Association between reproductive traits and four microsatellites in Brangus-Ibagé cattle Genet. Mol. Biol.
Oliveira,João F.C. de; Neves,Jairo P.; Almeida,Eric A.; Steigleder,Clara Sabina; Moraes,José Carlos F.; Gonçalves,Paulo Bayard D.; Weimer,Tania A..
The aim of the present study was to verify associations between reproductive efficiency and four microsatellite markers located in synteny with genes involved in the regulation of reproductive mechanisms. A sample of 107 females from a Brangus Ibagé population (5/8 Aberdeen Angus x 3/8 Nelore) was characterized for ETH225 (D9S1) and MM12E6 (D9S20) microsatellites, mapped on chromosome 9, and HEL5 (D21S15) and AFZ1 (D21S37) on chromosome 21. Associations between the genetic markers and reproductive efficiency were determined by one-way analysis of variance using calving interval (CI), live weight at calving (LWC), live weight at first calving (LW1C) and live weight at second calving (LW2C) as dependent variables. The genotypes were classified according to...

Association of the estrogen receptor gene Pvu II restriction polymorphism with expected progeny differences for reproductive and performance traits in swine herds in Brazil Genet. Mol. Biol.
Santana,Bárbara Amélia Aparecida; Biase,Fernando H.; Antunes,Robson Carlos; Borges,Mauricio; Franco,Mauricio Machaim; Goulart,Luiz Ricardo.
Estrogen has an important function in swine reproduction and growth. A Pvu II restriction enzyme polymorphism has been proven to be an important genetic variation in the estrogen receptor gene (ESR) and may be considered as a candidate gene for use in pig production but there is no data regarding the prevalence of this polymorphism in the Brazilian pig population. We used DNA samples from the following three purebred pig breeds: Large White (336 females and 26 males), Landrace (304 females and 27 males) and Pietrain (125 females and 11 males). The ESR genotyping was performed using PCR-RFLP. For each breed, genotypes for the ESR gene were compared independently for expected progeny differences (EPD) in litter size (LS), average daily weight gain (DWG)...

Estrogen and progesterone receptors in human papilloma virus-related cervical neoplasia BJMBR
Coelho,F.R.G.; Prado,J.C.M.; Pereira Sobrinho,J.S.; Hamada,G.; Landman,G.; Pinto,C.A.; Nonogaki,S.; Villa,L.L..
Estrogen (ER) and progesterone (PR) receptors in the normal uterine cervix, cervical intraepithelial neoplasia and invasive carcinoma were studied in consecutive samples from Hospital do Câncer, São Paulo, between 1996 and 1997. Tissue was collected by removing a fragment of the tumoral area using a 5-mm diameter biopsy punch, followed by removal of a macroscopically normal area as close as possible from the tumor. Histopathological confirmation was obtained for all specimens analyzed. A total of 24 normal tissues, 17 cases of cervical intraepithelial neoplasia and 7 of invasive carcinomas were studied. The ER/PR ratio was determined by immunohistochemistry using monoclonal antibodies specific for each receptor. Adjacent tissue slides were submitted to...

Immunohistochemical Localization of Estrogen Receptor in the Embryonic Gonad of Male Quail Embryo During Gonadal Differentiation International Journal of Morphology
Intarapat,Sittipon; Sailasuta,Achariya; Satayalai,Orawan.
In birds, male embryo the gonads develop bilateral testes, in which both left and right sides produce functional spermatozoa, whereas female embryo, only the left gonad develops into a functional ovary. Estrogen plays a key role in avian sex determination in both sexes by binding to the estrogen receptor (ER). Surprisingly, chicken estrogen receptor (cER) mRNA is expressed in both sexes; moreover; its expression is only expressed in the left male gonad. The present study aimed to localize ER protein in the left gonad of male quail embryo using immunohistochemistry. The 8-day-old male quail embryos whose embryonic sex distinguished by gonadal morphology were studied. Histology of the left male gonad displayed thin cortex containing 1 to 2 layers of the...

Interaction between estrogen receptor and retinol-binding protein-4 polymorphisms as a tool for the selection of prolific pigs Genet. Mol. Biol.
Gonçalves,Iara Denise Vasconcellos; Gonçalves,Paulo Bayard Dias; Silva,Janandra Cortese da; Portela Jr,Valério Valdetar; Borges,Luiz Felipe Kruel; Oliveira,João Francisco Coelho; Lovatto,Paulo Alberto.
The aim of the present study was to investigate the association of the estrogen receptor (ER-PvuII) and retinol-binding protein 4 (RBP4-MspI) gene polymorphisms and their interactions with prolificacy in a commercial synthetic pig line reared in Brazil. A total of 10,374 piglet records from 218 sows and 817 litters were used for litter size analysis. Only females with three or four farrowings were included in the analysis. The mean litter size ranged from 5.0 to 19.5 piglets. DNA was extracted from leukocytes by a standard method, and ER-PvuII and RBP4-MspI polymorphisms were characterized by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique. The association between alleles or genotypes and reproductive performance...

Profile of thyroid hormones in breast cancer patients BJMBR
Saraiva,P.P.; Figueiredo,N.B.; Padovani,C.R.; Brentani,M.M.; Nogueira,C.R..
Estrogen involvement in breast cancer has been established; however, the association between breast cancer and thyroid diseases is controversial. Estrogen-like effects of thyroid hormone on breast cancer cell growth in culture have been reported. The objective of the present study was to determine the profile of thyroid hormones in breast cancer patients. Serum aliquots from 26 patients with breast cancer ranging in age from 30 to 85 years and age-matched normal controls (N = 22) were analyzed for free triiodothyronine (T3F), free thyroxine (T4F), thyroid-stimulating hormone (TSH), antiperoxidase antibody (TPO), and estradiol (E2). Estrogen receptor ß (ERß) was determined in tumor tissues by immunohistochemistry. Thyroid disease incidence was higher in...

Transcriptional regulation of bidirectional gene pairs by 17-β-estradiol in MCF-7 breast cancer cells BJMBR
Garcia,S.A.B.; Nagai,M.A..
Using cDNA microarray analysis, we previously identified a set of differentially expressed genes in primary breast tumors based on the status of estrogen and progesterone receptors. In the present study, we performed an integrated computer-assisted and manual search of potential estrogen response element (ERE) binding sites in the promoter region of these genes to characterize their potential to be regulated by estrogen receptors (ER). Publicly available databases were used to annotate the position of these genes in the genome and to extract a 5’flanking region 2 kb upstream to 2 kb downstream of the transcription start site for transcription binding site analysis. The search for EREs and other binding sites was performed using several publicly available...

Transcriptional up-regulation of PHLDA1 by 17β-estradiol in MCF-7 breast cancer cells BJMBR
Marchiori,A.C.; Casolari,D.A.; Nagai,M.A..
Most breast cancer risk factors are associated with prolonged exposure of the mammary gland to high levels of estrogens. The actions of estrogens are predominantly mediated by two receptors, ERα and ERβ, which act as transcription factors binding with high affinity to estrogen response elements in the promoter region of target genes. However, most target genes do not contain the consensus estrogen response elements, but rather degenerated palindromic sequences showing one or more mutations and other ER-binding sites such as AP-1 and SP-1. Using the differential display reverse transcription-polymerase chain reaction technique, our group identified several genes differentially expressed in normal tissue and in ER-positive and ER-negative primary breast...
