Chicken embryos kept in culture medium were bombarded using a high helium gas pressure biolistic device. To optimize the factors that affect transformation efficiency, the lacZ gene under control of the human cytomegalovirus immediate early enhancer/promoter was used as a reporter gene. There was an inverse relationship between survival rate and transformation efficiency. The best conditions obtained for high embryo survival and high transformation efficiency were achieved with 800 psi helium gas pressure, 500 mmHg vacuum, gold particles, an 8 cm DNA-coated microparticle flying distance to the embryo and embryo placement 0.5 cm from the center of the particle dispersion cone. Duas linhagens de bactérias fitopatogênicas do gênero Pseudomonas e duas do gênero Erwinia produtoras de bacteriocinas, foram submetidas a diferentes tratamentos visando estimar a estabilidade desse caráter. Houve eliminação da produção de bacteriocinas após tratamento com duas concentrações de brometo de etídio e a porcentagem de eliminação variou com o hospedeiro e com a concentração da droga. Não houve eliminação em temperaturas elevadas e também a preservação da linhagem por três anos em laboratório não causou perda do caráter. A eliminação da produção de bacteriocinas com brometo de etídio sugere que os genes envolvidos tenham localização plasmidiana. |

Citrus black spot (CBS) is a plant disease of worldwide occurrence, affecting crops in Africa, Oceania, and South America. In Brazil, climate provides favorable conditions and CBS has spread to the Southeast and South regions. CBS is caused by the fungus Guignardia citricarpa (anamorph: Phyllosticta citricarpa) and its control is based on the use of fungicides, such as benzimidazoles. In South Africa, the disease was kept under control for 10 years with benomyl, until cases of resistance to high concentrations of this fungicide were reported from all citrus-producing areas. This study was developed to evaluate selection effects on gene frequencies in a synthetic beef cattle cross (5/8 Charolais, 3/8 Zebu) named Canchim. A sample of 154 Canchim animals, representing three generation classes, was analyzed for seven molecular markers. Thirty-six Charolais cattle were also studied for comparison. A highly significant variation (P < 0.01), with a linear increase of the allele coding for valine at position 127 of the growth hormone peptide, was observed through generations. This allele was found in the Charolais sample but is not observed in Brazilian Bos indicus breeds.
