Rapid diagnosis of rabies in suspected human cases influences post-exposure prophylaxis for potential contacts of the patient and ensures appropriate patient management. Apart from the central nervous system (CNS), rabies virus (RABV) is usually present in small sensory nerves adjacent to hair follicles of infected humans. We used an RT-PCR, with primers targeted to the 3' terminal portion of the nucleoprotein gene (N), to test neck-skin samples of nine patients who had rabies in order to validate a diagnostic method that could serve as an additional tool for rabies diagnosis, particularly in antemortem samples. Six of eight postmortem samples were found to be positive for rabies by RT-PCR, and one of two samples collected antemortem was positive with this...

Diagnosis of infectious bronchitis: an overview of concepts and tools Rev. Bras. Ciênc. Avic.
Villarreal,LYB.
Infectious bronchitis (IB) casues multi-systemic infection in chickens with signs similar caused by other poultry pathogens and thus a high diagnostic accuracy can only be achieved by s series of laboratory assays. This article reviews in a brief way the traditional virus assays such as embryo innoculation, tracheal rings and virus neutralization assays for the direct detection of Avian infectious bronchitis virus (IBV) and methods based on gene molecular biology and some assays for the detection of anti-IBV antibodies, including ELISA. A critical view on each technique is also provived by the author.

Diagnosis of oyster herpes-like virus: development and validation of molecular, immunological and cellular tools "VINO" - Final Report 4th January 1999 to 3rd January 2002 ArchiMer
Renault, Tristan; Davison, Andrew; Xhonneux, Florence; Dorange, Germaine; Culloty, Sarah; Novoa, Beatriz; Dixon, Peter.
Little information is available on viral infections that affect bivalve molluscs. Such a lack of data is due ta a certain inadequacy of the diagnosis methods that are employed when massive mortality events occur. Most laboratories involved in mollusc pathology still analyse samples through light microscopy. World-wide, there is thus currently a lack of information concerning the occurrence of bivalve herpesvituses. This is probably due to the lack of suitable diagnostic tools. The basic method for identification and examination of suspect samples is predominantly histopathology. This enables the identification of any cellular changes, but is not conclusive identification of bivalve herpesviruses. This technique doesn't allow, by itself, to detect viruses...

Renault, Tristan; Davison, Andrew; Xhonneux, Florence; Dorange, Germaine; Culloty, Sarah; Novoa, Beatriz; Dixon, Peter.
The aim of the programme is to develop specific tools for diagnosing herpes-like virus infections in bivalves and to validate these reagents by using them in different European laboratories involved in shellfish epidemiological surveys. This will done using techniques developed by the partners to characterise viruses by studying their genomes and immunologically reactive proteins, to cultivate oyster cells and vertebrate cell lines and to perform epidemiological surveys among bivalves. The programme objectives are: 1 - Obtaining the complete oyster herpes-like virus DNA sequence with determination of the genome structure. 2 - Comparing oyster herpes-like viruses with other viruses belonging to the Helpesviridae family on the basis of sequence data and...

Herpesvirus infections have been reported in different bivalves species around the world. Specific diagnostic tools are needed in order ta assess the causative raIe of these agents in mortalities observed among bivalves There is currently a lack of information concerning the occurrence of bivalve herpesviruses. This is probably due ta the lack of suitable diagnostic tools. The basic method for examination of bivalve samples is predominantly histology. This enables the identification of any cellular changes, but is not conclusive for virus identification. Transmission electron microscopy is a necessity for visual confirmation of viral infections. However, histology and electron microscopy are time consuming and inadequate for epidemiological studies. The...

Diagnosis of patients with Prader-Willi and Angelman Syndromes: the importance of an overall investigation Genet. Mol. Biol.
Varela,Monica Castro; Fridman,Cintia; Koiffmann,Célia Priszkulnik.
Seventy-two patients with clinical diagnoses of Prader-Willi (PWS; n = 28 patients) or Angelman syndromes (AS; n = 44 patients) were submitted to chromosome analysis, SNRPN-SNURF exon 1 methylation assay, and microsatellite genotyping. Analysis of the methylation pattern confirmed the PWS diagnosis in 18 out of 28 patients and the AS diagnosis in 20 out of 44 patients. FISH and microsatellite analysis detected a deletion in 30 patients (14 PWS and 16 AS). Eight patients had normal FISH results (4 PWS and 4 AS); microsatellite markers showed that these patients had a uniparental disomy (UPD). Based on this study, we propose a strategy for the routine diagnosis of these syndromes that consists of the following steps: 1) methylation analysis, which does not...

Diagnosis of pulmonary tuberculosis by score system in children and adolescents: a trial in a reference center in Bahia, Brazil BJID
Sant'Anna,Clemax Couto; Santos,Marilene Augusta R. C.; Franco,Rosana.
Since 2002, the Brazilian Ministry of Health has recommended a score system for tuberculosis diagnosis of children and adolescents that does not need bacteriological positivity, because most cases in this age group have few bacteria. An observational, transversal study was carried out at the outpatient health care service of the reference medical service in Salvador, Bahia, including 164 patients with pulmonary tuberculosis, with ages ranging between 1 and 15 years of age, who were treated from 1990 to 2001. The gold standard used to establish the diagnosis was clinical, radiological, epidemiological and based on follow-up data. The score system for diagnosis purposes was tested retrospectively. The median age and the average age of the 164 patients were 6...

Diagnostic and prognostic value of procalcitonin for early intracranial infection after craniotomy BJMBR
Yu,Y.; Li,H.J..
Intracranial infection is a common clinical complication after craniotomy. We aimed to explore the diagnostic and prognostic value of dynamic changing procalcitonin (PCT) in early intracranial infection after craniotomy. A prospective study was performed on 93 patients suspected of intracranial infection after craniotomy. Routine peripheral venous blood was collected on the day of admission, and C reactive protein (CRP) and PCT levels were measured. Cerebrospinal fluid (CSF) was collected for routine biochemical, PCT and culture assessment. Serum and CSF analysis continued on days 1, 2, 3, 5, 7, 9, and 11. The patients were divided into intracranial infection group and non-intracranial infection group; intracranial infection group was further divided into...

Diagnostic value of circulating lncRNA ANRIL and its correlation with coronary artery disease parameters BJMBR
Hu,Yao; Hu,Jing.
This study aimed to detect the expression of the long non-coding RNA (lncRNA) antisense non-coding RNA in the INK4 locus (ANRIL) and evaluate its correlation with disease risk, stenosis degree, inflammation, as well as overall survival (OS) in coronary artery disease (CAD) patients. A total of 230 patients who underwent diagnostic coronary angiography were consecutively recruited and assigned to CAD group (n=125) or control group (n=105) according to presence or absence of CAD. Gensini score was calculated to assess the severity of coronary artery damage. Plasma samples were collected and the expression ANRIL was detected in all participants. High-sensitivity C-reactive protein (hs-CRP), erythrocyte sedimentation rate (ESR), and cytokines including tumor...
