The objective of this work was the optimization of the conditions of in vitro culture for callus production of Silybum marianum (L.) Gaertn. (Asteraceae). Sections of cotyledons, previously disinfected by washing successively with ethanol 70º, NaClO (10% w/v) and Tween 20 (0.05% v/v) and rinsing with sterile distilled water, were used as explants. For its initial culture, B5 medium supplemented with BA and 2,4-D solidified with phytagel was used, and a 63% survival was achieved. To obtain callus, two solid media were assayed (S1 and S2) using B5 medium supplemented with growth regulators (BA and 2,4-D or NAA and BA, respectively). A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. The combinations and concentrations of PGRs were shown significant variations for the frequency of callus formation, appearence of callus and the potential of callus differentiation. ABSTRACTThe present investigation was aimed to detect the specific polypeptide(s) appeared during the sequential stages of differentiation. Among different explants, only nodal explants showed good results for callusing. To define preliminary aspects of walnut (Juglans regia L.) callogenesis in indirect propagation systems, this work analyzed callogenic induction in adult leaves of walnut in broadleaf trees (BTM), Murashige and Skoog (MS) Driver Kuniyuki Walnut (DKW) and Woody Plant (WPM) media. Leaf segments of 1 cm², previously sterilized, were placed in each culture medium, supplemented with 21.4 µM naphtalene acetic acid (NAA) and 8.8 µM 6-benzylaminopurine (BAP). A method for the isolation of coupled mitochondria from the callus of Araucaria angustifolia is described for the first time. Mitochondria were isolated from embryogenic callus of A. angustifolia. They were metabolically active, able to sustain oxidative phosphorylation as shown by respiratory control ratio values, which were about 2.4 when respiring on succinate as substrate. Oxygen uptake experiments, using freeze-thawed disrupted mitochondria, showed the presence of alternative rotenone-insensitive NAD(P)H dehydrogenases, which were stimulated by Ca2+. The procedure now described for the isolation of A. angustifolia mitochondria is an important new tool, allowing the investigation of mitochondrial bioenergetics and metabolism and physiology of plants. |

In the present study, in vitro mutagenesis techniques were applied to investigate the effects of gamma irradiation at 0, 10, 20, 30, 40, 50 and 60 Gy on physiological changes in callus of Gerbera jamesonii Bolus ex. Hook f. Biochemical changes in chlorophyll and soluble protein content of pre- and post- irradiated Gerbera callus were studied. Non-irradiated callus demonstrated the highest amount of chlorophyll content as compared to callus irradiated at 10, 20, 30, 40, 50 and 60 Gy. Callus growth and plant regeneration from long-term callus cultures were studied in two elite clones of Asparagus officinalis cv. Argenteuil, to establish a suitable protocol for a prospective in vitro selection program. Callus initiation and growth was evaluated on MS medium with 3% sucrose, 0.9% agar, 1 mg.l-1 kinetin, and three levels of 2,4-D. The highest callus relative growth was obtained on medium with 1.5 mg.l-1 2,4-D and 1 mg.l-1 kinetin.
