Assessing Helicobacter pylori motility and biofilm formation in subinhibitory concentrations of antimicrobials. Repositório Alice
FAURO, C. A. M.; TESSARI, L. C. M.; SANTOS, N. R. F. dos; CORREIA, N. S.; BONACORSI, C.; BAILÃO, A. M.; BAEZA, L. C.; VERBISCK, N. V.; DONOFRIO, F. C..
Numerous studies have shown that subinhibitory concentrations of antimicrobials can alter bacterial virulence factors. This study evaluates motility and biofilm formation by H. pylori 43504 grown in subinhibitory concentrations of amoxicillin (AMX), clarithromycin (CLA), or tetracycline (TET). For the swimming and swarming motility assays, H. pylori 43504 suspensions were prepared with the strain alone or with the strain in AMX, CLA, or TET at ½ MIC. Next, the media were incubated at 37 ºC, under microaerophilia. To assess biofilm formation in the presence of one of the antimicrobials at subinhibitory antimicrobial concentrations, bacterial suspensions (109 CFU/mL) were prepared in 2.5% FBS containing AMX, CLA, or TET at ½ MIC. After incubation for 10...

Correlation between Helicobacter pylori infection, gastric diseases and life habits among patients treated at a university hospital in Southeast Brazil BJID
Módena,José Luiz Proença; Acrani,Gustavo Olszanski; Micas,André Fernando Ditondo; Castro,Marly de; Silveira,Wanderley Dias da; Módena,José Luiz Pimenta; Oliveira,Ricardo Brandt de; Brocchi,Marcelo.
Helicobacter pylori is considered a significant agent in the development of various gastric diseases. However, the diseases caused by this bacterium are known as being multi-factorial, with the genotype, immune system and life habits of the host playing important roles in the establishment of the clinical outcome. Also, H. pylori exhibit a high degree of genetic variability, contributing to the complexity of the host-pathogen relationship. These observations, considered together with the widely-varying origins and social habits of the Brazilian population, lead us to speculate about the influence of these life habits on H. pylori infection and the clinical outcome. Therefore, in this study we evaluated the relationship between H. pylori infection and...

Eradication of Helicobacter pylori infection in patients with duodenal ulcer and non-ulcer dyspepsia and analysis of one-year reinfection rates BJMBR
Della Libera,E.; Rohr,M.R.S.; Moraes,M.; Siqueira,E.S.; Ferrari Jr.,A.P..
Helicobacter pylori (HP) infection is endemic worldwide. The proposed treatment is expensive and there are few reports regarding reinfection rates in Brazil. The aim of this study was to compare the eradication rates obtained with two therapeutic options and to evaluate reinfection one year after treatment. This was a prospective randomized trial with 55 patients. Thirty-nine patients had active duodenal ulcer (DU) and 16 non-ulcer dyspepsia (NUD), and all tested positive for HP. Diagnosis was based on at least two positive tests: ultrarapid urease test, histology and/or culture. Patients were randomized to two groups: group OMC treated with 40 mg omeprazole (once a day), 500 mg metronidazole and 250 mg clarithromycin (twice daily) for 7 days, or group NA...

Investigation of the association between clinical outcome and the cag pathogenicity-island and other virulence genes of Helicobacter pylori isolates from patients with dyspepsia in Eastern Turkey BJM
Ozbey,Gokben; Demirel,Ulvi; Aygun,Cem; Ertas,Hasan Basri.
The aims of our work were to determine the presence of the cag pathogenicity-island (cag PAI) and other virulence genes of Helicobacter pylori recovered from patients with gastritis and peptic ulcer, and to investigate the correlation of these virulence genes with clinical outcome. The presence of the cagA, the promoter regions of cagA, cagE, cagT, and the left end of cag-PAI (LEC), cag right junction (cagRJ), the plasticity region open reading frames (ORFs), vacA and oipA genes among 69 H. pylori isolates were determined by polymerase chain reaction. Intact cag PAI was detected in only one (1.4%) isolate. The cagA gene was identified in 52.1% and 76.2% of isolates from patients with dyspepsia (gastritis and peptic ulcer), respectively. The plasticity...

Abstract Considering that Helicobacter pylori, a bacterium able to colonize the upper gastrointestinal tract and cause mucosal injury, not always can be effectively eradicated by the traditional approaches, there is an interest in alternative therapies until a vaccine be available. Honey is a food supplement with high carbohydrate content and antioxidant activity, as well as broad antimicrobial spectrum. After analyzing the physicochemical and in vitro antimicrobial properties of an Apis mellifera honey from the Atlantic forest of Alagoas / Brazil, the purpose of the present work was evaluate its in vivo effects against Helicobacter pylori in the gastric mucosa of Wistar rats. First, it was verified the success of inoculation/infection of the pathogen in...
