Assessment of genetic variability within and among coffee progenies and cultivars using RAPD markers Genet. Mol. Biol.

The RAPD technique associated with restriction digestion of genomic DNA was used to assess the genetic variability within and among nine populations of Coffea arabica, including six progenies belonging to the Sarchimor germplasm, the progeny PR 77054-40-10 (Catuaí Vermelho IAC 81 x Icatu), and two commercial cultivars (IAPAR 59 and Catuaí Vermelho IAC-81). These populations were evaluated using analysis of molecular variance (AMOVA), genetic similarity among progenies, and percentage of polymorphic loci. A total of 99 RAPD markers were evaluated of which 67 (67.67%) were polymorphic. AMOVA showed that 38.5% and 61.5% of the genetic variation was distributed among and within populations, respectively. Breeding potential and genetic diversity of "híbrido do timor" coffee evaluated by molecular markers. Repositório Alice

AFLP, RAPD and SSR molecular markers were used to study the genetic diversity and genetic structure of the Híbrido de Timor germplasm. The principal coordinate analysis, UPGMA cluster analysis based on genetic dissimilarity of Jaccard, Bayesian model-based cluster analysis, percentage of polymorphic loci, Shannon?s information index and Nei gene diversity were employed to assess the genetic diversity. The analyses demonstrated a high genetic diversity among Híbrido de Timor accessions. UPGMA and Bayesian cluster analyses grouped the accessions into three clusters. The genetic structure of Híbrido de Timor is reported. The management of Híbrido de Timor germplasm variability and its potential use in breedin programs are discussed.

Caffeine inheritance in interspecific hybrids of Coffea arabica x Coffea canephora (Gentianales, Rubiaceae) Genet. Mol. Biol.

Caffeine inheritance was investigated in F2 and BC1F1 generations between Coffea arabica var. Bourbon Vermelho (BV) and Coffea canephora var. Robusta 4x (R4x). The caffeine content of seeds and leaves was determined during 2004 and 2005. Microsatellite loci-markers were used to deduce the meiotic pattern of chromosome pairing of tetraploid interspecific hybrids. Genetic analysis indicated that caffeine content in seeds was quantitatively inherited and controlled by genes with additive effects. The estimates of broad-sense heritability of caffeine content in seeds were high for both generations. Genetic relationship in Coffea species and parentage determination of interspecific hybrids using ISSR (Inter- Simple Sequence Repeat) markers Genet. Mol. Biol.

Inter-simple sequence repeat (ISSR) markers were used to evaluate genetic divergence among eight Coffea species and to identify the parentage of six interspecific hybrids. A total of 14 primers which contained different simple sequence repeats (SSR) were used as single primers or combined in pairs and tested for PCR amplifications. Two hundred and thirty highly reproducible fragments were amplified, which were then used to estimate the genetic similarity and to cluster the Coffea species and hybrids. High levels of interspecific genetic variation were revealed. Partial resistance to fruit necrosis associated to Colletotrichum spp. among arabic coffee genotypes BABT

The aim of this study was to evaluate the resistance to fruit necrosis associated to Colletotrichum spp. in Coffea arabica L.. Twenty-two arabic coffee genotypes were performed in a randomized block design, with three replications and ten plants per plot. They were evaluated for the variables fruit necrosis, luminosity index inside of the plant canopy and fruits per productive nodes of the plants. There was genetic variability for the resistance to fruit necrosis. Different partial resistance levels, varying from highly susceptible (score = 3.89) to moderately resistant (score = 1.77) were observed. The genotypes that presented moderate resistance were the cultivars IPR-100, IPR-103, IPR-105 and IPR-108 and the coffee selection IAPAR-00023.

Performance of intraspecific hybrids (Kouillou x Robusta) of Coffea canephora Pierre. Repositório Alice

The experiment was carried out in 2005 in the municipality of Ouro Preto do Oeste at the experimental station of Embrapa Rondônia. 256 genotypes were evaluated, from nine hybridizations among parents of Kouillou (Cpafro 121, Cpafro 194, Cpafro 199 and Emcapa 03) and Robusta (Robusta 1675, Robusta 2258 and Robusta 640). The experimental design was a randomized complete block design, where each cross was represented by a maximum of 32 plants, distributed in four replications. The spacing used was 3.5 x 1.5 m. The average of six harvests and the yield of hulled coffee was 74.90 bags ha-1. Highlight for progeny is Emcapa 03 x Cpafro 194 with average yield of 97.71 bags ha-1.
