Citrus exocortis viroid and Hop stunt viroid doubly infecting grapevines in Brazil. Repositório Alice

Viroids, non-protein-coding small (246-40 I nt) eircular single-stranded RNAs with autonornous replication, are currently classified into two farnilics. Within the farnily Pospiviroidae, Citrus exocortis viroid (CEVd) belongs to lhe genus Pospiviroid while Hop stunt viroid (HSVd) is the single rnembcr of the genus Hostuviroid. These pathogens are distributcd worldwide and infect a large nurnbcr of hosts. In Brazil, isolates of CEVd and HSVd have been detected in both citrus and grapevine. Sequence analysis of three citrus viroids infecting a single Tunisian citrus tree (Citrus, reticulata, Clementine) Genet. Mol. Biol.

We report the nucleotide sequences of three citrus viroids belonging to three different genera: Citrus exocortis viroid (CEVd), Hop stunt viroid (HSVd) and Citrus viroid-III (CVd-III) isolated from a single natural infected Citrus reticulata var. Clementine tree growing in a tree nursery in Manouba (near Tunis Capital). We describe the sequence variability of these viroids from their natural host without using an alternative passage by an indicator host or an artificial inoculation. This work confirms that naturally occurring viroid infections contain a mixture of sequence variants. These are the first sequences of citrus viroids from Africa.

The so-called "quebra-galho" clone of Tahiti lime is very popular in Brazil, because the small size of the trees is suitable for high-density plantings. These Tahiti lime trees are easily recognized because they show bark-cracking symptoms, that have been claimed to be associated with "exocortis" infection. Viroid infection of three "quebra-galho" Tahiti lime trees from a farm near Araraquara in the state of São Paulo, Brazil, was assessed by Northern-blot hybridization using viroid-specific probes. Similarly, eight clones of Tahiti lime from different origins and available at Topara Nursery, near Chincha, Peru, were also tested for viroids.
