Production of a cloned calf from a fetal fibroblast cell line BJMBR

The present study examined the in vitro and in vivo development of bovine nuclear-transferred embryos. A bovine fetal fibroblast culture was established and used as nucleus donor. Slaughterhouse oocytes were matured in vitro for 18 h before enucleation. Enucleated oocytes were fused with fetal fibroblasts with an electric stimulus and treated with cytochalasin D and cycloheximide for 1 h followed by cycloheximide alone for 4 h. Reconstructed embryos were cultured for 7-9 days and those which developed to blastocysts were transferred to recipient cows. Of 191 enucleated oocytes, 83 (43.5%) were successfully fused and 24 (28.9%) developed to blastocysts. Recombinant antigen-based immuno-slot blot method for serodiagnosis of syphilis BJMBR

Three recombinant antigens of Treponema pallidum Nichols strain were fused with GST, cloned and expressed in Escherichia coli, resulting in high levels of GST-rTp47 and GST-rTp17 expression, and supplementation with arginine tRNA for the AGR codon was needed to obtain GST-rTp15 overexpression. Purified fusion protein yields were 1.9, 1.7 and 5.3 mg/l of cell culture for GST-rTp47, GST-rTp17 and GST-rTp15, respectively. The identities of the antigens obtained were confirmed by automated DNA sequencing using ABI Prism 310 and peptide mapping by Finningan LC/MS. Rhizobacterial promotion of eucalypt rooting and growth BJM

A total of 107 rhizobacterial isolates, obtained from the rhizosphere of eucalypt clones were tested as rooting inducers of cuttings and mini-cuttings planted in substrate composed of carbonized rice husk and vermiculite (1:1). Cuttings and mini-cuttings were planted in conical plastic tubes containing treated and untreated (control) substrate and kept under intermittent mist irrigation at 26-28ºC. After 35 days, rooting percentage and dry root matter of cuttings were evaluated. Ten isolates capable of providing gains of up to 110% in root formation and up to 250% in root biomass over non-inoculated control cuttings were selected. Seedborne transmission of Zucchini yellow mosaic virus and Cucumber mosaic virus in Styrian Hulless group of Cucurbita pepo National Institute of Agronomic Research

A total of 62 seed lots originating from hulless pumpkin fruit exhibiting typical symptoms of ZYMV infection were tested in insect-free greenhouses in three consecutive years. Of those, 25 seed lots showed seedborne virus transmission that ranged from 0.29 to 15.34 % of the seeds. In total 19997 seeds were sowed, 16039 plants were monitored and 225 plants were seedborne virus infected. Of those, 166 had ZYMV and 59 had mixed infection of ZYMV and CMV as identified by ELISA serological tests, by indicator plants or by RT-PCR. The average rate of seedborne transmission of ZYMV was 1.4 %. Coat protein gene of CMV and ZYMV were characterized and compared.

Taura syndrome of penaeid shrimp: cloning of viral genome fragments and development of specific gene probes ArchiMer

The ssRNA genome extracted from purified Taura Syndrome Virus (TSV) was transcribed into double-stranded, blunt-ended cDNA and was used to construct cDNA libraries either in pUC 18 or in pBluescript II KS-vectors. Twelve recombinant plasmids chosen after screening of the libraries were subjected to restriction enzyme digestions for determination of size inserts and restriction maps. A conservative estimate of 5% losses due to disease means that the finfish aquaculture industry loses over $1 billion annually on a global scale. Drought stress could induce expressions of various genes that are involved in stress tolerance and responses. Recent studies were emphasized on a gene encoded for the 9-cis epoxycarotenoid dioxygenase (NCED) enzyme which catalyzes the biosynthesis of abscisic acid (ABA) in plants. However, little is known about drought tolerance genes in maize. We designed primers from the VP14 sequence in the NCBI gene bank for the study of expression of drought tolerance gene in SW 2301 drought tolerant and KSX 4605 drought sensitive lines.
