A biodegradation study of forest biomass by Aspergillus niger F7: correlation between enzymatic activity, hydrolytic percentage and biodegradation index BJM
Sharma,Nivedita; Kaushal,Richa; Gupta,Rakesh; Kumar,Sanjeev.
Aspergillus niger F7 isolated from soil was found to be the potent producer of cellulase and xylanase. The residue of forest species Toona ciliata, Celtris australis, Cedrus deodara and Pinus roxburghii was selected as substrate for biodegradation study due to its easy availability and wide use in industry. It was subjected to alkali (sodium hydroxide) treatment for enhancing its degradation. Biodegradation of forest waste by hydrolytic enzymes (cellulase and xylanase) secreted by A. niger under solid state fermentation (SSF) was explored. SSF of pretreated forest biomass was found to be superior over untreated forest biomass. Highest extracellular enzyme activity of 2201±23.91 U/g by A. niger was shown in pretreated C. australis wood resulting in...

Dorneles,Athos Odin Severo; Pereira,Aline Soares; Sasso,Victória Martini; Possebom,Gessieli; Tarouco,Camila Peligrinotti; Schorr,Márcio Renan Weber; Rossato,Liana; Ferreira,Paulo Ademar Avela; Tabaldi,Luciane Almeri.
ABSTRACT Potato cultivation is widespread around the world, being exposed to several abiotic stresses, including soils with high aluminum (Al) availability. Silicon (Si) is recognized for alleviating the stress caused by Al in various plant species. Thus, the aim of this study was to investigate the potential of Si to mitigate the oxidative stress caused by Al in potato genotypes, exhibiting differential sensitivity toward this element. Plants of the Al-sensitive genotype (SMIJ319-7) and Al-tolerant genotype (SMIF212-3) were grown for two weeks in a hydroponic system with the nutrient solution containing combinations of Al (0 and 1.85 mM) and Si (0, 0.5 and 1.0 mM). At the end of the experiment, photosynthetic parameters, pigment content, root and shoot...

Analysis of methodologies for the study of composition and biochemical carbohydrate changes in harvest and postharvest onion bulbs Phyton
Abrameto1,MA; Pozzo Ardizzi,CM; Gil,MI; Molina,LM.
Costly investments in storage and transport of onion bulbs makes it increasingly important to identify cultivars with the best chance of long-term storage. This paper discusses our own and other researchers' results and laboratory procedures on onion's pungency, soluble solids, dry matter, respiration rate, carbohydrates and catabolism-related enzymes under different storage conditions. The variability in parameters such as carbohydrate composition, pungency, soluble solids and soluble uronic acid concentration in water is growth dependent, and defines the quality of onion bulbs at harvest time. During storage, these parameters can be modified by environmental conditions, presence of pathogens or termination of dormancy. Under postharvest conditions,...

Biochemical and physiological analysis in carrot seeds from different orders of umbels Rev. Ciênc. Agron.
Silva,Patrícia Pereira da; Sekita,Marcelo Coelho; Dias,Denise Cunha Fernandes dos Santos; Nascimento,Warley Marcos.
ABSTRACT In carrot seed production, harvesting of the umbels is generally staggered, or sometimes the whole plant is harvested with the umbels at different stages of maturation; in that case, there is a risk of harvesting immature seeds and of losses due to natural dehiscent, or to physiological deterioration as a result of unfavourable environmental factors. The present study aimed to evaluate the physiological and biochemical changes related to the maturation of 'Planalto' carrot seeds from diferent umbels. The samples were collected 120 days after anthesis, with the primary, secondary and tertiary umbels being harvested separately. Seeds were analysed for germination, first count and germination rate, at temperatures of 25 ºC (optimum) to 35 ºC...

Yap,MKK; Tan,NH; Fung,SY.
The lethal and enzymatic activities of venom from Naja sumatrana (Equatorial spitting cobra) were determined and compared to venoms from three other Southeast Asian cobras (Naja sputatrix, Naja siamensis and Naja kaouthia). All four venoms exhibited the common characteristic enzymatic activities of Asiatic cobra venoms: low protease, phosphodiesterase, alkaline phosphomonoesterase and L-amino acid oxidase activities, moderately high acetylcholinesterase and hyaluronidase activities and high phospholipase A2. Fractionation of N. sumatrana venom by Resource® S cation exchange chromatography (GE Healthcare, USA) yielded nine major protein peaks, with all except the acidic protein peak being lethal to mice. Most of the protein peaks exhibit enzymatic...

Characterization of polyphenol oxidase in two cocoa (Theobroma cacao L.) cultivars produced in the south of Bahia, Brazil Ciênc. Tecnol. Aliment.
MACEDO,Adrielle Souza Leão; ROCHA,Fátima de Souza; RIBEIRO,Margareth da Silva; SOARES,Sergio Eduardo; BISPO,Eliete da Silva.
Abstract The reactions leading to the formation of precursors of chocolate flavor are performed by endogenous enzymes present in the cocoa seed. Polyphenol oxidase (PPO) presence and activity during fermentation of cocoa beans is responsible for the development of flavor precursors and is also implicated in the reduction of bitterness and astringency. However, the reliability of cocoa enzyme activities is complicated due to variations in different genotypes, geographical origins and methods of fermentation. In addition, there is still a lack of systematic studies comparing different cocoa cultivars. So, the present study was designed to characterize the activity of PPO in the pulp and seeds of two cocoa cultivars, PH 16 and TSH 1188. The PPO activity was...

Differential survivorship of congeneric ornamental fishes under forecasted climate changes are related to anaerobic potential Genet. Mol. Biol.
Gonçalves,Luciana Mara Fé; Silva,Maria de Nazaré Paula da; Val,Adalberto Luis; Almeida-Val,Vera Maria Fonseca de.
Abstract Two Amazonian closely related tetras – cardinal Paracheirodon axelrodi and green neon P. simulans – were artificially acclimatized to environmental chambers mimicking future climate change scenarios (mild, moderate and extreme), using a microcosm facility. P. simulans survived (100%) to all scenarios after 30 days exposure, while P. axelrodi presented decreasing survival percentages according to environmental severity. These differences may be the reflection of distinct natural acclimatization to microhabitats between the species, which differ in thermal conditions. Survival responses might be related to differences in relative gene expression of lactate dehydrogenase (Ldh), suggesting that P. axelrodi anaerobic potential is lower or non-existent...

Diurnal changes in storage carbohydrate metabolism in cotyledons of the tropical tree Hymenaea courbaril L. (Leguminosae) Rev. Bras. Bot.
Amaral,Lourdes Isabel Velho do; Santos,Henrique P.; Rossatto,Davi Rodrigo; Buckeridge,Marcos Silveira.
The cotyledons of Hymenaea courbaril store large amounts of xyloglucan, a cell wall polysaccharide that is believed to serve as storage for the period of seedling establishment. During storage mobilisation, xyloglucan seems to be degraded by a continuous process that starts right after radicle protrusion and follows up to the establishment of photosynthesis. Here we show evidence that events related to the hydrolases activities and production (α-xylosidase, β-galactosidase, β-glucosidase and xyloglucan endo-β-transglucosilase) as well as auxin, showed changes that follow the diurnal cycle. The period of higher hydrolases activities was between 6pm and 6am, which is out of phase with photosynthesis. Among the enzymes, α-xilosidase seems to be more important...

Effect of growth hormone on fatty acid synthase gene expression in porcine adipose tissue cultures Genet. Mol. Biol.
José,Andrea A.F.B.V.; Alvares,Lúcia. E.; Delgado,Eduardo F.; Coutinho,Luiz L.; Lanna,Dante P.D..
We describe an efficient in vitro assay to test growth hormone effects on mRNA levels and fatty acid synthase (FAS, EC. 2.3.1.85) activity. Swine adipose tissue explants were long-term cultured in medium containing growth hormone and FAS mRNA levels and enzyme activity were measured. We quantified FAS transcripts by competitive reverse transcriptase PCR (RT-PCR) using total RNA from cultured adipose tissue explants and RT-PCR standard-curves were constructed using a cloned 307 bp segment of native FAS cDNA and a shorter fragment from which a 64 bp (competitor, 243 bp) internal sequence had been deleted. A known amount of competitor was added to each PCR as an internal control and µ-actin transcripts were also measured to correct for differences in total...

Endophytic fungi producing of esterases: evaluation in vitro of the enzymatic activity using pH indicator BJM
Lisboa,Helen Cristina Fávero; Biasetto,Carolina Rabal; Medeiros,João Batista de; Araújo,Ângela Regina; Silva,Dulce Helena Siqueira; Teles,Helder Lopes; Trevisan,Henrique Celso.
A sensitive and efficient colorimetric method was optimized for detection of esterase enzymes produced by endophytic fungi for development of High-Throughput Screening (HTS). The fungi were isolated and obtained previously from plant species of Cerrado and Atlantic Forest located in areas of environmental preservation in the State of Sao Paulo / Brazil, as part of the project "Chemical and biological prospecting endophytic fungi associated to plant species of Cerrado and Atlantic Forest". The compounds ethyl butyrate, ethyl acetate and methyl propionate were used as standards esters which were hydrolyzed by extracellular enzyme from endophytic fungi (EC. 3.1.1.1 -carboxylesterases) for production of carboxylic acids. Thus, the reduction of the pH increases...

Ethylene and lettuce seed germination Scientia Agricola
Nascimento,Warley Marcos.
Ethylene can stimulate seed germination and overcome dormancy in many species. For instance, the inhibitory effect of high temperature on lettuce seed germination can be overcome by exogenous ethylene. Involvement of ethylene in seed germination is a widely accepted fact, but the mechanistic details are poorly understood. A critical factor in ethylene studies is the response of seed germination properties to various ethylene inhibitors. Lettuce seed has been used as a model to study the role of ethylene in seed germination at high temperature. The role of ethylene in germination is not known but either ethylene evolution or addition of 1-aminocyclopropane-1-carboxilic acid (ACC) have been associated with increased activity of endo-beta-mannanase, a...
