ABSTRACT Potato cultivation is widespread around the world, being exposed to several abiotic stresses, including soils with high aluminum (Al) availability. Silicon (Si) is recognized for alleviating the stress caused by Al in various plant species. Thus, the aim of this study was to investigate the potential of Si to mitigate the oxidative stress caused by Al in potato genotypes, exhibiting differential sensitivity toward this element. Plants of the Al-sensitive genotype (SMIJ319-7) and Al-tolerant genotype (SMIF212-3) were grown for two weeks in a hydroponic system with the nutrient solution containing combinations of Al (0 and 1.85 mM) and Si (0, 0.5 and 1.0 mM). At the end of the experiment, photosynthetic parameters, pigment content, root and shoot... |

Costly investments in storage and transport of onion bulbs makes it increasingly important to identify cultivars with the best chance of long-term storage. This paper discusses our own and other researchers' results and laboratory procedures on onion's pungency, soluble solids, dry matter, respiration rate, carbohydrates and catabolism-related enzymes under different storage conditions. The variability in parameters such as carbohydrate composition, pungency, soluble solids and soluble uronic acid concentration in water is growth dependent, and defines the quality of onion bulbs at harvest time. During storage, these parameters can be modified by environmental conditions, presence of pathogens or termination of dormancy. Under postharvest conditions,... |

ABSTRACT In carrot seed production, harvesting of the umbels is generally staggered, or sometimes the whole plant is harvested with the umbels at different stages of maturation; in that case, there is a risk of harvesting immature seeds and of losses due to natural dehiscent, or to physiological deterioration as a result of unfavourable environmental factors. The present study aimed to evaluate the physiological and biochemical changes related to the maturation of 'Planalto' carrot seeds from diferent umbels. The samples were collected 120 days after anthesis, with the primary, secondary and tertiary umbels being harvested separately. Seeds were analysed for germination, first count and germination rate, at temperatures of 25 ºC (optimum) to 35 ºC... |

The lethal and enzymatic activities of venom from Naja sumatrana (Equatorial spitting cobra) were determined and compared to venoms from three other Southeast Asian cobras (Naja sputatrix, Naja siamensis and Naja kaouthia). All four venoms exhibited the common characteristic enzymatic activities of Asiatic cobra venoms: low protease, phosphodiesterase, alkaline phosphomonoesterase and L-amino acid oxidase activities, moderately high acetylcholinesterase and hyaluronidase activities and high phospholipase A2. Fractionation of N. sumatrana venom by Resource® S cation exchange chromatography (GE Healthcare, USA) yielded nine major protein peaks, with all except the acidic protein peak being lethal to mice. Most of the protein peaks exhibit enzymatic... |

ABSTRACT The lipases have ability to catalyze diverse reactions and are important in different biotechnological applications. The aim of this work was to isolate and characterize microorganisms that produce lipases, from different food industry effluents localized in Pelotas, RS/Brazil. Bacteria were identified using Gram stain and biochemical tests (Vitek 2(r)). Fungi were identified according to macro and micromorphology characteristics. The extracellular lipase production was evaluated using the Rhodamine B test and the enzymatic activity by titration. Twenty-one bacteria were isolated and identified as Klebsiella pneumoniae ssp. pneumoniae, Serratia marcescens, Enterobacter aerogenes, Raoultella ornithinolytica and Raoultella planticola. Were... |

BackgroundCrotalus durissus terrificus venom (CdtV) is one of the most studied snake venoms in Brazil. Despite presenting several well known proteins, its L-amino acid oxidase (LAAO) has not been studied previously. This study aimed to isolate, characterize and evaluate the enzyme stability of bordonein-L, an LAAO from CdtV.Methods The enzyme was isolated through cation exchange, gel filtration and affinity chromatography, followed by a reversed-phase fast protein liquid chromatography to confirm its purity. Subsequently, its N-terminal amino acid sequence was determined by Edman degradation. The enzyme activity and stability were evaluated by a microplate colorimetric assay and the molecular mass was estimated by SDS-PAGE using periodic acid-Schiff... |

The causal agents of apple bull?s-eye rot in southern Brazil have recently been described as Neofabraea actinidiae and N. brasiliensis. Isolates of both species were evaluated for response of mycelial growth index (MGI) to different temperatures, enzyme production, mycelial growth inhibition and effective concentrations (EC 50 and EC 100 ) of the fungicides tri fl umizole, pyrimethanil and thiophanate methyl, as well as aggres- siveness on fruits of "Fuji " hybrid and " Pink Lady " . There was signi ficantly lower mycelium growth in N. brasiliensis compared with N. actinidiae at all temperatures tested. Neither species grew at 3 and 32°C. There were minor differences in production of enzymes in the two species, with all N. brasiliensis isolates showing no... |

Abstract The reactions leading to the formation of precursors of chocolate flavor are performed by endogenous enzymes present in the cocoa seed. Polyphenol oxidase (PPO) presence and activity during fermentation of cocoa beans is responsible for the development of flavor precursors and is also implicated in the reduction of bitterness and astringency. However, the reliability of cocoa enzyme activities is complicated due to variations in different genotypes, geographical origins and methods of fermentation. In addition, there is still a lack of systematic studies comparing different cocoa cultivars. So, the present study was designed to characterize the activity of PPO in the pulp and seeds of two cocoa cultivars, PH 16 and TSH 1188. The PPO activity was... |

ABSTRACT: An experiment was conducted to study the effects of two sources and dietary levels of selenium (Se) on performance, Se concentration and glutathione peroxidase activity in plasma and liver of piglets post weaning. A total of 126 weaned pigs male and female Large White × Landrace, at average body weight of 7.2±0.67kg were allotted to seven treatments in a randomized block design, with two Se sources (organic and inorganic), inclusion levels, two levels for the inorganic source (0.30 and 0.60ppm) and four for the organic source (0.15, 0.30, 0.45 and 0.60ppm) and one control diet without Se supplementation containing 0.095ppm, and six replicates of three animals per pen. The experimental period was 35 days post-weaning. The organic source contained... |
