The aim of this work was to prepare the mares for embryo transfer. In group 1 (G1,treated, n=15), recipient mares in anoestrus or in a transition period were treated with 5.0, 3.0 and 2.0 mg of estradiol cipionate at the days 0, 1 and 2 respectively, beginning at the day of ovulation (DO). From the fourth day on, the mares this group received long-acting progesterone weekly, up to the 120 day of gestation. At D8, the embryo was collected from the donor and transferred to the recipient. At D12, the ultrasonographyc diagnosis of pregnancy was carried out. Abstract: Animal surgery procedures are being progressively restricted worldwide in the context of embryo transfer. In small ruminants, the needs for development of alternative and efficient non-surgical techniques for embryo transfer have been emphasized. Non-surgical embryo recovery is well consolidated in Brazil in cattle and goats, while in sheep it remains a challenge. In mammals, embryo implantation is an essential step in reproduction. Implantation is a phenomenon that involves crosstalk between the blastocyst and the maternal endometrium. However, the molecular basis of the connections between the blastocyst and endometrium is not yet fully understood. Amphiregulin is a member of the epidermal growth factor family and is known to be expressed in the luminal epithelium of the mouse uterus on 3.5 days post coitum (dpc). Resumo: 0 trabalho tem o objetivo de contribuir corn programas de transferência de embriôes em caprinos no Brasil através do conhecimento das respostas aos tratamentos de sincronizaçao do estro e superovulaçao e da melhor data de colheita de embriôes em relaçao ao estadio embrionario. Os dadas obtidos sao referentes a 19 cabras mestiças das Raças Parda Alpina, Moxoto e Anglo Nubiana (ME), 13 Saanen (SA) e 6 Anglo Nubianas (NB). Abstract: The variability of superovulatory responses is still a limiting factor for the consolidation of commercial embryo transfer programs in small ruminants (Bartlewski et al. 2016. Theriogenology, 86: 130-143). Superovulation protocols are traditionally performed with prior use of progesterone devices for short- or long-term duration (Oliveira et al. 2014. ABSTRACT The efficiency of an alternative freezing protocol for goat embryos of different morphology and quality was tested. Fifty-eight embryos on Day 6-7 stage were transferred as fresh or after freeze-thawing (n=29/group). For freezing, embryos were placed into 1.5M ethylene-glycol solution for 10min. During this time, they were loaded in the central part of 0.25mL straw, separated by air bubble from columns containing PBS/BSA 0.4% plus 20% BFS. Straws were then frozen using a freezing machine from 20ºC to -6ºC at a cooling rate of 3ºC/min, stabilization for 15min (seeding after 5min), from -6 C to -32ºC at 0.6 C/min,and plunged into liquid nitrogen. Abstract: Ewes have certain particularities in the cervical anatomy leading to the commercial use of surgical methods for collecting embryos in this species. However, the literature shows that Non-Surgical Embryo Recovery (NSER) is possible through specific protocols for cervical relaxation (Fonseca et al., 2019. Reprod Fertil Devel, 31:17-26). Abstract: This study aimed to monitor estrous cycle parameters of a human granulocyte colony-stimulating factor (hG-CSF)-transgenic founder female goat and to perform superovulation and embryo recovery (surgical or transcervical method) for further transfer to recipients to quickly obtain offspring.
