke on the earliest stages of human prenatal development. Studies in this area have included the use of animal models, patients undergoing in vitro fertilization, and embryonic stem cell models. Events leading to fertilization, such as cumulus expansion, hyperactivation of sperm motility, and oocyte pick-up by the oviduct are all impaired by smoke exposure in animal models. Steps crucial to...

Fasciolosis is a disease that affects the liver parenchyma and bile ducts of numerous animals, including humans, which causes economic losses and threatens public health. The present work aimed at reporting the distribution and factors connected with Fasciola hepatica infection in cattle in ten municipalities in southern Espírito Santo, Brazil, between 2008 and 2009. During this period, 50 rural properties in ten cities were analyzed and fecal samples from 10% of the cattle on each farm were collected. Of the 717 samples analyzed, 154 (21.33%) presented Fasciola hepatica eggs. As for the 50 rural properties studied, 32 (64%) had animals positive for fasciolosis. Throughout the evaluation, it was observed that 42 (84%) rural properties had flooded areas, 33...

Embryo quality is influenced by the culture conditions that affect in vitro maturation (IVM), fertilization (IVF) and culture (IVC) rates. The present study investigated the feasibility of producing bovine embryos after the replacement of fetal calf serum (FCS) and bovine serum albumin (BSA) by ovalbumin (OVA). The IVM and IVC medium were supplemented with 10% FCS, 4 mg/mL BSA, or 4 mg/mL OVA. The IVF medium was supplemented with 6 mg/mL BSA or OVA. For IVM, supplementation with FCS, BSA, and OVA did not affect nuclear maturation or cortical granule migration. Higher rates of formation of two pronuclei were obtained when FCS was employed for IVM (79.97%), regardless of the supplement used...

The objective was to investigate the effects of sex-sorting on early fertilization events in ovum pick up (OPU) derived oocytes fertilized in vitro with frozen-thawed sperm at different co-incubation lengths. Eighty-four OPU sessions were carried out in 18 cyclic, dry and non-stimulated Holstein Friesian and German black pied cows. Ovum pick up oocytes were matured in vitro for 24 hours and fertilized with frozen-thawed sex sorted or unsorted sperm from the same ejaculate. Fertilization was achieved by two experimental protocols: 1) short gamete co-incubation length: 4, 8 and 12 hours; and 2) long gamete co-incubation length: 18 and 24 hours. After in vitro fertilization, ova were fixed and stained to identify early fertilization events. Sperm penetration,...

In cattle, embryo development is characterized by the appearance of two distinct cell layers, the trophectoderm and the inner cell mass. The latter will undergo differentiation to form the embryonic disc consisting of the epiblast and hypoblast. The aim of this study was to ultrastructurally characterize the bovine embryo from different in vitro production techniques, with emphasis on trophectoderm and inner cell mass cells. Bovine embryos on day 7 (conception = D1) of pregnancy, derived via in vitro production techniques, were fixed for light and transmission electron microscopy processing. Results suggested that embryos produced by nuclear transfer of somatic cells and parthenogenesis showed significant changes in macroscopic and microscopic structure....

The advent of in vitro fertilization (IVF) in animals and humans implies an extraordinary change in the environment where the beginning of a new organism takes place. In mammals fertilization occurs in the maternal oviduct, where there are unique conditions for guaranteeing the encounter of the gametes and the first stages of development of the embryo and thus its future. During this period a major epigenetic reprogramming takes place that is crucial for the normal fate of the embryo. This epigenetic reprogramming is very vulnerable to changes in environmental conditions such as the ones implied in IVF, including in vitro culture, nutrition, light, temperature, oxygen tension,...
