Prostaglandin F-2 alpha (PGF(2 alpha)) is the primary luteolysin in the cow, and luteal endothelin-1 (ET-1) interacts with PGF(2 alpha) during the process of luteolysis. In contrast, a developing corpus luteum (CL) is refractory to exogenous administration of PGF(2 alpha). Thus, the present study was aimed to investigate the functional relationship between ET-1 and PGF(2 alpha) in the mid-CL (PGF(2 alpha)-sensitive) and early-CL (PGF(2 alpha)-refractory). In the mid-CL model, cows (n = 6/treatment) were assigned to receive one of five types of treatments on day 10 of the estrous cycle: (1) an injection of saline; control, (2) a 500 jig of PGF2. analogue (sufficient dose to induce luteolytis); full-PG, (3) an intraluteal injection of 0.25 mg ET-1; ET-1, (4)... |

The aim of this study was to evaluate the potential of luteal echotexture (mean pixel value and heterogeneity), as a tool for assessing luteal function during different phases of the estrous cycle in Toggenburg goats. Sonographic evaluations of the ovaries were performed daily in nulliparous goats (n = 21), using a 5MHz linear rectal probe, commencing at estrus (day 0). Blood samples were collected daily for plasma progesterone RIA and images recorded on VHS tape and then digitized in TIFF format at a resolution of 1500×1125 pixels. A representative elementary area (REA) of 5625 pixels (0.31cm2) of these images was analyzed using custom-developed software, for mean pixel value and heterogeneity. Mean plasma progesterone, luteal area and pixels all reached... |

Polycystic ovarian syndrome (PCOS) is a low grade inflammatory disease characterized by hyperandrogenemia and chronic anovulation. C-reactive protein (CRP), released by adipocytes, plays a key role in PCOS. Apis mellifera honeybee venom (HBV) contains a variety of biologically active components with various pharmaceutical properties. This study was designed to assess the possibility of HBV application as an anti-inflammatory therapeutic agent. To induce PCOS, 1 mg/100 g body weight estradiol valerate (EV) was subcutaneously (SC) injected into eight-week-old rats. After 60 days, 0.5 mg/kg HBV was administered SC for 14 consecutive days, and the results of PCOS treatment were investigated. Rats were then anesthetized with chloroform, and their ovaries and... |

This study aimed to evaluate the Color Doppler ultrasound as a substitute for laparoscopy for couting of corpora lutea (CL) in superovulated sheep. In conclusion, the Color Doppler ultrasonography is highly efficient to estimate the number of CLs in superovulated ewes. This represents an important advance because it replaces invasive laparoscopic procedure, avoids fasting, drugs use and unnecessary handling in animals that did not respond to the treatment. Therefore, the Color Doppler ultrasound can replace the laparoscopy for the assessment of superovulated sheep. |

ABSTRACT: The equine chorionic gonadotropin (eCG) is a glycoprotein produced in mare endometrial calices. In bovine, it is used in estrus synchronization protocols. However, studies have shown that it is potentially immunogenic and its effect can decrease after repetitive use. This study aimed to evaluate antral follicle dynamics, corpus luteum (CL) and ovulation rate in bos indicus cows submitted to an estrus synchronization protocol in association with eCG for eight times consecutively. Ten cyclical, multiparous, and pasture raised beef cows were divided into two groups: control group (n=5) and eCG group (n=5). In 30 day interval, all animals were synchronized with the same estrogen/progesterone based protocol, totalizing 8 re-synchronizations. Cows in... |

We have shown that luteal blood flow increases in the peripheral vasculature of the mature corpus luteum (CL) prior to the onset of luteolysis in response to prostaglandin F2α (PGF2α) in the cow, but this phenomenon does not occur in the early CL. We therefore hypothesize that this acute increase of luteal blood flow occurs by vasodilation of large blood vessels due to local release of nitric oxide (NO). In the present study, we characterized the CL vasculature together with endothelial NO synthase (eNOS) expression during the estrous cycle in the cow. Immunohistochemistry was used to quantify the number of arteriolovenous vessels (surrounded with smooth muscle actin-positive smooth muscle cells), capillaries (with von Willebrand Factor-positive... |

Endothelin-1 (ET-1) is a luteolytic mediator in the bovine corpus luteum (CL), and its action appears to be via endothelin type A receptor (ETR-A). Thus, the aim of the present study was to determine the effect of ETR-A antagonist on PGF(2)alpha-induced luteolysis in the cow. Cows on days 1012 of the estrous cycle were subjected to five intraluteal injections of the ETR-A antagonist LU 135252 in saline or only saline at -0.5, 2, 4, 6, and 8 h after PGF(2)alpha administration (=0 h). Serial luteal biopsies were conducted to determine the expression of mRNA in the luteal tissue. There were no significant differences in the decrease in plasma progesterone (P) concentrations and the mRNA expressions of steroidogenic acute regulatory protein and 3... |

Abstract: The Doppler ultrasonography allows the characterization of blood flow and estimation of the functionality of reproductive organs. Therefore, it shows a great potential for use in multiple ovulation and embryo transfer (MOET) programs in ruminants. This study aimed to assess the effect of luteal blood flow and number of corpora lutea (CL) at the moment of embryo transfer to recipient on the establishment of pregnancy in sheep. In conclusion, both the subjective luteal blood flow and the number of CL did not influence the pregnancy rate in recipients of the Lacaune breed. [Efeito do fluxo sanguíneo luteal e número de corpos lúteos na taxa de gestação após transferência em tempo fixo de embriões criopreservados em receptoras Lacaune avaliadas por... |

To evaluate the effect of using urea in the diet on ovarian follicular dynamics and plasma urea and progesterone concentration, an experiment was carried out with 29 Alpine goats randomly distributed into four diets, each one containing 0%; 0.73%; 1.46% or 2.24% of urea on total dry matter. The follicular dynamics was monitored daily by ultrasonography. The estrous cycles were characterized by occurrence of two, three, four or five follicular waves. The emergence day of the first follicular wave in cycles with two waves, the length of the third wave and the maximum diameter of the largest follicle of the third wave in cycles with three waves presented a quadratic behavior according to the percentage of urea in the diets. In cycles with four waves, the urea... |

Abstract: The effect of hCG administration on accessory corpus luteum (ACL) formation, CL area, and plasma progesterone (P4) concentration (ng/mL) seven days after breeding was studied in nulliparous Santa Inês sheep. Intravaginal 60 mg MAP sponges were inserted into ewes for six days and 300 IU eCG i.m. and 30 ?g d-cloprostenol latero-vulvar were administered 24 h before sponge removal. Ewes were naturally bred and, seven days after first mating (Day 0; D0), were treated with either 250 IU hCG (hCG group; n = 7) or 1 mL saline solution (control group; n = 7). Blood was collected to determine plasma P4 concentrations and sonograms were performed on Days 7, 10, 13, 16, 19, and 22. Number of CL on D7 was similar (P > 0.05) between hCG (1.3 ± 0.5) and... |

Cell-to-cell interaction via cell contact-dependent pathway is essentially important for maintenance and regulation of corpus luteum (CL) integrity and its physiological actions. The objective of the present study was to evaluate the mRNA expression of the cell adhesion molecules (CAMs) that are constituent factors of gap junctions [connexin (Cx) 43] and adherence junctions (VE-, E-, N-cadherin) in two types of endothelial cells from the mid CL and in CL tissue during the estrous cycle and PGF2α-induced luteolysis in the cow. Specific mRNA expression for Cx43 and N-cadherin was detected in cytokeratin-positive (CK+) and cytokeratin-negative (CK-) luteal endothelial cells (EC) and fully luteinized granulosa cells (LGC). E-cadherin mRNA was expressed in... |
