Acute hepatotoxicity of Crotalus durissus terrificus (South American rattlesnake) venom in rats J. Venom. Anim. Toxins incl. Trop. Dis.
França,RF; Vieira,RP; Ferrari,EF; Souza,RA; Osorio,RAL; Prianti-Jr,ACG; Hyslop,S; Zamuner,SR; Cogo,JC; Ribeiro,W.
Venom of the South American rattlesnake, Crotalus durissus terrificus (Cdt), presents myotoxic and neurotoxic outcomes, but reports on its effects on the liver are scarce. This study examined the hepatotoxicity resulting from Cdt venom administration (100, 200 and 300 µg/kg) in male Wistar rats. Animals were studies at 3, 6, 9 and 12 hours after venom injection. The hepatotoxicity was assessed through serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (AP), gamma glutamyl transferase (GGT), bilirrubin and also by histopathological evaluation. All the different concentrations of Cdt venom resulted in increased levels of hepatic enzymes, when compared with the control group, except for the 100 µg/kg dose,...

Age-related biological activity of South American rattlesnake (Crotalus durissus terrificus) venom J. Venom. Anim. Toxins incl. Trop. Dis.
Furtado,M. F. D.; Santos,M. C.; Kamiguti,A. S..
An in vitro and in vivo comparative study was performed on the effects of Crotalus durissus terrificus venoms from a mother and its 15 newborns. The venoms were tested for protein content, lethality, proteolytic, myotoxic, hemorrhagic, and phospholipase A2 activity. The minimum coagulant dose in plasma and human fibrinogen, protrhombin, and Factor II activations were analyzed. The venoms were also analyzed by polyacrylamide gel electrophoresis (PAGE). This showed that despite similar total protein content, the biological effects of the venoms were different. Venom from young snakes exhibited higher enzymatic and coagulant activities and higher myotoxicity compared to the mother’s. In addition, the PLA2 content paralleled myotoxicity. However, no difference...

ANTITUMOR EFFECT OF SNAKE VENOMS J. Venom. Anim. Toxins
SILVA,R. J. DA; FECCHIO,D.; BARRAVIERA,B..
The search for biological antitumor agents has been pursued for over half a century. Snake venom has been shown to possess a wide spectrum of biological activities. The objectives of the present review are to evaluate the existing controversies on this subject published in a number of papers and to propose probable explanations for the phenomena observed. We reported our results obtained in a study, in which we evaluated the action of the venoms of Crotalus durissus terrificus and Bothrops jararaca on Ehrlich ascites tumor cells. We noticed an important antitumor effect, mainly with Bothrops jararaca venom, as well as an increase in the functional activity of macrophages. We also observed an increase in the number of mononuclear and polymorphonuclear cells...

Biochemical and biological evaluation of gyroxin isolated from Crotalus durissus terrificus venom J. Venom. Anim. Toxins incl. Trop. Dis.
Barros,LC; Soares,AM; Costa,FL; Rodrigues,VM; Fuly,AL; Giglio,JR; Gallacci,M; Thomazini-Santos,IA; Barraviera,SRCS; Barraviera,B; Ferreira Junior,RS.
Gyroxin, a thrombin-like enzyme isolated from Crotalus durissus terrificus venom and capable of converting fibrinogen into fibrin, presents coagulant and neurotoxic activities. The aim of the present study was to evaluate such coagulant and toxic properties. Gyroxin was isolated using only two chromatographic steps - namely gel filtration (Sephadex G-75) and affinity (Benzamidine Sepharose 6B) - resulting in a sample of high purity, as evaluated by RP-HPLC C2/C18 and electrophoretic analysis that showed a molecular mass of 30 kDa. Gyroxin hydrolyzed specific chromogenic substrates, which caused it to be classified as a serine proteinase and thrombin-like enzyme. It was stable from pH 5.5 to 8.5 and inhibited by Mn²+, Cu²+, PMSF and benzamidine. Human...

Crotalus durissus terrificus crotapotin naturally displays preferred positions for amino acid substitutions J. Venom. Anim. Toxins incl. Trop. Dis.
Oliveira,Laudicéia Alves de; Ferreira Jr,Rui Seabra; Barraviera,Benedito; Carvalho,Francilene Capel Tavares de; Barros,Luciana Curtolo de; Santos,Lucilene Delazari dos; Pimenta,Daniel Carvalho.
Abstract Background Classically, Crotalus durissus terrificus (Cdt) venom can be described, according to chromatographic criteria, as a simple venom, composed of four major toxins, namely: gyroxin, crotamine, crotoxin and convulxin. Crotoxin is a non-covalent heterodimeric neurotoxin constituted of two subunits: an active phospholipase A2 and a chaperone protein, termed crotapotin. This molecule is composed of three peptide chains connected by seven disulfide bridges. Naturally occurring variants/isoforms of either crotoxin or crotapotin itself have already been reported. Methods The crude Cdt venom was separated by using RP-HPLC and the toxins were identified by mass spectrometry (MS). Crotapotin was purified, reduced and alkylated in order to...

Crotoxin, the major toxin from the rattlesnake Crotalus durissus terrificus, inhibits ³H-choline uptake in guinea pig ileum BJMBR
Kattah,L.S.; Santoro,M.M.; Diniz,C.R.; De Lima,M.E..
We examined the effect of crotoxin, the neurotoxic complex from the venom of the South American rattlesnake Crotalus durissus terrificus, on the uptake of ³H-choline in minces of smooth muscle myenteric plexus from guinea pig ileum. In the concentration range used (0.03-1 µM) and up to 10 min of treatment, crotoxin decreased ³H-choline uptake by 50-75% compared to control. This inhibition was time dependent and did not seem to be associated with the disruption of the neuronal membrane, because at least for the first 20 min of tissue exposure to the toxin (up to 1 µM) the levels of lactate dehydrogenase (LDH) released into the supernatant were similar to those of controls. Higher concentrations of crotoxin or more extensive incubation times with this toxin...

Does the rattle of Crotalus durissus terrificus reveal its dietary history? J. Venom. Anim. Toxins incl. Trop. Dis.
Martinez,Melissa Gaste; Ducatti,Carlos; Silva,Evandro Tadeu; Sant’Anna,Savio Stefanini; Sartori,Maria Márcia Pereira; Barraviera,Benedito.
Background Environmental devastation threatens the survival of many species, including venomous snakes such as the South American rattlesnake Crotalus durissus terrificus. This observation is based on the decrease of snakes collected and donated to Brazilian research institutes. Nevertheless, some individuals have managed to survive and procreate. The question is how these snakes are adapting in these new environmental conditions. Methods To answer it, the carbon-13 level of rattlesnakes and their feed (either laboratory or wild mice) was evaluated by isotope-ratio mass spectrometry. Thus, rattle segments from 16 adults and 15 offspring of captive snakes, and of three wild newborn C. d. terrificus were evaluated as well as 17 Mus musculus mice captured in...
