Interaction of insect actomyosin with adenosine triphosphate National Institute of Agronomic Research

Some physiocochemical observations were made on the interaction between actomyosin from honeybee thoracic muscle and adenosine triphosphate (ATP). ATP caused superprecipitation, viscosity drop, and light-scattering drop in insect actomyosin. On the other hand, actomyosin showed a powerful ATP-splitting enzyme action. The extent of these interactions of insect actomyosin with ATP was fairly coniparable with those of rabbit actomyosin. From the viewpoint of comparative biochemistry, these observations, in general, support the current theory that the ATP-actomyosin system is essential for muscular contraction.

In the present work, an endopin-like elastase inhibitor was purified for the first time from bovine muscle. A three-step chromatography procedure was developed including successively SP-Sepharose, Q-Sepharose and EMD-DEAE 650. This procedure provides about 300 mu g of highly pure inhibitor from 500 g of bovine diaphragm muscle. The N-terminal sequence of the muscle elastase inhibitor, together with the sequence of a trypsin-generated peptide, showed 100% similarity with the cDNA deduced sequence of chromaffin cell endopin 1. Hence, the muscle inhibitor was designated muscle endopin 1 (mEndopin 1). mEndopin 1 had a molecular mass of 70 kDa, as assessed by both gel filtration and SDS/PAGE. Observations préliminaires sur la structure de la strie Z dans le muscle alaire de l'abeille National Institute of Agronomic Research

L'étude au microscope électronique du muscle alaire de l'Abeille Apis mellifica nous a permis d'apporter certaines précisions sur l'ultrastucture de ce muscle. En particulier nous avons pu étudier les connexions existant entre les filaments primaires et la strie Z, en l'absence de bande I. Les muscles ont été fixés au tétroxyde d'osmium, déshydratés à l'alcool éthylique et inclus dans l'araldite selon la technique de Glauert et Glauert (11). Les coupes minces ont été effectuées au microtome Porter Blum et colorées à l'acide phosphotungstique en solution alcoolique durant 5 mn. Les clichés ont été pris au microscope RCA EMU-3C.

Structure, fatigue, and enzyme activities in "fast" insect muscles National Institute of Agronomic Research

Two groups of extensores trochanteris of the mesothoracic leg of Periplaneta americana were investigated: (1) the main depressor of the leg, which is also a flight muscle; (2) the coxal depressor of the leg. These muscle groups are known to be equally ‘fast’ in mechanical and electrical responses. The group used for walking as well as for flying has the greatest endurance. This is correlated with a larger number of sarcosomes, more glycogen, a better oxygensupply, a higher activity of some energy-supplying enzymes, and a structural difference of neuromuscular junctions.

Über den bau und die funktion der valvula vaginalis der bienenkönigin (Apis mellifica L.) National Institute of Agronomic Research

The valve-fold of the queen honeybee is an epithe1ian fold of the ventral vaginal wall which is interlaced with ascending muscle fibres. The valve-fold projects into the lumen of the vagina like a tongue-shaped roll just behind the caudal end of the median oviduct and directly above the sternal antecosta of the VII. segment. Opinions about the function of this fold are dlsagreeing. A short description of the histological structure of the valve-fold is given. Furthermore, reference is made to a premature, progressive degeneration and calcification of the valve-fold's musculature in most of the queen honeybees, dependent on age. In discussions concerning the function of the valve-fold, these causally not clarified degeneration processes must be considered.
