This investigation describes the anticancer, antioxidant and antimicrobial properties of the extracts of Combretum rupicola, a native plant from Northeast of Brazil. Methanolic, ethyl acetate and chloroform extracts from leaves of C. rupicola were evaluated in relation to their potential in inhibiting the cell growth and cytotoxic properties against nine human cancer cell lines (MCF-7, NCI-ADR/RES, OVCAR-3, PC-3, HT-29, NCIH460, 786-O, UACC-62, K-562). In addition, antioxidant activity of these extracts was measured using DPPH as radical scavenging assay and the ntimicrobial activities against nine pathogens were tested by agar diffusion method. A rapid primary screening method for antitumor using the oomycete Pythium aphanidermatum. Repositório Alice

Investigating bioactive components from microbial metabolites can provide potential source for drug discovery. There are a number of traditional antitumor compounds screening assays already described. In this study, we have developed an efficient method to search for antitumor compounds using the Oomycete Pythium aphanidermatum as model on agar plates for high throughtput screening. Antioomycete compounds showed cytotoxic effects in human cancer cells lines due to the possible similarity between the lipid cell membrane of the Pythium species and cancerous cells, suggesting the compounds act through similar mechanisms of inhibition. ABSTRACT Cancer is a public health problem due to its high worldwide morbimortality. Current treatment protocols do not guarantee complete remission, which has prompted to search for new and more effective antitumoral compounds. Several substances exhibiting cytostatic and cytotoxic effects over cancer cells might contribute to the treatment of this pathology. Some studies indicate the presence of such substances in scorpion venom. ABSTRACT Physiological processes, as autophagy, proliferation and apoptosis are affected during carcinogenesis. Restoring cellular sensitivity to apoptotic stimuli, such as the antineoplastic cocktails, has been explored as a strategy to eliminate cancer cells. Autophagy, a physiological process of recycling organelles and macromolecules can be deviated from homeostasis to support cancer cells survival, proliferation, escape from apoptosis, and therapy resistance. The relationship between autophagy and apoptosis is complex and many stimuli can induce both processes. Thermodynamics of molecular recognitions between antineoplastic drug taxol and phosphatidylcholine BABT

The aim of this study was to study the basic features of Taxol recognition with phospholipids by applying the thermodynamic and spectroscopic measurements. The obtained information could be used further for deductions on its precise cellular and pharmacological mechanisms of action, on improvements of its solubility properties by phospholipids, as well as for designing the novel lipidic carriers for drug delivery.

Genotoxicity of the cyclo-oxygenase-inhibitor sulindac sulfide in the filamentous fungus Aspergillus nidulans BJM

Sulindac sulfide is a non-steroidal anti-inflammatory drug (NSAID) with chemopreventive effect on human cancer cells. Due to the involvement of the somatic recombination in the carcinogenic process, sulindac sulfide's recombinogenic potential was evaluated by the Homozygotization Index (HI) in the filamentous fungus Aspergillus nidulans. The drug's recombinogenic potential was evaluated by its capacity to induce homozygosis of recessive genes from heterozygous diploid cells. Sulindac sulfide at 175 and 350 µM concentrations induced mitotic recombination in A. nidulans diploid cells, with HI values for genetic markers higher than 2.0, and significantly different from control HI values. Doxorubicin and etoposide induce somatic recombination in diploid cells of Aspergillus nidulans BJM

Doxorubicin and etoposide are intercalating agents that inhibit the action of the enzyme topoisomerase II. Both drugs present therapeutic activity in numerous human neoplasms In the present work the recombinagenic potential of these drugs was evaluated by ascomycete Aspergillus nidulans. Their effects on the asexual cycle of A. nidulans was also appraised. Two heterozygous diploid strains of A. nidulans, a wild (uvsH+//uvsH+) and a defective to the DNA repair (uvsH//uvsH) were used.
