A transcriptomic approach of salinity response in the euryhaline teleost, Dicentrarchus labrax 5
Boutet, Isabelle; Ky, Chin Long; Bonhomme, Francois.
Euryhaline teleosts possess the capacity to osmoregulate under various environmental conditions (freshwater to hypersaline water). This physiological capacity is generally monitored using enzyme activity assays (Na+/K+-ATPase...), hormones quantification (prolactine, growth hormone) or their mRNAs expression. To date, few studies addressed the genetic correlates of adaptation to varying salinity at a molecular level in such fish. In the sea bass Dicentrarchus labrax, genetic differentiation was observed at specific allozyme loci between lagoon- and open-sea populations. In the present study, we investigated transcriptomic response of D. labrax to salt- and freshwater acclimation in two organs involved in osmoregulation, gill and intestine. By using...

Acclimation of juvenile Mugil liza Valenciennes, 1836 (Mugiliformes: Mugilidae) to different environmental salinities 83
Lisboa,Viviana; Barcarolli,Indianara F.; Sampaio,Luís A.; Bianchini,Adalto.
Survival and physiological parameters associated with metabolism and osmoregulation were evaluated in juveniles of the Lebranche mullet Mugil liza acclimated to different water salinities (5, 10, 20, 30, and 40‰) for 15 days. Room temperature (25ºC) and photoperiod (12L:12D) were fixed. Fish were fed twice-a-day with commercial diet (28% crude protein) until satiation. After acclimation, whole body oxygen consumption was measured and fish were euthanized and sampled for blood, gills, and liver. Whole body oxygen consumption and plasma osmolality did not change in the range of salinities tested. The isosmotic point was estimated as 412.7 mOsmol kg-1 (13.5‰). Gill Na+,K+-ATPase activity tended to be lower at 20 and 30‰, while liver glycogen content was...

Accumulation of organic and inorganic solutes in NaCl-stressed sorghum seedlings from aged and primed seeds 63
Oliveira,Alexandre Bosco de; Alencar,Nara Lídia Mendes; Prisco,José Tarquinio; Gomes-Filho,Enéas.
Although it has been known that the seed's physiological potential affects its response to osmoconditioning and abiotic stresses, researches involving seed aging and priming associated to abiotic stresses are scarce. The aim of this work was to evaluate the role of seed priming on salt tolerance in sorghum seedlings from seeds with two vigor levels (aged or non-aged) and to verify the organic and inorganic solute contributions as osmoregulators in NaCl-stressed seedlings from aged and primed seeds. The combinations of two seed vigor levels (aged or not), two seed types (primed or not) and two salinity levels (exposed to NaCl at 100 mM or not) were evaluated. In low physiological quality seeds (aged seeds), priming provided an attenuation of salinity's...

Activity determination of Na+ K+ - ATPase and Mg++ - ATPase enzymes in the gill of Poecilia vivipara (Osteichthyes, Cyprinodontiformes) in different salinities 52
Amaral,Marcelo da Cunha; Bonecker,Ana Cristina Teixeira; Ortiz,Cláudio H. D..
This work aimed to know the tolerance mechanisms through the salinity variation by Na+ K+ - ATPase and Mg++ - ATPase and enzymes encountered in the gills of Poecilia vivipara. In field, the presence of this species was observed in salinities of 0 and 28‰. In laboratory, these fish were maintained in aquarium with mean salinity of 30‰ and positive responses were obtained. Some adult specimens, collected in a lagoon of the Coqueiros Beach, were utilized as matrixes. In the experiments the specimens used were those born in the test aquarium. For each salinity studied three replicates were made with three specimens for each one. The alevins were maintained in salinities of 5, 10, 15, 20, 25, 30 and 35‰ during a month for adaptation. Gills were extracted in...

Adaptations biochimiques intestinales chez la truite: Influence de l'aliment et de la salinite sur quelques proprietes membranaires 5
Leray, C.
The authors report the results of physiological and biochemical experiments which confirm the important role player by fish (Salmo gairdneri ) intestinal mucosa in their osmoregulatory process. The ability of the dietary lipids to modulate this function is described. It was demonstrated that the ionic apical permeability of the enterocytes are related to their membrane lipid composition. The importance of the essential fatty acids is emphasized. The time course changes of the apical permeability are of value in improving sea water adaptation, similar conclusions are reported for Nacl supplementation.

An economic evaluation of the osmoregulation gene technology to the Australian wheat industry 31
Farquharson, Robert J.; Morgan, J.M.; Brennan, John P..
Episodes of rainfall irregularity and soil moisture deficit have focused attention on the widespread limitation of water supply on winter cereal crop production in Australia. This has motivated a number of efforts at breeding for improved drought tolerance. A recent example involves a cellular adaptation which mitigates water loss through solute accumulation (osmoregulation or osmotic adjustment). An assessment of the performance of osmoregulation yield response in the presence of climate change found that wheat cultivars with this gene are unlikely to be adversely affected by hotter and drier conditions across the wheat belt. The results of an economic evaluation of potential future innovations (wheat cultivars) from the osmoregulation gene technology are...

Change of osmoregulatory and hematological parameters in tilapia (Oreochromis niloticus) after exposure to sublethal mercury concentrations 5
Handayani, Kiki Syaputri; Soegianto, Agoes; Lignot, Jehan-hervé.
The effects of Hg exposure on blood parameters and gill physiology of tilapia (Oreochromis niloticus) were analyzed. Fish maintained in freshwater were exposed for 7 days (d) to sublethal mercury concentrations (0.1 and 1 mg/L). Blood serum osmolality (SO), sodium (Na+), potassium (K+) and chloride (Cl−) ionic concentrations, and hematological parameters were assessed after 1 up to 7 d of exposure. Serum osmolality and ionic concentrations of exposed fish appeared differently affected throughout the experimental period compared to the controls. Osmolality was reduced at the 2 tested concentrations but Na+ and Cl− contents were only altered at 1 mg/L of Hg after 1 d of exposure and values rapidly returned to the control values thereafter. K+ content was...

Could a diet enriched with n-3 highly unsaturated fatty acids be considered a promising way to enhance the immune defences and the resistance of Penaeid prawns to environmental stress? 5
Chim, Liet; Lemaire, Pierrette; Delaporte, Maryse; Le Moullac, Gilles; Galois, Robert; Martin, Jean-louis.
The prawn Penaeus stylirostris (Stimpson), when fed for 28 days with n-3 highly unsaturated fatty acid (HUFA)-enriched feed pellets, demonstrated an enhanced resistance to variations in environmental parameters (a decrease in temperature and salinity over a 4-day period from 28 degreesC to 17 degreesC and from 35 parts per thousand to 10 parts per thousand respectively) and an improvement in their immune defence potential, i.e. increased agglutination titre of plasma and increased respiratory burst of haemocytes.

Description of the unusual digestive tract of Platax orbicularis and the potential impact of Tenacibaculum maritimum infection 5
Alix, Maud; Gasset, Eric; Bardon-albaret, Agnes; Noel, Jean; Pirot, Nelly; Perez, Valérie; Coves, Denis; Saulnier, Denis; Lignot, Jehan-hervé; Cucchi, Patricia N..
Background Ephippidae fish are characterized by a discoid shape with a very small visceral cavity. Among them Platax orbicularis has a high economic potential due to its flesh quality and flesh to carcass ratio. Nonetheless, the development of its aquaculture is limited by high mortality rates, especially due to Tenacibaculum maritimum infection, occurring one to three weeks after the transfer of fishes from bio-secure land-based aquaculture system to the lagoon cages for growth. Among the lines of defense against this microbial infection, the gastrointestinal tract (GIT) is less studied. The knowledge about the morphofunctional anatomy of this organ in P. orbicularis is still scarce. Therefore, the aims of this study are to characterize the GIT in...

Differential distribution of V-type H+-ATPase and Na+/K+-ATPase in the branchial chamber of the palaemonid shrimp Macrobrachium amazonicum 5
Boudour-boucheker, Nesrine; Boulo, Viviane; Charmantier-daures, Mireille; Grousset, Evelyse; Anger, Klaus; Charmantier, Guy; Lorin-nebel, Catherine.
V-H+-ATPase and Na+/K+-ATPase were localized in the gills and branchiostegites of M. amazonicum and the effects of salinity on the branchial chamber ultrastructure and on the localization of transporters were investigated. Gills present septal and pillar cells. In freshwater (FW), the apical surface of pillar cells is amplified by extensive evaginations associated with mitochondria. V-H+-ATPase immunofluorescence was localized in the membranes of the apical evaginations and in clustered subapical areas of pillar cells, suggesting labeling of intracellular vesicle membranes. Na+/K+-ATPase labeling was restricted to the septal cells. No difference in immunostaining was recorded for both proteins according to salinity (FW vs. 25 PSU). In the branchiostegite,...

Digestive tract ontogeny of Dicentrarchus labrax: Implication in osmoregulation 5
Giffard, I; Charmantier, G; Grousset, E; Aujoulat, F; Castille, R.
The ontogeny of the digestive tract (DT) and of Na+/K+-ATPase localization was investigated during the early postembryonic development (from yolk sac larva to juvenile) of the euryhaline teleost Dicentrarchus labrax reared at two salinities: seawater and diluted seawater. Histology, electron microscopy and immunocytochemistry were used to determine the presence and differentiation of ion transporting cells. At hatching, the DT is an undifferentiated straight tube over the yolk sac. At the mouth opening (day 5), it comprises six segments: buccopharynx, esophagus, stomach, anterior intestine, posterior intestine and rectum, well differentiated at the juvenile stage (day 72). The enterocytes displayed ultrastructural features similar to those of...

Dissolved oxygen and ammonia levels in water that affect plasma ionic content and gallbladder bile in silver catfish 65
Becker,Alexssandro Geferson; Garcia,Luciano de Oliveira; Kochhann,Daiani; Gonçalves,Jamile Fabbrin; Loro,Vania Lúcia; Baldisserotto,Bernardo.
Ionic contents (Na+, K+ and Cl-) of plasma and gallbladder bile (GB) of juveniles silver catfish, Rhamdia quelen (156.1±0.2g, 28.2±0.3cm), were determined in three different times (0, 6 and 24h) after exposure to: a) control or high dissolved oxygen (DO = 6.5mg L-1) + low NH3 (0.03mg L-1); b) low DO (3.5mg L-1) + low NH3; c) high DO + high NH3 (0.1mg L-1); and d) low DO + high NH3. High waterborne NH3 or low DO levels increased plasma and GB ion levels. These parameters might have followed different mechanisms to affect osmoregulation since a synergic effect of these variables was detected.

Evaporative water loss and dehydration during the night in hummingbirds 97
HARTMAN BAKKEN,BRADLEY; SABAT,PABLO.
Nectar-feeding birds oscillate between avoiding overhydration when they are feeding and preventing dehydration during fasts. Here, we examined how resting rates of total evaporative water loss (TEWL) and metabolic water production (MWP) influence water balance in the green-backed firecrown (Sephanoides sephanoides), a Chilean hummingbird. We hypothesized that a circadian rhythm in TEWL would assuage the dehydration risk that hummingbirds face during the night. However, we did not find support for this idea. In resting hummingbirds, rates of TEWL during the day (54 ±6 µLh-1,n = 8) and night (65 ± 12 h-1, n = 5) were similar. Rates of MWP were also similar between the day (22 ±3 µL h-1, n = 8) and night (23 ± 2 µL h-1, n = 5). MWP rates were significantly...

Growth and production components of West Indian cherry cultivated with saline waters and potassium fertilization 91
Lima,Geovani S. de; Pinheiro,Francisco W. A.; Dias,Adaan S.; Gheyi,Hans R.; Soares,Lauriane A. dos A.; Silva,Saulo S. da.
ABSTRACT Due to water limitations in terms of both quantity and quality in the semi-arid region of northeastern Brazil, the use of waters with high concentrations of salts become necessary in irrigated agriculture. Thus, this study aimed to evaluate the growth and production of grafted West Indian cherry under saline water irrigation and potassium fertilization. The experiment was carried out in drainage lysimeters under protected environment conditions, using a Regolithic Neosol with sandy loam texture. Treatments resulted from the combination of two levels of electrical conductivity - ECw (0.8 and 3.8 dS m-1) of irrigation water and four doses of potassium (50, 75, 100 and 125% of the recommendation of Musser), arranged in a randomized block design, with...

IMPROVING THE TOLERANCE OF Vicia faba AGAINST ENVIRONMENTAL SALINITY RESULTED FROM THE IRRIGATION WITH SEA WATER BY USING KNO3 AND (NH4)2SO4 AS CHEMICAL OSMOREGULATORS 37
MOHAMED,KAMEL.
The familiar solutes, Na+, K+, Ca2+, Mg2+, Cl-, PO4(3-), SO4(2-), soluble carbohydrates, amino acids and soluble proteins, which play a role in osmotic adjustment were estimated to investigate the role of potassium nitrate and ammonium sulphate as osmoregulators and their effects on the solutes composition. Vicia faba L. was cultivated and irrigated with 5, 10, 15 and 20 % (v:v) sea water. The plants were divided to three groups. The first was irrigated with sea water only. The second was treated with 5 mM KNO3 while the third was treated with 5mM (NH4)2SO4. The plants were left to grow until flowering stage. The results indicated that the non treated group increased the soluble carbohydrates in the roots to avoid the influx of sodium. The treatment with...

Incidence de la salinite sur les equilibres de distribution hydrique et ioniques et sur le metabolisme respiratoire dans le muscle blanc, le muscle rouge et la branchie chez le muge Chelon labrosus 5
Gallis, J; Piquemal, S; Lambert, J; Sergent, P; Rousseau, V.
Oxygen consumption, water, K super(+) and Na super(+) contents were measured in white muscle, red muscle and gill epithelium in the mullet Chelon labrosus) in relation with the duration of fresh water or sea water (1500mOsm) adaptation. The unit respiratory activities in gill and red muscle where respectively 5 and 3 timesmore elevated than in white muscle. The pericellular K super(+)/Na super(+) ratio of the 3 tissues decreased with the fresh water adaptation duration. In the gill, the intracellular K super(+)/Na super(+) ratio was stable while the parameter decreased strongly in the white muscle. The results are discussed in relation to energetic cost of homocellular regulation and homoepithelial regulation.

Inter-individual variability in freshwater tolerance is related to transcript level differences in gill and posterior kidney of European sea bass 5
L'Honoré, Thibault; Farcy, Emilie; Blondeau-bidet, Eva; Lorin-nebel, Catherine.
Acclimation to low salinities is a vital physiological challenge for euryhaline fish as the European sea bass Dicentrarchus labrax. This species undertakes seasonal migrations towards lagoons and estuaries where a wide range of salinity variations occur along the year. We have previously reported intraspecific differences in freshwater tolerance, with an average 30% mortality rate. In this study, we bring new evidence of mechanisms underlying freshwater tolerance in sea bass at gill and kidney levels. In fresh water (FW), intraspecific differences in mRNA expression levels of several ion transporters and prolactin receptors were measured. We showed that the branchial Cl-/HCO3- anion transporter (slc26a6c) was over-expressed in freshwater intolerant fish,...

Ion fluxes in silver catfish (Rhamdia quelen) juveniles exposed to different dissolved oxygen levels 83
Rosso,Felipe Link de; Bolner,Keidi C. S.; Baldisserotto,Bernardo.
Low dissolved oxygen levels in the water (hypoxia) can be provoked by oxygen consumption by fish and other organisms, organic matter decomposition, phytoplankton blooms, and temperature increase. The objective of the present study was to investigate Na+, Cl-, K+, and ammonia fluxes in silver catfish (Rhamdia quelen) exposed to different dissolved oxygen levels. Juveniles (9 ± 1g) maintained at 6.0 mg.L-1 dissolved oxygen were transferred to four 40 L aquaria with different dissolved oxygen levels (in mg.L-1): 6.0, 4.5, 3.5, and 2.5. In another series of experiments, juveniles were acclimated at 6.0 or 2.5 mg.L-1 dissolved oxygen levels, and then placed in two 40 L aquaria with 6.0 mg.L-1 dissolved oxygen. Ion uptake pathways in European sea bass Dicentrarchus labrax 5
Blondeau-bidet, Eva; Hiroi, Junya; Lorin-nebel, Catherine.
Ion uptake mechanisms are diverse in fish species, certainly linked to duplication events that have led to the presence of a multitude of paralogous genes. In fish, Na+ uptake involves several ion transporters expressed in different ionocyte subtypes. In the European sea bass Dicentrarchus labrax, several key transporters potentially involved in Na+ uptake have been investigated in seawater (SW) and following a 2 weeks freshwater (FW) acclimation. Using gel electrophoresis, we have shown that the Na+/H+-exchanger 3 (nhe3, slc9a3) is expressed in gills and kidney at both salinities. Quantitative realtime PCR analysis showed a significantly higher nhe3 expression in fresh water (FW) compared to SW. Its apical localization in a subset of gill ionocytes in...
