Mycobacterium avium ssp. paratuberculosis (MAP) is the etiologic agent of paratuberculosis and it potentially plays a role in Crohn?s disease. In humans, the main route of transmission of MAP might be the intake of contaminated milk and dairy products. Considering that MAP has already been detected in many types of cheese in different counties, and that Coalho cheese is an important dairy product in northeastern Brazil, the aim of this study was to report the first detection of MAP in retail Coalho cheese in Brazil by PCR and culture. Of 30 retail Coalho cheese samples, 3 (10%) amplified fragments of a similar size to that expected (626 bp) were obtained and viable MAP was recovered by culture from 1 (3.3%) sample. The DNA from the positive culture sample...

Detection of Mycobacterium avium subsp. paratuberculosis in a cattle/pudu interface Arq. Bras. Med. Vet. Zootec.
Salgado,M.; Aleuy,O. A.; Sevilla,I. A.; Troncoso,E..
ABSTRACTMycobacterium avium subsp. paratuberculosis (Map) is the etiological agent of paratuberculosis. In Chile, information about Map isolation from both domestic ruminant and wildlife species has been accumulating, but it has to be extended to other species. The present study focuses specifically on one wild grazing species, the pudu (Pudu puda), one of the less known South American deer considered an endangered species that shares pastures with cattle in southern Chile, where the greatest part of the country's dairy cattle population is located. Convenient samples from 3 pudus were collected from one dairy farm where Map infection had previously been confirmed in cattle. All three pudus shed the bacterium in feces and the isolates are the same type of...

Albuquerque,Pedro Paulo Feitosa de; Santos,André de Souza; Souza Neto,Orestes Luiz de; Kim,Pomy de Cássia Peixoto; Cavalcanti,Erika Fernanda Torres Samico Fernandes; Oliveira,Júnior Mário Baltazar de; Mota,Rinaldo Aparecido; Pinheiro Júnior,José Wilton.
Abstract The aim of this study was to detect the IS900 region of Mycobacterium avium subsp. paratuberculosis (MAP) in bovine milk samples using real-time polymerase chain reaction (qPCR) and conventional PCR, and to study the agreement between these tests. A total of 121 bovine milk samples were collected from herds considered positive for MAP, from the State of Pernambuco, Brazil. MAP DNA was detected in 20 samples (16.5%) using conventional PCR and in 34 samples (28.1%) using qPCR. MAP DNA was detected in all of the 6 animal farms studied. Moderate agreement was found between qPCR and conventional PCR results, where the sensitivity and specificity of conventional PCR in relation to qPCR were 50% and 96.6%, respectively. Thus, the IS900 region of MAP was...

Multiple-locus variable number-tandem repeat analysis (MLVA) of Mycobacterium avium subspecies paratuberculosis (MAP) isolates may contribute to the knowledge of strain diversity in Argentina. Although the diversity of MAP has been previously investigated in Argentina using IS900-RFLP, a small number of isolates were employed, and a low discriminative power was reached. The aim of the present study was to test the genetic diversity among MAP isolates using an MLVA approach based on 8 repetitive loci. We studied 97 isolates from cattle, goat and sheep and could describe 7 different patterns: INMV1, INMV2, INMV11, INMV13, INMV16, INMV33 and one incomplete pattern. INMV1 and INMV2 were the most frequent patterns, grouping 76.3% of the isolates. We were also...

Paratuberculosis: an update BJM
Lilenbaum,W.; Marassi,C.D.; Oelemann,W.M.R..
Paratuberculosis is a chronic enteritis that affects ruminants and is caused by Mycobacterium avium paratuberculosis (Map). The disease is worldwide spread and causes important economic losses. In Brazil, the bacillus was isolated in the south and northeast regions of the country and in Rio de Janeiro, but there are no enough epidemiological studies about its occurrence. Isolation of Map from tissues or fecal samples is 100% specific, but Map shows the most fastidious growth of all mycobacteria. Incubation lasts 8 -12 weeks, with a dependency on exogenous mycobactin J. Diagnostic tests based on specific DNA sequences allow fast and secure identification, and PCR has been used to confirm positive culture results and to identify Map in feces, milk and...

Improvement of an in-house ELISA for bovine paratuberculosis serology in Brazil BJM
Marassi,Carla Dray; Fonseca,Leila de Souza; Ristow,Paula; Ferreira,Rachel; Lilenbaum,Walter; Oelemann,Walter M.R..
Paratuberculosis (Johne's disease) is a chronic enteritis in cattle caused by infection with Mycobacterium avium subsp. paratuberculosis (Map). In Brazil, few reports describe the isolation of the organism. The gold standard test for Map is its isolation from tissues or feces. Moreover, bacterial growth is slow and test results are available only after three to five months of incubation Furthermore, shedding of bacilli at levels detectable by fecal culture is irregular and does not occur during the early stages of infection, which compromises the sensitivity of this methodology. The most common immunological tests to identify Map infection are complement fixation test (CFT), agar gel immunodiffusion (AGID) and enzyme-linked immunoassay (ELISA). In Brazil,...

LAMP technology: Rapid identification of Brucella and Mycobacterium avium subsp. paratuberculosis BJM
Trangoni,Marcos D.; Gioffré,Andrea K.; Cerón Cucchi,María E.; Caimi,Karina C.; Ruybal,Paula; Zumárraga,Martín J.; Cravero,Silvio L..
In this study, we developed new sets of primers to detect Brucella spp. and M. avium subsp. paratuberculosis (MAP) through isothermal amplification. We selected a previously well-characterized target gene, bscp31, specific for Brucella spp. and IS900 for MAP. The limits of detection using the loop-mediated isothermal amplification (LAMP) protocols described herein were similar to those of conventional PCR targeting the same sequences. Hydroxynaphtol blue and SYBR GreenTM allowed direct naked-eye detection with identical sensitivity as agarose gel electrophoresis. We included the LAMP-based protocol in a rapid identification scheme of the respective pathogens, and all tested isolates were correctly identified within 2 to 3 h. In addition, both protocols...

Interference of tuberculosis on the performance of ELISAs used in the diagnosis of paratuberculosis in cattle BJM
Lilenbaum,Walter; Ferreira,Rachel; Marassi,Carla Dray; Ristow,Paula; Oelemann,Walter Martins Roland; Fonseca,Leila de Souza.
Forty-four cows from five herds infected with tuberculosis (TB) and without paratuberculosis (PTB), and 21 cows from a herd without either infection were studied. The cattle presented concordant results in both the skin test and gamma-interferon assay for TB and two commercial ELISAs for PTB. Animals were divided according to TB test results into Group A with 28 TB-infected animals, Group B with 16 TB-negative animals from infected herds, and Group C with 21 TB-negative cows from a tuberculosis-free herd (which were used as controls). Twenty of 28 animals from Group A (71.4%), 6/16 from Group B (37.5%) and none from Group C were reactive to PTB ELISAs, suggesting that these commercial kits were unable to distinguish between PTB and TB. It is proposed that...

Lipoarabinomanano (LAM) de Mycobacterium spp: Respuesta inmune inducida en terneros InVet
Jolly,A.; Colavecchia,S.; Jar,A.; Fernández,E.; Bernardelli,A.; Mundo,S..
