A study on the production of Bacillus thuringiensis on different culture media, for insect control. Repositório Alice
CAPALBO, D. M. F.; MORAES, I. O..
Bacillus thuringiensis has highly specific insecticidal activity and holds considerable advantage over chemical controls which often have long residual action and may be toxic to nontarget organisms. In order to carry out large-scale field trials with that bioinsecticide, its sufficient local supply must be available. This study was undertaken to determine a suitable fermentation medium composition and optimal conditions for large-scale, low-cost production of B. thuringiensis. Submerged fermentation process was proposed utilizing liquid agroindustrial residues like sugar-cane molasses, coconut water, liquid residue from beer industry and the wastewater from cassava flour industry. Temperature, aeration and agitation were controlled. Analysis of pH,...

ANTAGONISM AGAINST VIBRIO CHOLERAE BY BACTERIAL DIFFUSIBLE COMPOUND IN THE FECAL MICROBIOTA OF RODENTS Rev. Microbiol.
Silva,Simone Helena da; Vieira,Enio Cardillo; Nicoli,Jacques Robert.
In an ex vivo agar plate assay, we monitored the appearance of an inhibitory halo against Vibrio cholerae from the feces of Wistar and Fischer rats aged 10 to 42 days. The frequency of Wistar rats showing halo increased from 0% (10 days) to a maximum of 80.0% (29 days) and then decreased to 53.3% (42 days). A similar pattern was obtained with Fischer rats but with a lower intensity (maximum frequency of 50.0% by day 36). In a separate experiment, when Wistar rats were fed a low-protein diet for 7 days, the inhibitory halo decreased drastically. Three apparently different colony morphologies were isolated from the dominant fecal microbiota: a facultative anaerobe (FAN) and two strict anaerobes (SAN). The ex vivo inhibitory test showed a halo around the...

LANDA B. B.; HERVAS, A.; BETTIOL, W.; JIMENEZ-DIAZ, R.M..
The antagonistic activity against in vitro growth of Fusarium oxysporum f.sp. ciceris was determined for 74 bacterial isolates obtained from the rhizosphere of chickpeas grown in two field soils with different histories of Fusarium wilt, and for seven isolates of Pseudomonas spp. from culture collections. Twenty-four isolates of Bacillus spp. and Pseudomonas chlororaphis 30-84 showed a strong antagonism against three races (0, 1 and 5) of F.o. ciceris tested. Three selected Bacillus isolates and P. chlororaphis 30-84 were further tested against 30 isolates of races 0, 1 and 5 of F.o ciceris, races 0,1 and 2 of F.o. melonis, F.o. Phaseoli and nonpathogenic F. oxysporum. Bacillus isolates differed in their antagonistic activity and were less inhibitory to...

Antagonistic and protective effects against Salmonella enterica serovar typhimurium by Lactobacillus murinus in the digestive tract of gnotobiotic mice BJM
Vasconcelos,Ana Luiza Sampaio; Nicoli,Jacques Robert; Nardi,Regina Maria Drummond.
In the present study, production of antagonistic substances by Lactobacillus murinus against enteropathogenic bacteria was evaluated in vivo as well as a possible protective effect against an oral challenge with Salmonella Typhimurium using a gnotobiotic animal model. A higher mean survival time (P < 0.05) was observed in L. murinus-associated animals (7.89 ± 3.83 days) when compared with the control ones (4.44 ± 0.73 days). Lactobacillus murinus exerted a potent antagonistic effect in vivo against S. sonnei and to a lesser extent against S. Typhimurium as revealed by inhibitory halos around the feces of Lactobacillus-associated animals. Diameters of inhibitory halos around intestinal contents increased along the intestine tract, following...

Mendoza,José Luis Hernández; Pérez,María Isabel Sánchez; Prieto,Juan Manuel González; Velásquez,Jesús DiCarlo Quiroz; Olivares,Jesús Gerardo García; Langarica,Homar Rene Gill.
Abstract Sampling of agricultural soils from the Mexican northeastern region was performed to detect Trichoderma spp., genetically characterize it, and assess its potential use as a biologic control agent against Macrophomina phaseolina. M. phaseolina is a phytopathogen that attacks over 500 species of cultivated plants and causes heavy losses in the regional sorghum crop. Sampling was performed immediately after sorghum or corn harvest in an area that was approximately 170 km from the Mexico-USA border. Sixteen isolates were obtained in total. Using colony morphology and sequencing the internal transcribed spacers (ITS) 1 and 4 of 18S rDNA, 14 strains were identified as Trichoderma harzianum, T. koningiopsis and T. virens. Subsequently, their antagonistic...

Endophytes are being considered for use in biological control, and the enzymes they secrete might facilitate their initial colonization of internal plant tissues and direct interactions with microbial pathogens. Microbial proteases are also biotechnologically important products employed in bioremediation processes, cosmetics, and the pharmaceutical, photographic and food industries. In the present study, we evaluated antagonism and competitive interactions between 98 fungal endophytes and Alternaria alternata, Colletotrichum sp., Phyllosticta citricarpa and Moniliophthora perniciosa. We also examined the proteolytic activities of endophytes grown in liquid medium and conducted cup plate assays. The results showed that certain strains in the assemblage of...

Antifungal potential of crude extracts of Trichoderma spp. Biota Neotropica
Marques,Eder; Martins,Irene; Mello,Sueli Correa Marques de.
Abstract Antibiosis is the mechanism by which certain microorganisms respond to the presence of others, secreting compounds or metabolites capable of inhibiting or impeding their development. The crude extract of Trichoderma contains a mixture of secondary compounds, which may show antibiotic effect, and has been used for the prospect of this fungus for biological control and other industrial purposes. Faced with the increasing demand of agriculture for ecologically compatible alternatives for the management of diseases, this work aimed to investigate the spectrum of action of Non-Volatile Metabolites (NVMs) of Trichoderma isolates against different plant pathogenic fungi. The antagonistic potential of NVMs was evaluated through the incorporation method of...

Bioassay for selection of biocontroller bacteria against bean common blight (Xanthomonas axonopodis pv. phaseoli) BJM
Zanatta,Zarela G.C.N.; Moura,Andréa B.; Maia,Luciano C.; Santos,André S. dos.
Emphasis has been given on selection of micro-organism for biological control. However, in order to evaluate the biological control potential of a great number of micro-organisms in a small period of time it is necessary to develop an efficient bioassay. Seven hundred and sixty bacterial isolates from different habitats, were selected for compatibility with Rhizobium leguminosarum bv. phaseoli (SEMIA 4077 e SEMIA 4080). Among them 596 isolates were ineffective against both rhizobia. Bean seeds immersed in suspension of each one of these isolates were agitated for 5 hours at 10ºC and sowed in non-sterilized soil. The plants were kept in greenhouse. After the development of cotyledonary and primary leaves, these were removed and bioassayed for Xanthomonas...

Characterization of Bacillus isolates of potato rhizosphere from andean soils of Peru and their potential PGPR characteristics BJM
Calvo,Pamela; Ormeño-Orrillo,Ernesto; Martínez-Romero,Esperanza; Zúñiga,Doris.
Bacillus spp. are well known rhizosphere residents of many crops and usually show plant growth promoting (PGP) activities that include biocontrol capacity against some phytopatogenic fungi. Potato crops in the Andean Highlands of Peru face many nutritional and phytophatogenic problems that have a significant impact on production. In this context is important to investigate the natural presence of these microorganisms in the potato rhizosphere and propose a selective screening to find promising PGP strains. In this study, sixty three Bacillus strains isolated from the rhizosphere of native potato varieties growing in the Andean highlands of Peru were screened for in vitro antagonism against Rhizoctonia solani and Fusarium solani. A high prevalence (68%) of...
