Animal preparation prior to artificial insemination (AI) in goats usually involves a combination of intravaginal devices containing progesterone/progestagens plus PGF2?? analogues and gonadotropins, being eCG the most used. The use of intravaginal devices soaked in synthetic progesterone analogues requires milk discard for 60 days. Progesterone soaked intravaginal devices, as synthetic ones, are associated with more animal handling and discomfort. In addition, it leads to environmental impacts because it generates more residues. On the other hand, eCG has presented continuous restriction due to animal welfare issues associated with its production (Vilanova et al., Animals, 9:1053, 2019) and it has been substituted by hCG for this purpose. This study tested... |

Abstract: The use of B-mode and color Doppler ultrasonography may replace the use of surgical methods (laparoscopy or laparotomy) in the evaluation of the superovulatory response in sheep, with an accuracy around 74% and 82%, respectively (Oliveira et al. 2018. Reprod Dom Anim, 53:742-750). The ultrasound determination of ovarian response in embryo donor goats still needs to be proven, especially because the non-invasive technique is mandatory when the embryo collection is done by transcervical route. The aim of this study was to correlate the quantification of corpora luteal (CLs) determined by Bmode ultrasound evaluation with the number of recovered structures obtained by transcervical embryo collection of superovulated goats. In view of the above, it is... |

In vivo embryo production performs an important role in small ruminants, however the efficiency of this biotechnology has high variability. In this context, the occurrence of luteal dysfunction is exacerbated in superovulated females (Rodriguez et al., 2015) and it is an unwanted response wich may be related to low embryo production. Thus, it is possible to highlight the need to expand knowledge related to the corpora lutea functionality, as well as determinative tools for ultrasound diagnosis. The aim of this study was to test correlations between biometric variables, luteal vascularization and serum progesterone concentrations. In conclusion, luteal vascularization is an indicative of corpus luteum functionality in superovulated ewes. |

Abstract: The primary goal of this study was to employ ultrasonography to examine the ovaries of ewes undergoing superovulatory treatment for correlations between antral follicular blood flow and ovarian responses/embryo yields. Five Santa Inês ewes were subjected to a short- (Days 0?6, Group 1) and five to a long-term progesterone-based protocol (Days 0?12, Group 2) to synchronize estrus and ovulations after the superovulatory treatment. Porcine FSH (pFSH, 200 mg) was administered in 8 decreasing doses over 4 days, starting on Days 4 and 10 in Groups 1 and 2, respectively. After CIDR removal, all ewes were bred by a ram and embryos were recovered surgically 7 days later. Transrectal ovarian ultrasonography was performed the day before and on all 4 days of... |

Abstract: The objective was to evaluate if supplemental LH given at the end of FSH treatment would synchronize the time of ovulation and increase the ovulation rate and embryo yield in Santa Ines ewes. Twenty superovulatory (SOV) programs were accomplished in cross-over design (60d interval). On D0, a CIDR device was inserted, and the device was replaced with a new one 7 days later, when 37.5µg of d-cloprostenol was administered. On D12, we started the SOV treatment, administering 256mg of pFSH 8 times, 12h apart. On D14, the CIDR was removed, and 200IU of eCG and 37.5µg of d-cloprostenol were administered. On D15, the ewes were allocated into one of two groups, a Control group (n=10) that received no supplemental LH and a LH group (n=10) treated with... |

Abstract: The compatibility between the embryo development stage and the estrous cycle moment of the embryo recipient is an important factor in the multiple ovulation and embryo transfer (MOET) program. This aspect contributes significantly to the pregnancy establishment. Thus, the present study aimed to evaluate the effect of estrus synchronization and superovulation protocol on the distribution of ovine embryos at different stages of development. In conclusion, the protocol of estrus synchronization and superovulation was efficient in the synchronization of ovulation, which reflected in the prevalence of embryos in the stage of compact morula, when the collection is performed between days D6 and D7 after the first natural mating. This time relative do... |

For the success of an embryo transfer program, it is necessary to achieve a great synchronization between the embryo donor and recipients. For this, hormonal protocols are commonly used, aiming at a greater number and quality of the viable products, in a shorter period of time. The aim of this study was to assess the effect of the hormonal protocol of estrus synchronization and superovulation in goats on the quality and stage of embryos obtained. In conclusion, the hormonal protocol used for superovulation of goats was effective enough to guarantee embryos in adequate stages and quality for fresh embryo transfer and cryopreservation, avoiding both undesired very young and hatched embryos. Abstract: The objective of this study, was to evaluate the exogenous FSH dose effect on gonadotrophic treatment over ewes ovulatory follicle dynamics. Twenty four Santa Inês ewes were submitted to estrus synchronization with intravaginal progesterone device (CIDR®, Pfizer, Hamilton, New Zealand) inserted on Day 0 and remaining until the Day 8. On Day 0 and Day 8 were intramuscular (IM) administered 0.125 mg of PGF2? synthetic analogue (Sincrocio®, Ouro Fino, Cravinhos, Brazil). The gonadotrophic treatment started 48 hours before the progesterone removal (Day 6) when females were randomly allocated into three experimental groups according to the total dose of exogenous porcine FSH (Folltropin-V®, Bioniche, Belleville, Canada): G200 (n = 8) - 200 mg; G133 (n... |

Abstract: The effect of hCG administration on accessory corpus luteum (ACL) formation, CL area, and plasma progesterone (P4) concentration (ng/mL) seven days after breeding was studied in nulliparous Santa Inês sheep. Intravaginal 60 mg MAP sponges were inserted into ewes for six days and 300 IU eCG i.m. and 30 ?g d-cloprostenol latero-vulvar were administered 24 h before sponge removal. Ewes were naturally bred and, seven days after first mating (Day 0; D0), were treated with either 250 IU hCG (hCG group; n = 7) or 1 mL saline solution (control group; n = 7). Blood was collected to determine plasma P4 concentrations and sonograms were performed on Days 7, 10, 13, 16, 19, and 22. Number of CL on D7 was similar (P > 0.05) between hCG (1.3 ± 0.5) and... |
