BackgroundThe state of Pará encompasses 26% of Brazilian Amazon where an enormous diversity of arboviruses has been found. This study aimed to assess the prevalence and distribution of hemagglutination-inhibition antibodies against antigens of six Flavivirus(yellow fever virus, Ilheus virus, Saint Louis encephalitis virus, Cacipacore virus, Bussuquara virus and Rocio virus) in water buffaloes in Pará state, Brazil. Los cultivos celulares de mosquitos son frecuentemente utilizados para el aislamiento, identificación y caracterización de arbovirus. Para el estudio de los virus dengue (VDEN) y virus de fiebre amarilla (VFA) se emplean, principalmente, la línea celular C6/36 de Aedes albopictus y la línea celular AP61, obtenida de Aedes pseudoscutellaris. La línea celular de A. aegypti AEGY28, previamente obtenida a partir de tejidos embrionarios del vector, se utilizó en el presente trabajo para evaluar la susceptibilidad a la infección por VDEN y VFA. Searching for the best real-time RT-PCRs to detect Zika virus infections: the importance of comparing several protocols BJMBR

Clinical manifestations of Zika, dengue, and chikungunya virus infections are very similar, making it difficult to reach a diagnosis based only on clinical grounds. In addition, there is an intense cross-reactivity between antibodies directed to Zika virus and other flaviviruses, and an accurate Zika diagnosis is best achieved by real-time RT-PCR. However, some real-time RT-PCR show better performance than others. To reach the best possible Zika diagnosis, the analytic sensitivity of some probe-based real-time RT-PCR amplifying Zika virus RNA was evaluated in spiked and clinical samples. El virus West Nile es un flavivirus patógeno para humanos en el Viejo Mundo que ha experimentado recientemente un proceso de emergencia en el continente americano. Desde su introducción en 1999 en EEUU, se ha convertido en un patógeno de preocupación para las poblaciones de aves silvestres al haber provocado allí eventos masivos de mortalidad y porque numerosas especies de aves han experimentado disminuciones significativas de sus poblaciones. Para 2001 el virus ya se había dispersado fuera de EEUU, extendiéndose por América del Sur, América Central y el Caribe. Debido a la falta de un sistema de vigilancia activo para esta patología, no se conoce su verdadero impacto sobre las poblaciones silvestres en esas regiones. Las infecciones transmitidas por Flavivirus se encuentran entre las enfermedades transmisibles con mayor incidencia en el mundo. La mayoría de ellas se manifiestan clínicamente como un síndrome febril que puede estar o no acompañado de diversos síntomas. La severidad de estas infecciones es variable con casos asintomáticos y otros que pueden llegar a ser letales. La razón de esta variabilidad en la presentación clínica, se desconoce en humanos. Dengue virus-induced regulation of the host cell translational machinery BJMBR

Dengue virus (DV)-induced changes in the host cell protein synthesis machinery are not well understood. We investigated the transcriptional changes related to initiation of protein synthesis. The human hepatoma cell line, HepG2, was infected with DV serotype 2 for 1 h at a multiplicity of infection of one. RNA was extracted after 6, 24 and 48 h. Microarray results showed that 36.5% of the translation factors related to initiation of protein synthesis had significant differential expression (Z-score ≥ ±2.0). The yellow fever 17D vaccine virus: molecular basis of viral attenuation and its use as an expression vector BJMBR
The yellow fever (YF) virus is the prototype flavivirus. The use of molecular techniques has unraveled the basic mechanisms of viral genome structure and expression. Recent trends in flavivirus research include the use of infectious clone technology with which it is possible to recover virus from cloned cDNA. Using this technique, mutations can be introduced at any point of the viral genome and their resulting effect on virus phenotype can be assessed. This approach has opened new possibilities to study several biological viral features with special emphasis on the issue of virulence/attenuation of the YF virus.
