The continuous production and release of chemicals into the environment has led to the need to assess their genotoxicity. Numerous organophosphorus compounds with different structures have been synthesized in recent years, and several oxaphosphole derivatives are known to possess biological activity. Such chemical compounds may influence proliferating cells and cause disturbances of the genetic material. In this study, we examined the cytotoxicity and genotoxicity of 4-bromo-N,N-diethyl-5,5-dimethyl-2,5-dihydro-1,2-oxaphosphol-2-amine 2-oxide (Br-oxph). In A. cepa cells, Br-oxph (10-9 M, 10-6 M and 10-3 M) reduced the mitotic index 48 h after treatment with the two highest concentrations, with no significant effect at earlier intervals. Mercury is a xenobiotic metal that is a highly deleterious environmental pollutant. The biotransformation of mercury chloride (HgCl2) into methylmercury chloride (CH3HgCl) in aquatic environments is well-known and humans are exposed by consumption of contaminated fish, shellfish and algae. The objective of the present study was to determine the changes induced in vitro by two mercury compounds (HgCl2 and CH3HgCl) in cultured human lymphocytes. Short-term human leukocyte cultures from 10 healthy donors (5 females and 5 males) were set-up by adding drops of whole blood in complete medium. Evaluation of the genotoxic potential of the Hypericum brasiliense (Guttiferae) extract in mammalian cell system in vivo Genet. Mol. Biol.

Plants of the genus Hypericum, long used in folk medicine, contain active compounds which present, anti-septic, diuretic, digestive, expectorant, vermifugal, anti-depressive and other properties. The possible clastogenic effect of a H. brasiliense extract was tested in vivo on the bone marrow cells of Wistar rats. The extract was administered by gavage at doses of 50, 150 and 300 mg/kg body weight. Experimental and control animals were submitted to euthanasia 24 h after the treatment for micronucleus (MN) and chromosome preparations. H. brasiliense extract did not induce statistically significant increases in the average numbers of MN or chromosome aberrations in the test systems employed.

In vivo evaluation of the mutagenic potential and phytochemical characterization of oleoresin from Copaifera duckei Dwyer Genet. Mol. Biol.

We characterized the chemical constituents of Copaifera duckei oleoresin and used dermal application to Wistar rats to evaluated its possible mutagenic and cytotoxic activities on peripheral blood reticulocytes and bone marrow cells. Chemical characterization of the oleoresin revealed the presence of sesquiterpene hydrocarbons, an unidentified neutral diterpene and diterpene acids. To evaluate mutagenicity evaluation the rats were treated with 10, 25 and 50% of the LD50 dose of the oleoresin for three consecutive days and peripheral blood collected after 0, 24, 48 and 72 h for micronucleus analysis. Thyroid hormones stimulate aerobic metabolism which may lead to oxidative stress accompanied by damage to various cellular macromolecules, including DNA. Extracts of the fruits of Caesalpinia ferrea Mart. (Leguminosae) are widely consumed in folkloric medicine in Brazil and several other countries without any genetic toxicity evaluation. In this study we investigated, the clastogenic and cytotoxic potential of the crude aqueous extract of the fruits of C. ferrea in Wistar rat bone marrow cells using the micronucleus and chromosomal aberration test systems. The objective of the present study was to determine the frequency of somatic chromosomal anomalies and Y chromosomal microdeletions (azoospermia factor genes, AZF) in infertile males who seek assisted reproduction. These studies are very important because the assisted reproduction techniques (mainly intracytoplasmic sperm injection) bypass the natural selection process and some classical chromosomal abnormalities, microdeletions of AZF genes or some deleterious genic mutations could pass through generations. Biological dosimetry (biodosimetry) is based on investigations of induced biological effects (biomarkers) in order to correlate them with radiation dose. Among the indicators employed in biodosimetry, scoring of chromosome aberrations is the most reliable method to quantify individual exposure to ionizing radiation. The technique, applied to circulating lymphocytes, has been developed into a routine procedure to evaluate the dose in the case of real or suspected accidental exposure. Мультиаберрантные клетки и пикноз ядер у гидробионтов из района с повышенным содержанием естественных радионуклидов IBSS Repository

Исследованы аберрации хромосом в клетках гидробионтов на о. Икария в Эгейском море, где вблизи гидро­термальных источников наблюдается повышенное содержание естественных радионуклидов. У ракообраз­ных и червей в этих районах обнаружены наиболее высокий уровень хромосомного мутагенеза, мультиабер-рантные клетки и пикноз ядер. Предполагается, что генотоксические эффекты индуцированы комплексным влиянием низкой величины рН, высокой температуры воды и повышенного содержания естественных ра­дионуклидов.
