Acid and enzymatic hydrolysis to recover reducing sugars from cassava bagasse: an economic study BABT

The objective of this work was to study the acid and enzymatic hydrolysis of cassava bagasse for the recovery of reducing sugars and to establish the operational costs. A statistical program "Statistica", based on the surface response was used to optimize the recovery of reducing sugars in both the processes. The process economics was determined considering the values of reducing sugars obtained at laboratory scale, and the operations costs of a cylindrical reactor of 1500 L, with flat walls at the top and bottom. The reactor was operated with 150 kg of cassava bagasse and 1350 kg of water. The yield of the acid hydrolysis was 62.4 g of reducing sugars from 100 g of cassava bagasse containing 66% starch. Activity of fungal phytases stored in two ways in response to the period of storage at room temperature Ciência Rural

Two distinct experiments were conducted simultaneously with phytases of Aspergillus oryzae and A. niger for determining enzyme activity in response to storage period (up to 180 days at room temperature - RT). In the first experiment, enzymes were stored as they were acquired (pure form) and the activity was measured periodically during 180 days of storage at RT. In the second experiment, the phytases were incorporated to a supplement containing vitamins, minerals, and amino acids; and then this supplement was stored at RT up to 180 days, so that every 30 days of storage was collected one aliquot from each replicate for determining enzymatic activity. Summer mortality of the oyster result from the interaction of many factors. A respiratory assumption proposes a link between the supposed causes of the energy state, the cultivation methods and the medium. This work is focused towards the search for specific metabolic markers of environmental stresses. The cDNA of genes coding for PK and PEPCK were cloned and sequenced in order to detect their regulation level. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The objective of this work was to assess the functionality of the glycolytic pathways in the bacterium Xylella fastidiosa. To this effect, the enzymes phosphoglucose isomerase, aldolase, glyceraldehyde-3-phosphate dehydrogenase and pyruvate kinase of the glycolytic pathway, and glucose 6-phosphate dehydrogenase of the Entner-Doudoroff pathway were studied, followed by cloning and expression studies of the enolase gene and determination of its activity. These studies showed that X. fastidiosa does not use the glycolytic pathway to metabolize carbohydrates, which explains the increased duplication time of this phytopatogen. Carbon sources effect on pectinase production from Aspergillus japonicus 586 BJM

The effect of different carbon sources on the pectinesterases, endo- and exo-polygalacturonase activities from Aspergillus japonicus 586 was evaluated in liquid media (Manachini solutions) supplemented with different substrate concentrations. The culture medium was inoculated with 5.10(6) spores/ml and mantained under agitation (140 rpm), at 30°C, during 122 h. The enzyme evaluation was carried out 24 h after filtration. Chicken serologic response to Salmonella enterica serotype Typhimurium assessed by Elisa Rev. Bras. Ciênc. Avic.

This study evaluated two enzyme-linked immunosorbent assays (ELISA) in the detection of chicken serologic response against Salmonella enterica sorotype Typhimurium. The assays have used as detecting antigen the soluble bacterial proteins of a non-flagellated strain of Salmonella Typhimurium (AgTM), and antibody conjugated to peroxidase or alkaline phosphatase. According to the results, optimal dilutions of antigen (concentration 5.49 mg/mL) and serum samples in both assays were 1:20,000 and 1:1,000, respectively. In such conditions, the ELISA/AgTM was able to detect serological response to Salmonella Typhimurium. Cross-reactions to Salmonella serotypes Gallinarum and Pullorum were seen, but not with other serotypes such as Enteritidis.

Chromatographic characterization of products isolated from chrome shavings BABT

Gel permeation chromatography and high performance liquid chromatography were employed for separation and chemical characterization of products isolated from chrome shavings. After enzymatic hydrolysis, the products isolated were peptides of higher molecular weight. Peptides of lower molecular weight and free aminoacids were the main products using sulfuric acid in chrome shavings solubilization. Glycine (17%) , glutamic acid (10.6%) , alanine (9.2%) , and arginine (8.2%) were the principal amino acids found. Phenylalanine(1.8%) was the main aromatic amino acid , while tryptophane was completely absent.

Comparison of growth, digestive system maturation and skeletal development in sea bass larvae reared in an intensive or a mesocosm system ArchiMer

The quality of development in intensive or mesocosm hatchery-reared Dicentrarchus labrax larvae was investigated using physiological indicators assessing ontogeny. Larvae were reared in intensive (120 L tanks) and in mesocosm systems (20 m(3) enclosures) with the same feeding sequence, excluding the wild zooplankton naturally available in mesocosms. Faster growth was recorded since early development [16 day after hatching (DAH)] in the mesocosm. Co-production of Proteases and Bioactive Protein Hydrolysates from Bioprocessing of Feather Meal BABT

Abstract: Feather meal conversion through submerged cultivations with Bacillus strains (CL33A, CL14) yielded proteases and protein hydrolysates. After 4-day (CL33A) and 10-day (CL14) cultivations, protease activities reached 461 U/mL; hydrolysates presented antioxidant (radicals-scavenging, 57-77%; Fe2+-chelation, 14-28%; Fe3+-reduction) and antidiabetic (dipeptidyl peptidase-IV inhibition, 49-52%) potentials. The obtained bioproducts present prospective commercial/industrial applications.

Fish sauces are traditionally used in most Asian countries. Whatever the process, the principle is the same i.e. a fish autolysis in saline media followed by a fermentation step. Not less than 18 months are needed to obtain a brown liquid rich in salt and amino acids. This PhD work aims to determine if an addition of proteases can speed up the process while keeping the nutritional and sensorial properties. The experiment was conducted to evaluate the addition of phytase on productive performance and egg quality of Japanese quails fed diets containing different levels of phytase. It was used 320 Japanese quails at 167 days of age, weighing 182.3 ± 3.8 g distributed in a completely randomized design with four experimental diets and eight repetitions of ten birds per experimental unit.
