Assessment of Genetic Variability of Fish Personality Traits using Rainbow Trout Isogenic Lines ArchiMer
Millot, Sandie; Pean, Samuel; Labbe, Laurent; Kerneis, Thierry; Quillet, Edwige; Dupont-nivet, Mathilde; Begout, Marie-laure.
The study of inter-individual variability of personality in fish is a growing field of interest but the genetic basis of this complex trait is still poorly investigated due to the difficulty in controlling fish genetic origin and life history. When available, isogenic lines that allow performing independent tests on different individuals having identical genotype constitute a very relevant experimental material to disentangle the genetic and environmental components of behavioural individuality. We took advantage of heterozygous isogenic lines to investigate the personality in rainbow trout through the analysis of their reactions to different experimental situations. To this end, seven to ten rainbow trout isogenic lines were screened for their spatial...

Body composition and sensory quality of wild and farmed brown-trout (Salmo trutta) and of farmed rainbow-trout (Oncorhynchus mykiss) Ciência Rural
Antão-Geraldes,Ana Maria; Hungulo,Sílvia Rafael; Pereira,Etelvina; Teixeira,Amílcar; Teixeira,Alfredo; Rodrigues,Sandra.
ABSTRACT: Body composition (total crude protein, lipid, ash, dry matter and moisture) and fatty acid profiles were compared between wild and farmed brown trout and between farmed rainbow trout. Farmed brown trout contained the highest amount of crude protein (18.39%), whereas farmed rainbow trout contained higher levels of crude lipid (2.35%). Thirty six fatty acids were found, including sixteen saturated fatty acids (SFA), nine monounsaturated fatty acids (MUFA) and eleven polyunsaturated fatty acids (PUFA). The most abundant SFA in all fish were palmitic acid and stearic acid. However, the most abundant fatty acids in all trout samples were MUFA and PUFA. MUFA were the most abundant fatty acid in farmed brown trout because of high abundance of oleic...

Comparison of brown trout (Salmo trutta) reared in fresh water and sea water to freshwater rainbow trout (Oncorhynchus mykiss): II. Phosphorus balance ArchiMer
Dosdat, Antoine; Metailler, Robert; Desbruyeres, Elisabeth; Huelvan, Christine.
Brown trout and rainbow trout (average weight 100 g) were reared in fresh water at 12 degrees C under the same conditions before transferring brown trout to sea water, in order to compare phosphorus utilisation in both species. Apparent phosphorus availability, orthophosphate excretion and phosphorus accretion in the fish were directly determined. Thus, actual phosphorus mass balance was built. Rainbow trout raised in fresh water had a higher phosphorus retention coefficient (maximum 50 %) than brown trout reared in fresh water (maximum 45 %). Transferring brown trout to sea water induced a reduction in phosphorus retention (maximum 39 %), Orthophosphate excretion, ranging 7-20 mg phosphorus per kg wet weight per day, represented 10-20 % of ingested...

Cryopreservation of semen from functional sex-reversed genotypic females of the rainbow trout, Oncorhynchus mykiss BABT
Ninhaus-Silveira,Alexandre; Foresti,Fausto; Tabata,Yara Aiko; Rigolino,Marcos Guilherme; Veríssimo-Silveira,Rosicleire.
Cryopreservation of semen from sex-reversed females of rainbow trout aims at rationalizing the production of stocks composed by 100% females. Semen from normal males (M) and two types of genotypic females (R and G), sex-reversed by the oral administration of 17alpha-methyltestosterone, were used. R was obtained by the fertilization of normal eggs with semen of sex-reversed females while G via gynogenetic reproduction. Semen was diluted in an extender solution (glucose 5,4 g, egg yolk 10 ml, dimetil sulfoxide 10 ml, water 80 ml) at 1:3 ratio (semen/extender), stored in straws of 0.5 ml and freezed in a dry container Cryopac CP-65, at -180ºC. Thawing was performed with water at 70ºC for 3 seconds. There were no significant fertilization rate differences...

Hepatic phosphoenolpyruvate carboxykinase gene expression is not repressed by dietary carbohydrates in rainbow trout (Oncorhynchus mykiss) ArchiMer
Panserat, Stéphane; Plagnes Juan, E; Breque, Jeanine; Kaushik, Sadasivam.
Phosphoenolpyruvate carboxykinase (PEPCK) is a rate-limiting enzyme in hepatic gluconeogenesis and therefore plays a central role in glucose homeostasis. The aim of this study was to analyse the nutritional regulation of PEPCK gene expression in rainbow trout (Oncorhynchus mykiss), which are known to use dietary carbohydrates poorly. A full-length hepatic PEPCK cDNA (2637 base pairs with one open reading frame putatively encoding a 635-residue protein) was cloned and found to be highly homologous to mammalian PEPCKs. The presence of a putative peptide signal specific to a mitochondrial-type PEPCK in the deduced amino acid sequence suggests that this PEPCK gene codes for a mitochondrial form, In gluconeogenic tissues such as liver, kidney and intestine,...

How to assess fin damage in rainbow trout, Oncorhynchus mykiss? ArchiMer
Person, Jeannine; Le Bayon, Nicolas; Gros, Sylvie.
Fin condition was examined in 100-500 g hatchery-reared rainbow trout sampled in a large range of environmental conditions of an experimental farm. A method that is simple to use, requires little training, and does not need costly sophisticated equipment or reference to a model is proposed to assess fin erosion (defined as a decrease in size and profile alteration of fins). A procedure to reliably measure the length of the longest ray present in two selected fins (dorsal and pectoral) is described. Fin general profile is assessed using five levels of erosion based on the precise rank of intact or eroded rays to limit subjective bias. The suitability of the two descriptors of fin erosion under laboratory, farm or field conditions is discussed.

Impact of rainbow trout on aquatic invertebrate communities in subtropical mountain streams of northwest Argentina Ecología austral
Molineri,Carlos.
The present study was conducted to assess the effect of rainbow trout introduction on benthic invertebrate communities, stratified as epibenthos and infauna. Two main questions are explored: 1) do the trout-invaded streams show a different community structure than the streams with autochthonous fish?, and 2) does the presence of trout affect differentially the epibenthos and the infauna? Epibenthic and infaunal samples, drift samples and fish stomach content were sampled four times from three stations to assess the impact of rainbow trout (Oncorhynchus mykiss) on the invertebrate aquatic fauna. Only one species of native fish was recorded in the studied sites, the siluriform Trichomycterus corduvense. Both fish species were found always in allopatry. The...

Ionic composition of endolymph in teleosts: Origin and importance of endolymph alkalinity ArchiMer
Payan, P; Kossmann, Hans; Watrin, A; Mayer-gostan, N; Boeuf, Gilles.
Ionic (Na+, K+, Cl-, PO43-, pH), total CO2, total calcium and protein concentrations in the plasma and endolymph of the inner ear were compared in trout Oncorhynchus mykiss and turbot Scophthalmus maximus. In both species, saccular endolymph was characterized by high levels of K+ and total CO2 and in trout by an alkaline pH, The kinetic characteristics of proton secretion across the saccular epithelium of trout were investigated using a titration technique in which isolated saccules were mounted as closed sacs. The rate of proton secretion depends strongly on the pH of the Ringer's solution and secretion stops at a pH below 7.2, Proton secretion is driven by an energy-dependent mechanism involving basolateral ouabain-sensitive Na+/K+ exchangers. Proton...

Modeling the weight gain of freshwater-reared rainbow trout ( Oncorhynchus mykiss ) during the grow-out phase R. Bras. Zootec.
Janampa-Sarmiento,Peter Charrie; Takata,Rodrigo; Freitas,Thiago Mendes de; Freire,Licius de Sá; Pereira,Marcelo Menezes de Britto; Lugert,Vincent; Heluy,Guilherme Melgaço; Pereira,Marcelo Maia.
ABSTRACT We used five nonlinear models to calculate the weight gain of rainbow trout (122.11±15.6 g) during the final grow-out phase of 98 days under three different feed types (two commercials diets, A and B, and one experimental diet, C) in triplicate groups. We fitted the von Bertalanffy growth function with allometric and isometric scaling coefficient, Gompertz, Logistic, and Brody functions to weight (g) at age data of 900 fish, distributed in nine tanks. The equations were fitted to the data based on the least squares method using the Marquardt iterative algorithm. The accuracy of the fitted models was evaluated using a model performance metrics, combining mean squared residuals (MSR), mean absolute error (MAE), and Akaike’s Information Criterion...

Nutritional regulation and tissue specificity of gene expression for proteins involved in hepatic glucose metabolism in rainbow trout (Oncorhynchus mykiss) ArchiMer
Panserat, Stéphane; Plagnes Juan, E; Kaushik, Sadasivam.
Rainbow trout (Oncorhynchus mykiss) are known to use dietary carbohydrates poorly. One of the hypotheses to explain the poor utilisation of dietary glucose by these fish is a dysfunction in nutritional regulation of hepatic glucose metabolism. In this study, we obtained partial clones of rainbow trout cDNAs coding for a glucose transporter (Glut2), and for the enzymes 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (6PF-2K/F-2,6BPase), fructose-1,6-bisphosphatase (FBPase) and pyruvate kinase (PI(), Their deduced amino acid sequences were highly similar to those of mammals (up to 80% similarity), In a study of nutritional regulation, the Glut2 gene was highly expressed in the liver irrespective of the nutritional status of the trout, in agreement with...

Otolith growth in trout Oncorhynchus mykiss: supply of Ca2+ and Sr2+ to the saccular endolymph ArchiMer
Payan, P; Borelli, G; Priouzeau, F; De Pontual, Helene; Boeuf, G; Mayer Gostan, N.
Kinetic and pharmacological characteristics of Ca2+ fluxes across the saccular epithelium of trout were studied using a perfused isolated inner ear. Ca-45(2+) influx from the Ringer solution to the endolymph was 3-4 nmoles h(-1) mu(-1) endolymph, which corresponds to a global turnover rate of the endolymph calcium of 200% h(-1). Ca2+ entry into the proximal endolymph was faster than into the distal fluid. Net Ca2+ movement across the saccular epithelium depended on the direction and intensity of the chemical gradient of calcium between the Ringer solution and the endolymph. Increasing the calcium concentration in the Ringer solution up to 4.4 mmol l(-1) provoked an accumulation of Ca2+ in both proximal and distal endolymphs, and equilibrium was reached...

Reproduction uniparentale et polyploidie induites chez la truite arc-en-ciel (Oncorhynchus mykiss) et chez les bivalves Crassostrea gigas, Ruditapes philippinarum et Chlamys varia ArchiMer
Diter, Alain.
Methods of gynogenesis, androgenesis and polyploidy induction were developed with the aim of genetic improvement in the rainbow trout, the Pacific oyster, the Manila clam and the black scallop. In the trout: · haploid androgenesis was induced by means of a pressure shock applied on diploid eggs, probably at the time of karyogamy. Diploid androgenetic individuals were directly produced from eggs fertilized with sperm from tetraploid male. · a heat shock treatment effective for suppressing the first egg mitosis was developed. This treatment allows the production of homozygous gynogenetic lines (checked using enzyme electrophoresis technique) from eggs inseminated with ultraviolet irradiated sperm. The treatment also induced tetraploidy in eggs normally...

The combined effects of feeding time and dietary fat levels on feed intake, growth and body composition in rainbow trout ArchiMer
Gelineau, Anne; Bolliet, Valérie; Corrraze, Geneviève; Boujard, Thierry.
The combined effects of feeding time (morning and evening) and dietary fat concentration on feed intake, growth, body composition and lipid tissue distribution were examined in rainbow trout fed on demand. To that purpose, diets with low (LE, 6% lipid) or high energy concentrations (HE, 24% lipid) were used in four treatments that combined provision of the same (HE-HE or LE-LE) or different (HE-LE, LE-HE) diets at morning and evening meals. Digestible energy intakes of the LE-HE (229 U kg(-1) d(-1)) and HE-HE (269 U kg(-1) d(-1)) groups were significantly different. There was no significant difference in the amount of energy intake between the two meals of the day in any of the treatments. The main effect of dietary treatment was on lipid intake; fish in...

The spawning migration of anadromous rainbow trout in the Santa Cruz River, Patagonia (Argentina) through radio-tracking Ecología austral
Riva Rossi,Carla; Arguimbau,Milagros; Pascual,Miguel.
Adult anadromous rainbow trout (Oncorhynchus mykiss) from the Santa Cruz River were implanted with radio-transmitters in May 2000 as they ascended the river from the ocean to locate spawning areas, assess the range and timing of the spawning migration, and determine the composition of the spawning populations. Radio-tagged fish revealed geographic and temporal migration patterns and previously unidentified spawning areas. Telemetry data indicated that anadromous fish dwell and spawn in the main stem river. Range of the total detected upstream migration varied between 2-280 km. Spawning begins by mid winter and peaks in the spring, matching the raise in water temperature, which ranged from 4ºC at the beginning of the spawning season to 10ºC at the end of...

Voluntary feed intake and discrimination of diets containing a novel fluoroquinolone in self-fed rainbow trout ArchiMer
Boujard, Thierry; Le Gouvello, Raphaela.
The objective of the study was to evaluate the capacity of rainbow trout to discriminate diets containing micro-granulated or powdered novel fluoroquinolone in comparison with a positive (containing sulfamerazine sodium) and a negative control (placebo). In a first trial, groups of trout had access to a self-feeder containing the placebo feed for 10 days, and then to one of the four feeds during another period of 10 days. Fish reduced their voluntary feed intake (VFI) by more than 50 % when they had access to the positive control, and by more than 20 % when they had access to feed containing micro-granulated premix. There was no significant decrease in VFI when the fish had access to feed containing powdered premix, but in this latter case, an increase in...
