Detection of ostreid herpesvirus 1 (OsHV-1) DNA in seawater by PCR: influence of water parameters in bioassays ArchiMer

Since 1991, herpesvirus infections have been reported among larvae and juveniles of various bivalves. Most of the studies focused on detection of viral infections of economically important species. However, the persistence of bivalve herpesviruses; in the marine environment is poorly documented. The present study concerns the role of seawater parameters in Ostreid Herpesvirus I (OsHV-1) detection by polymerase chain reaction (PCR). Viral DNA extracted from purified particles or virions present in infected oyster larvae were detected by PCR after storage in different media at different temperatures. Recherche par PCR d'OsHV-1 (Ostreid Herpesvirus type 1), dans des échantillons d'eau de claires ostréicoles ArchiMer

Depuis 1991, des mortalités massives associées à des infections à herpèsvirus sont rapportées chez des larves et des juvéniles de différentes espèces de bivalves dans différentes régions du globe. Un nouveau groupe au sein de la famille des Herpesviridae a été créé afin d'y intégrer les herpèsvirus d'invertébrés marins avec actuellement pour unique représentant le virus infectant l'huître creuse, Crassostrea gigas, OsHV-1. Des études ont porté sur la mise en évidence d'OsHV-1 chez les coquillages d'intérêt économique après développement d'outils de diagnostic rapides et fiables. The presence of a herpes-type virus has been demonstrated in various species of marine shellfish (bivalves and gastropoda) throughout different regions of the world (Europe, North and South America, Asia, Australia), and is frequently associated with high mortality levels. Nevertheless, apart from the sanitary monitoring in mollusc farming, very little data is available about the detection of this type of virus in the natural populations of bivalves and about the persistence of such viruses in sea water.
