A programme of zoosanitary survey of the populations of black-lip pearl oyster (P. margaritifera) has been developed in French Polynesia during 1996 and 1997. Based on histological and ultrastructural examinations, the study of pearl oysters sampled in several pearl farms from the Society Islands, Tuamotu Atolls and Gambler Archipelago, has been particularly turned to enzootic pathological problems. Particular attention has been given to the shell disease and to the necrosis of the adductor muscle, symptoms that had accompanied the mass mortality in 1985.Virus-like particles, about 40 nm in diameter, were detected in membrane bound vesicles from the necrosized muscle tissue. The gametogenic cycle and the reproductive effort of the blacklip pearl oyster, Pinctada margaritifera, cultivated in Takapoto lagoon were studied for a 1-year period (March 1997-April 1998) by bimonthly observations of gonadal sections, dry tissue weights and gonadal index in a population of pearl oyster composed of three age-groups. pearl oysters attained sexual maturity in the end of their first year (height approximate to 40 mm), implying that P. margaritifera is a late-maturing species in comparison with other Pteriidae. This species was also confirmed to be a marked protandrous successive hermaphrodite in culture, with 100 % of males at first maturity and 75 % in older pearl oyster (height > 120 mm). Producing high quality cultured black pearls from Pinctada margaritifera is one of the major challenges for the "pearl oyster" industry in French Polynesia. In order to assess donor effect on cultured pearl quality, wild Pinctada margaritifera originating from the Tuamotu Archipelago were used in a duplicated grafting experiment. After 12 months of culture, nucleus retention was assessed and seven pearl quality traits recorded on the 454 cultured pearls harvested from the experiment.
