Using double-strand RNA (dsRNA) high-throughput sequencing, we identified five RNA viruses in a bean golden mosaic virus (BGMV)-resistant common bean transgenic line with symptoms of viral infection. Four of the identified viruses had already been described as infecting common bean (cowpea mild mottle virus, bean rugose mosaic virus, Phaseolus vulgaris alphaendornavirus 1, and Phaseolus vulgaris alphaendornavirus 2) and one is a putative new plant rhabdovirus (genus Cytorhabdovirus), tentatively named bean-associated cytorhabdovirus (BaCV). Tbe rep gene of Bean golden mosaic virus (BGMV) is essential for virus replication, and rep gene rnutations with amino acid codons cbanges in the putative NTP-binding rnotif D262R (MI) was created. Phaseolus vulgaris rransforrnation was achieved with a helium driven particle acceleration device. Tbe vector contained the bar gene [resistance to arnmonium glyphosinate (AG)] and besides the rep gene mutant. A total of J 7 initial - R(O) - transformants were analyzed. From the R(O) plant M1/4, which had mutation MI, six progeny plants with resistance to AG were obtained, and four remained BGMV symptomless after inoculation. Genetic diversity of begomoviruses infecting soybean, bean and associated weeds in Northwestern Argentina Trop. Plant Pathol.

The subtropical Northwestern region of Argentina (provinces of Tucumán, Salta, Jujuy, Santiago del Estero and Catamarca) suffers from a high incidence of the whitefly Bemisia tabaci, and the detection of begomoviruses is also common. The Northwest is the main bean-growing region of the country, and approximately 10% of Argentina's soybean crop is grown in this area. We have used a PCR-based assay to establish the identity and genetic diversity of begomoviruses associated with bean and soybean crops in Northwestern Argentina. Since the 1990s geminiviruses have emerged as devastating pathogens in tropical and subtropical regions as well as in temperate regions. Recent studies accomplished molecular hybridization-based detection of three begomoviruses infecting soybean and bean crops in Argentina, Bean golden mosaic virus (BGMV), Soybean blistering mosaic virus (SbBMV) and Tomato yellow spot virus (ToYSV). The aims of the present study were to determine incidence and prevalence of the three known geminiviruses in soybean and bean crops in Argentina following the same procedure and to characterize the sites where Bemisia tabaci-geminivirus complex was found based on climate data. A new bipartite begomovirus (family Geminiviridae) was detected on cowpea (Vigna unguiculata) plants exhibiting bright golden mosaic symptoms on leaves under field conditions in Brazil. Complete consensus sequences of DNA-A and DNA-B components of an isolate of the virus (PE?088) were obtained by nanopore sequencing and confirmed by Sanger sequencing. The genome components presented the typical genomic organization of New World (NW) begomoviruses.
