Disinfection and in vitro establishment of Acacia mearnsii nodal segments PFB - Pesquisa Florestal Brasileira

Due to low rates of rooting it is difficult to propagate selected clones of Acacia mearnsii by cuttings. The purpose of this study was to define a protocol for disinfection of nodal segments for in vitro introduction, aiming to improve the species genetically for forest plantation. Combination of different products concentration (70% ethyl alcohol, sodium hypochlorite, plant preservative mixture and mercuric chloride), immersion period and size of nodal segment were tested. The best disinfection treatment was the combination of 70% ethanol, 0.4% mercuric chloride, sodium hypochlorite at 2% in nodal segments of 10 mm, with 80% survival.

The Cerambycidae Oncideres impluviata (Germar, 1823) is an important insect pest for Acacia mearnsii De Wild in Southern Brazil. The damage caused by their girdling behavior reduces tree productivity, specially in the early years of plant establishment, when girdling is performed on the main trunk of trees. Here, we used a fragment of the mtDNA COI gene to analyze the genetic diversity, population structure and demography of O. impluviata in Southern Brazil, as well as to present the first hypothesis of phylogenetic relationships among species of the genus Oncideres. Induction of leafy galls in Acacia mearnsii De Wild seedlings infected by Rhodococcus fascians BABT

Plantlets of blackwattle (Acacia mearnsii De Wild) were inoculated with the bacterium Rhodococcus fascians and cultured in vitro. Leafy galls appeared at the cotyledonary nodes in 75% of the infected plants. The galls were separated from the plants and cultured on a medium containing three-quarters-strength MS salts (Murashige and Skoog, 1962), MS vitamins, 2% sucrose and an antibiotic (cephalothin), supplemented with or without 0.2% activated charcoal. Histological studies conducted from the sixth to the twenty-second day after plant infection revealed the presence of newly formed meristematic centers, first in the axillary region, then on the petioles and lamina of the leaflets around the apical meristem. Selecting superior individuals requires a careful genetic evaluation. For this reason, repeated measurements of biometric traits need to be carried out in the same individual. This index is a strategic tool aimed to assist in providing accurate observations and further selection. The objectives of this study were to estimate the repeatability coefficient in many seed traits for black wattle clones; test the best method to estimate the optimal number of measurements required; and to analyze genetic dissimilarity.
