Anticandidal effect of endophytic bacteria isolated from Equisetum arvense L. against Candida albicans and Candida glabrata BABT
Das,Gitishree; Patra,Jayanta Kumar; Choi,Jaehyuk; Baek,Kwang-Hyun.
ABSTRACT Equisetum arvense, a fern species possesses a number of pharmaceutical prospective. In the present study, a total of 103 endophytic bacteria isolated from E. arvense and were evaluated for their anticandidal property against five Candida species, two C. albicans, C. glabrata, C. saitoana and C. geochares. Out of them fifty one were identified as per the morphological and molecular characterisation using 16S rRNA gene sequencing and among them, ten promising endophytic bacteria were mentioned in the present study. Among ten endophytic bacteria, Psychrobacillus insolitus and Curtobacterium oceanosedimentum exerted highest anticandidal effect against C. albicans KACC 30062 and C. glabrata KBNO6P00368, with diameter of inhibition zones of 21.30±0.41...

Synergistic effects of tacrolimus and azole antifungal compounds in fluconazole-susceptible and fluconazole-resistant Candida glabrata isolates BJM
Denardi,Laura Bedin; Mario,Débora Alves Nunes; Loreto,Érico Silva; Santurio,Janio Morais; Alves,Sydney Hartz.
In vitro interaction between tacrolimus (FK506) and four azoles (fluconazole, ketoconazole, itraconazole and voriconazole) against thirty clinical isolates of both fluconazole susceptible and -resistant Candida glabrata were evaluated by the checkerboard microdilution method. Synergistic, indifferent or antagonism interactions were found for combinations of the antifungal agents and FK506. A larger synergistic effect was observed for the combinations of FK506 with itraconazole and voriconazole (43%), followed by that of the combination with ketoconazole (37%), against fluconazole-susceptible isolates. For fluconazole-resistant C. glabrata, a higher synergistic effect was obtained from FK506 combined with ketoconazole (77%), itraconazole (73%), voriconazole...

Multidrug-resistant Candida glabrata strains obtained by induction of anidulafungin resistance in planktonic and biofilm cells BJPS
Hatwig,Camila; Balbueno,Eduardo A.; Bergamo,Vanessa Z.; Pippi,Bruna; Fuentefria,Alexandre M.; Silveira,Gustavo P..
Candida glabrata has emerged as a common cause of serious life-threatening fungal infections, largely owing to their low susceptibility to azole antifungals. Recent guidance indicates the use of echinocandins as the first-choice drug for the treatment of systemic infections of C. glabrata; however, C. glabrata resistance to echinocandins is reportedly increasing. Herein, we present the induction of anidulafungin resistance in planktonic and sessile cells of C. glabrata and the development of fluconazole cross-resistance. MICs of 21 clinical C. glabrata strains were determined by a broth microdilution method using anidulafungin and fluconazole. Biofilm formation on a tracheal catheter was determined using 1- × 1-cm2 polyvinyl polychloride catheter...

Proteomic analysis of cytosolic proteins associated with petite mutations in Candida glabrata BJMBR
Loureiro y Penha,C.V.; Kubitschek,P.H.B.; Larcher,G.; Perales,J.; Rodriguez León,I.; Lopes-Bezerra,L.M.; Bouchara,J.P..
The incidence of superficial or deep-seated infections due to Candida glabrata has increased markedly, probably because of the low intrinsic susceptibility of this microorganism to azole antifungals and its relatively high propensity to acquire azole resistance. To determine changes in the C. glabrata proteome associated with petite mutations, cytosolic extracts from an azole-resistant petite mutant of C. glabrata induced by exposure to ethidium bromide, and from its azole-susceptible parent isolate were compared by two-dimensional polyacrylamide gel electrophoresis. Proteins of interest were identified by peptide mass fingerprinting or sequence tagging using a matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometer. Tryptic...

A new biosurfactant produced by Candida glabrata UCP 1002: characteristics of stability and application in oil recovery BABT
Luna,Juliana Moura de; Sarubbo,Leonie; Campos-Takaki,Galba Maria de.
The production of a new biosurfactant by Candida glabrata UCP1002 was studied to evaluate the influence of the concentration of the cotton seed oil, glucose and yeast extract. The dynamics of the growth and surfactant production were showed for all the cultivation conditions studied. The best emulsification of the n-hexadecane, quantified by the emulsifying index was observed in the medium containing 7.5% cotton seed oil, 5% glucose and 0.3% yeast extract. The isolated biosurfactant showed a CMC of 2.5% and the surface tension at that point showed to be 31mN/m. The potential application of the biosurfactant in oil recovery from the sand, in acid and alkaline environments and over exposure to high salinity and different temperatures was demonstrated by the...

Candida Glabrata Esophagitis: new case reports and management BJM
Macêdo,Danielle Patrícia Cerqueira; Silva,Vanessa Karina Alves da; Farias,Aline Mary de Almeida; Melo,Luciana Resende Bandeira de; Wilheim,Ana Botler; Neves,Rejane Pereira.
Candida esophagitis (CE) is a common opportunistic infection in the immunocompromised host. C. glabrata is rarely cited as agent of CE and has been underestimated due to lack of proper identification. In this study, two cases of C. glabrata esophagitis in AIDS and chagasic patients are reported. Diagnosis of Candida species should be considered an important key for the ideal choice of antifungal therapy against this mycosis.

Occurrence of killer Candida glabrata clinical isolates BJM
O,Arroyo-Helguera; Alejandro,De Las Penas; Irene,Castaño.
In this work we characterized the occurrence of killer activity in 64 Candida glabrata clinical isolates under different conditions. We found that only 6.25 % of the clinical isolates tested were positive for killer activity against a Saccharomyces cerevisiae W303 sensitive strain. Sensitivity of killer activity to different values of pH and temperatures was analyzed. We found that the killer activity presented by all isolates was resistant to every pH and temperature tested, although optimal activity was found at a range of pH values from 4 to 7 and at 37ºC. We did not observe extrachromosomal genetic elements associated with killer activity in any of the positive C. glabrata isolates. The killer effect was due to a decrease in viability and DNA...

Candida glabrata among Candida spp. from environmental health practitioners of a Brazilian Hospital BJM
Savastano,Catarina; Silva,Elisa de Oliveira; Gonçalves,Lindyanne Lemos; Nery,Jéssica Maria; Silva,Naiara Chaves; Dias,Amanda Latercia Tranches.
Abstract The incidence of the species Candida albicans and non-albicans Candida was evaluated in a Brazilian Tertiary Hospital from the environment and health practitioners. In a 12-month period we had a total positivity of 19.65% of Candida spp. The most recurring non-albicans Candida species was C. glabrata (37.62%), generally considered a species of low virulence, but with a higher mortality rate than C. albicans. Subsequently, C. parapsilosis (25.74%) and C. tropicalis (16.86%) were the second and third most commonly isolated species. Considering the total samples collected from the emergency room and from the inpatient and the pediatric sector, 19.10% were positive for Candida spp., with the predominance of non-albicans Candida species (89.42%). The...
