Genetic divergence among hybrids of 'Cravo' mandarin with 'Pêra' sweet orange Scientia Agricola

Molecular markers have been used as tools in breading programs of sexual hybridation, allowing the genetic characterization of a large number of genotypes. The RADP markers are the most used since the employed techniques are simple and of low cost. To evaluate the genetic divergence among F1 hybrids of 'Cravo' mandarin (Citrus reticulata Blanco) and 'Pêra' sweet orange (C. sinensis (L.) Osbeck), this study analyses the variability and similarity of the hybrids among themselves and with their parents. Sorghum anthracnose, caused by Colletotrichum sublineolum is a serious disease in Brazil. The fungus is highly variable pathogenically with many physiological races. The present study was made to evaluate the usefulness of molecular methods (SDS-PAGE, RAPD, ARDRA and rDNA sequencing) for identifying C. sublineolum pathotypes isolated from sorghum. Although protein profile (SDS-PAGE) showed a relative low level of polymorphism, differences could be observed due to presence/absence of some polypeptides as well as to intensity of some bands among pathotypes. Genetic Diversity among Flue-cured Tobacco Cultivars Based on RAPD and AFLP Markers BABT

The aim of this work was to study the genetic diversity among flue-cured tobacco cultivars. RAPD and AFLP analyses were used to assess the genetic similarity among selected accessions of flue-cured tobacco. Seventy eight RAPD and 154 AFLP polymorphic bands were obtained and used to assess the genetic diversity among 28 tobacco accessions. The cultivar relationships were estimated through the cluster analysis (UPGMA) based on RAPD data and AFLP data. The accessions were grouped into three major clusters and these shared common ancestry clustered together.

Genetic diversity among forty coffee varieties assessed by RAPD markers associated with restriction digestion BABT

The genetic variability of 40 accessions of_C. arabica was evaluated using a combination of the random amplified polymorphic DNA (RAPD) technique and restriction digestion of genomic DNA. The genetic variability and the relatedness among all accessions were initially evaluated using 195 RAPD primers which revealed a very low level of genetic variation. To improve the efficiency in the detection of polymorphism, the genomic DNA of all accessions were submitted to digestion with restriction endonucleases prior to PCR amplification. The fungus Stemphylium solani causes leaf blight of tomato (Lycopersicon esculentum) in Brazil. In recent years, severe epidemics of a new leaf blight of cotton (Gossipium hyrsutum) caused by S. solani occurred in three major cotton-growing Brazilian states (PR, MT and GO). Molecular analysis was performed to assess the genetic diversity among the S. solani isolates from cotton, and to verify their relationship with representative S. solani isolates from tomato. The genetic relationships among 81 maize accessions consisting 79 landraces and two improved varieties, maintained by farmers in southern Brazil were investigated using Random Amplified Polymorphic DNA (RAPD). Thirty-two highly informative primers amplified 255 markers of which 184 (72.2%) were polymorphics. Based on the RAPD markers, a dendrogram was constructed using the UPGMA method. The range of genetic similarity was from 0.78 to 0.91. The molecular data grouped the accessions into two main clusters, which were correlated according to kernel colors. Trichosporon asahii is an opportunistic pathogen, associated with a high mortality rate in immunocompromised patients. In this study, ten isolates, recovered from oral cavity and urine of patients in Intensive Care Units (ICU) over six months, were identified by classical and molecular methods, typed by RAPD and tested in vitro for susceptibility to fluconazole, itraconazole, 5-flucytosine and amphotericin B. Oenocarpus mapora is an Amazonian palm species commonly used by native populations for food and in folk medicine. We measured genetic variability, using RAPD markers, of material kept in a germplasm bank composed of accessions sampled from the Brazilian Amazon. These included 74 individuals from 23 accessions sampled from 9 localities in three States of the Brazilian Amazon. Jaccard genetic similarities were calculated based on 137 polymorphic bands, amplified by 15 primers. This article takes an ecological approach to the genetic diversity of Rosewood (Aniba rosaeodora Ducke) in a central Amazonian terra firme forest north of Manaus. Planted Rosewood setting, under partial shaded canopy, were assessed in terms of fruiting production, frugivory, and seed dispersal. Using RAPD molecular analysis procedures, the influence of the spatial distribution of adult trees on the genetic diversity (polymorphism) of saplings was assessed with genetic samples from 34 reproductive trees and 60 saplings. Genetic diversity of guava (Psidium guajava L.) from Central Mexico revealed by morphological and RAPD markers Phyton

Guava fruit produced in Calvillo, Aguascalientes (Mexico) is considered to be of the best quality in this country. Nevertheless, growers from this place empirically know that there is a noticeable variation among individual trees within the same orchard, and variation among individuals from different orchards. In order to have a clear evidence of this, morphology analysis of guava was performed taking data from seventy nine individuals out of thirty six orchards, while RAPD was performed on a subset of twenty six individuals. Similarity was found for morphology data ranging from 87-100%, while similarity from RAPD data ranged from 30-100%. Combined data of RAPD and morphology showed similarity greater than 80%. In this study, random amplified polymorphic DNA (RAPD) markers were applied to analyze the genetic diversity of samples of the Neotropical catfish Hypostomus ancistroides, collected from four sites (S1, S2, S3 and S4) along an urban stream in Southern Brazil. The 11 primers used in RAPD analysis amplified 147 loci, 76 (51.7%) of which were polymorphic. The proportions of polymorphic loci observed in the four samples were: 29.93% (S1), 31.97% (S2), 23.81% (S3) and 38.77% (S4). The average heterozygosity within sampling localities ranged from 0.1230 to 0.1526 and unbiased genetic distances ranged from 0.0253 to 0.0445. The analyze of 98 P. griseola isolates collected at Damolandia and Inhumas county, State of Goias, Brazil, revealed great genetic diversity. According to the method used and at a distance level of 62,5%, isolates were clustered in 4 groups. A non linkage among RAPD markers and the pathotypes used in this study was observed.

Genetic diversity of the common bean Phaseolus vulgaris L. determined by DNA-based molecular markers. Repositório Alice

Weselected 28 potential common bean (Phaseolus vulgaris L.) progenitors to determine their genetic diversity using RAPD technique. They were divided into two groups based on their type of storage protein (phaseolin); 18 of them had type "S" phaseolin and the other 10 had phaseolin. DNA samples of all the cultivars were amplified in vitro with oligonucleotide primiers of random sequences. A total of 276 amplification products, 144 of them being polymorphic, were obtained with 45 different primers. Multivariate analysis of these data divided the cultivars into groups and, in addition, allowed a more precise intra and intergroup distinction.
