Bacterial strains from floodplain soils perform different plant-growth promoting processes and enhance cowpea growth Scientia Agricola
Costa,Elaine Martins da; Carvalho,Fernanda de; Nóbrega,Rafaela Simão Abrahão; Silva,Jacqueline Savana; Moreira,Fatima Maria de Souza.
ABSTRACT Certain nodulating nitrogen-fixing bacteria in legumes and other nodule endophytes perform different plant-growth promoting processes. The objective of this study was to evaluate 26 bacterial strains isolated from cowpea nodules grown in floodplain soils in the Brazilian savannas, regarding performance of plant-growth promoting processes and ability to enhance cowpea growth. We also identified these strains by 16S rRNA sequencing. The following processes were evaluated: free-living biological nitrogen fixation (BNF), solubilization of calcium, aluminum and iron phosphates and production of indole-3-acetic acid (IAA). The abilities to nodulate and promote cowpea growth were evaluated in Leonard jars. Partial sequencing of the 16S rRNA gene...

Growth regulators and darkness increase efficiency in in vitro culture of immature embryos from peppers Scientia Agricola
Manzur,Juan Pablo; Calvache-Asensio,María de las Nieves; Rodriguez-Burruezo,Adrian.
Common pepper (Capsicum annuum L.) is one of the most important vegetables in the world, and extensive breeding efforts are being made to develop new improved strains of this species. In this regard, in vitro culture of immature embryos may help breeders accelerate breeding cycles and overcome interspecific barriers, among other applications. In this study, we have optimized a protocol for in vitro culture of immature embryos of C. annuum. Levels of indole-3-acetic acid (IAA) and zeatin have been tested to improve the efficiency (germination rates) of this technique in C. annuum embryos at the four main immature stages (i.e. globular, heart, torpedo, and early cotyledonary) from four varietal types of this species (California Wonder, Piquillo, Guindilla,...

Indole ring oxidation by activated leukocytes prevents the production of hypochlorous acid BJMBR
Ximenes,V.F.; Paino,I.M.M.; Faria-Oliveira,O.M.M. de; Fonseca,L.M. da; Brunetti,I.L..
Hypochlorous acid (HOCl) released by activated leukocytes has been implicated in the tissue damage that characterizes chronic inflammatory diseases. In this investigation, 14 indole derivatives, including metabolites such as melatonin, tryptophan and indole-3-acetic acid, were screened for their ability to inhibit the generation of this endogenous oxidant by stimulated leukocytes. The release of HOCl was measured by the production of taurine-chloramine when the leukocytes (2 x 10(6) cells/mL) were incubated at 37ºC in 10 mM phosphate-buffered saline, pH 7.4, for 30 min with 5 mM taurine and stimulated with 100 nM phorbol-12-myristate acetate. Irrespective of the group substituted in the indole ring, all the compounds tested including indole,...

Plant growth regulators optimization for in vitro cultivation of the orchid Guarianthe skinneri (Bateman) Dressier & W.E.Higgins Gayana Botánica
Coello,Christian Yanelly; Miceli,Clara Luz; Orantes,Carolina; DenDooven,Luc; Gutiérrez,Federico Antonio.
An in vitro culture procedure was developed to induce shoots and roots of Guarianthe skinneri (Orchidaceae) plantlets regenerated from seed-derived protocorms on a Murashige and Skoog (MS) médium supplemented with 6-benzyladenine (BA), indole-3-acetic acid (IAA), a-naftalenacetic acid (NAA) and gibberellic acid (GA3). A máximum of 10.6 shoots was obtained with 16.1 μM NAA, 17.1 μM IAA, 6.3 x 10-9 μM GA3 and 0.0023 μM BA. A máximum of 4.0 roots on each shoot with 5.4 μM NAA, 17.1 μM IAA, 0.001 μM AG3 and 4.6 x 10-9 μM BA. Máximum shoot and root length was obtained with a minimum of GA3, but a máximum IAA. GA3, was the main factor controlling shoot and root induction and elongation.
