Applications of nucleoside-based molecular probes for the in vivo assessment of tumour biochemistry using positron emission tomography (PET) BABT
Wiebe,Leonard I..
Positron emission tomography (PET) is a non-invasive nuclear imaging technique. In PET, radiolabelled molecules decay by positron emission. The gamma rays resulting from positron annihilation are detected in coincidence and mapped to produce three dimensional images of radiotracer distribution in the body. Molecular imaging with PET refers to the use of positron-emitting biomolecules that are highly specific substrates for target enzymes, transport proteins or receptor proteins. Molecular imaging with PET produces spatial and temporal maps of the target-related processes. Molecular imaging is an important analytical tool in diagnostic medical imaging, therapy monitoring and the development of new drugs. Molecular imaging has its roots in molecular biology....

Approche de l'état physiologique de l'huître creuse Crassostrea gigas au cours d'un cycle saisonnier à Marennes-Oléron ArchiMer
Moal, Jeanne; Samain, Jean-francois; Bodoy, Alain; Le Coz, Jean-rene.
The energy charge was measured for two Crassostrea gigas populations, during an annual survey at Marennes Oleron, to appreciate the condition of health of these oysters. It is assumed that the actual decrease in production of this area is related to a biomass overload which affects the oysters by lowering their scope for growth. Energy charge, an index developped by ATKINSON (1968), defines the energy equilibrium of organisms. It is the adenylic nucleotide ratio: E.C = (ATP + 1/2ADP)/ATP + ADP + AMP). We have measured this parameter to determine the state of health of these oysters. A methodological study has been conducted previously to define the conditions of sampling, conservation and extraction for a HPLC assay. The HPLC method (high performance...

BASiNET - Biological Sequences NETwork: a case study on coding and non-coding RNAs identification. Repositório Alice
ITO, E. A.; KATAHIRA, I.; VICENTE, F. F. da R.; PEREIRA, L. F. P.; LOPES, F. M..
With the emergence of Next Generation Sequencing (NGS) technologies, a large volume of sequence data in particular de novo sequencing was rapidly produced at relatively low costs. In this context, computational tools are increasingly important to assist in the identification of relevant information to understand the functioning of organisms. This work introduces BASiNET, an alignment-free tool for classifying biological sequences based on the feature extraction from complex network measurements. The method initially transform the sequences and represents them as complex networks. Then it extracts topological measures and constructs a feature vector that is used to classify the sequences. The method was evaluated in the classification of coding and...

Effect of nucleotides on broiler performance and carcass yield Rev. Bras. Ciênc. Avic.
Pelícia,VC; Sartori,JR; Zavarize,KC; Pezzato,AC; Stradiotti,AC; Araujo,PC; Mituo,MAO; Madeira,LA.
This study aimed at evaluating the effect of nucleotides on the performance and carcass yield of broilers fed diets with no antibiotic growth promoters (AGP), anticoccidials, or animal feedstuffs. In the trial, 600 Ross 308 male broilers were distributed in a completely randomized experimental design into six treatments with four replicates of 25 birds each. Treatments consisted of a control diet (CD), CD + AGP, CD + 0.04%, CD + 0.05%, CD + 0.06%, and CD + 0.07% nucleotides. The experimental diets did not contain anticoccidials, and birds were vaccinated against coccidiosis at three days of age. No significant differences were detected among broilers submitted to the different treatments in none of the studied parameters. Under the conditions of this...

Improvement in RNA extraction from S. cerevisie by optimization in the autolysis and NH3 hydrolysis BABT
Oliveira,Antonio Martins; Oliva Neto,Pedro de.
The optimization of autolysis of Saccharomyces cerevisiae from brewery was studied aiming at the maximum ribonucleic acid extraction and yeast extract production. The best conditions for yeast autolysis was 55.2ºC, pH= 5.1 and 9.8% NaCl for 24h of processing, without the NH3 use. In these conditions, the RNA yield was 89.7%, resulting in 51.3% of dehydrated yeast extract with 57.9% protein. The use of 12.2% NH3 at 60ºC after autolysis (8h) and plasmolysis (8h) was not viable due to the reduction in the RNA yield from 89.7to78.4%. On the other hand, the thermal shock at 60ºC for 15 minutes prior to autolysis provided an increase in the yield from 89.7 to 91.4%. The autolysis, including NaCl plasmolysis in the optimized conditions was efficient, economic and...

Myostatin (GDF8) single nucleotide polymorphisms in Nellore cattle. Repositório Alice
GRISOLIA, A. B.; D´ANGELO, G. T.; PORTO NETO, L. R.; SIQUEIRA, F.; GARCIA, J. F..
The myostatin gene, also known as GDF8 (growth differentiation factor 8), is located on bovine chromosome 2 (BTA2); it has three exons and two introns. Myostatin is specifically expressed during embryonic development and in adult skeletal muscle, functioning as a negative regulatory protein. Several cattle breeds (Piedmontese, Belgian Blue and Blond?Aquitaine, and others) show polymorphisms in this gene; these polymorphisms are directly related to the double muscling phenotype. We looked for polymorphisms in the Nellore cattle myostatin gene and compared them with those known for taurine breeds. Seven regions, covering the three exons of this gene, were amplified by polymerase chain reaction and sequenced, including the untranslated region. DNA from 30...

Nucleotide frequencies in human genome and Fibonacci numbers. Repositório Alice
YAMAGISHI, M. E. B.; SHIMABUKURO, A. I..
Abstract. This work presents a mathematical model that establishes an interesting connection between nucleotide frequencies in human single-stranded DNA and the famous Fibonacci's numbers. The model relies on two assumptions. First, Chargaff's second parity rule should be valid, and second, the nucleotide frequencies should approach limit values when the number of bases is sufficiently large. Under these two hypotheses, it is possible to predict the human nucleotide frequencies with accuracy. This result may be used as evidence to the Fibonacci string model that was proposed to the sequence growth of DNA repetitive sequences. It is noteworthy that the predicted values are solutions of an optimization problem, which is commonplace in many of nature's...

Oyster adenytate energy charge: response to levels of food ArchiMer
Moal, Jeanne; Le Coz, Jean-rene; Samain, Jean-francois; Daniel, Jean-yves; Bodoy, Alain.
Adenylatc Encrgy Charge (AEC) values of Crassostrea gigas (9 and 18 months old) cultured in ponds in Marennes-Oléron Bay (Atlantic coast, France) are reported. Two ponds were loaded with two different initial oyster biomasses in order to obtain different food conditions. The growth of oysters was different in the two ponds which corroborated the effectivences of attaining different food conditions. AEC level was different depending on the pond and was therefore related to food availability per unit oyster biomass. Well-fed oysters exhibited greater AEC values than poorly-fed oysters, throughout thc study. Information on nutritional situations for wild or cultured populations can therefore be obtained by AEC. In cach pond, the seasonal AEC variations were...

Ribonuclease Production by Aspergillus species Rev. Microbiol.
Gomes,Eleni; Silva,Roberto da; Serzedello,Alcides.
Ribonuclease production by Aspergillus flavipes, A. sulphureus and A. fischeri in semi-synthetic medium, after 24-144 hours at 30ºC under shaking, was studied. After cultivation, the medium was separated from micelia by filtration and the resultant solution was used as enzymatic extract. The highest amount of biomass and RNase was obtained after 96 hours of cultivation. The enzymes produced by three species presented similar characteristics, with optimum temperature at 55ºC and two peaks of activity at pH 4.5 and 7.0. A. flavipes RNases were more sensitive to temperature: 50% of the initial activity was lost after 1 hour at 70ºC. After this heat treatment, RNase of A. sulphureus lost 30% of this activity and that of A. fischeri only 16%. The nucleotides...

The aim of this experiment was to evaluate the effect of sources of trophic action (glutamine alone, glutamine associated with glutamic acid and yeast) associated with vaccination against coccidiosis on the performance and morphometry of the small intestine of broilers. In the trial, 1,200 broiler chicks were distributed according to a completely randomized design with a 3 x 2 + 2 (trophic action x vaccination or not + control - free trophic factor) factorial arrangement, with five replicates of 30 birds each. Vaccination negatively affected performance parameters and the morphometry of the intestinal mucosa, but at the end of the experimental period, among the broilers vaccinated against coccidiosis, the group fed glutamine presented better recovery from...
