The sequencing of prfA, encoding the transcriptional regulator of virulence genes, in 26 low virulence field Listeria monocytogenes strains showed that eight strains exhibited the same single amino-acid substitution: PrfAK220T. These strains exhibited no expression of PrfA-regulated proteins and thus no virulence. This substitution inactivated PrfA, since expression of the PrfAK220T mutant gene in an EGDΔprfA strain did not restore the haemolytic and phosphatidylcholine phospholipase C activities, in contrast to the wild-type prfA gene. The substitution of the lysine at position 220 occurred in the helix αH. However, the data showed that the PrfAK220T protein is dimerized just as well as its wild-type counterpart, but does not bind to PrfA boxes....

A new RNA picorna-like virus in the cotton pink bollworm Pectinophora gossypiella (Lep. : Gelechiidae) in Egypt IRD

Un nouveau virus infectant le ver rose du cotonnier, #Pectinophora gossypiella$, a été isolé et purifié à partir de larves mortes récoltées en champ de coton. Les virions purifiés de 27 plus ou moins 1 nm de diamètre, ont une forme icosaédrique et contiennent un génome à ARN. Trois protéines capsidaires de 31,7, 32,6 et 47,4 Kd ont été trouvées dans les virions. Le virus purifié n'est pas hautement infectieux pour les larves, cependant la durée du stade chrysalide est supérieure chez les lots infectés artificiellement. Dans les tissus atteints, les virions sont dispersés dans le cytoplasme des cellules de l'intestin ou bien groupés en amas paracristallins de même type que chez d'autres insectes infectés avec des picornavirus. Absolute feeding design, a realistic way for fish nutrients requirements determination IRD

Feeding experiment is designed on the basis of food allowance rather than feed formulation. Food allowances are fixed in the attempt to provide the desired amount of nutricients per body mass of fish per day (in terms of g.kg-1.d-1 or J.kg-1.d-1). By this way, previous determination of optimum feeding ration is not required, and utilization of neutral feed complement (e.g. cellulose) is unnecessary. Experiment interpretation of this design is presented for a tipalia species, #Sarotherodon melanotheron$. Interrelationship of commonly used indices for growth and nutrition is discussed. Alimentation complémentaire de 38 nourrissons sénégalais IRD

Une enquête de consommation alimentaire par pesée a été faite pendant 3 jours chez 38 nourrissons sénégalais afin d'apprécier la part de l'alimentation complémentaire dans la couverture de leurs besoins nutritionnels. Les apports recommandés en énergie et en protéines sont satisfaits respectivement à 49 et à 74%. Les besoins nutritionnels sont couverts à des niveaux variables : calcium 38%, fer 24%, vitamine A 39%, B1 51%, B2 43%, PP 82%, C 54%, folates 8%, B12 263%. En tenant compte de l'estimation des apports assurés par le lait maternel, les apports recommandés en énergie et en protéines sont satisfaits pendant la période étudiée. (Résumé d'auteur)

La valeur énergétique nette d’un aliment est déductible de sa teneur en énergie digestible selon une équation valable pour le porc à tous les stades physiologiques. Elle modifie la hiérarchie des matières premières en abaissant celles riches en protéines ou en fibres et en relevant celles riches en amidon ou en matières grasses. La protéine idéale théorique, de valeur biologique 1, est l’équivalent de l’énergie nette pour la couverture du besoin azoté. L’approche expérimentale de cette protéine par la suppression des excès d’acides aminés, laisse espérer une importante marge de progrès du rendement d’utilisation de l’azote absorbé. Ce rendement mesure une disponibilité métabolique de l’acide aminé limitant peu dépendante des matières premières utilisées....

Over the last 10 years, DNA sequences of more than 600 bacterial species have been deposited in databases and are now available to search any gene, motif, or regulatory sequence of interest. Although genome data are instrumental in phylogenetic analysis and in silico design of metabolic and regulatory networks, only a very small fraction of the information has been experimentally validated. A striking example is lipoproteins predicted from genome sequences. Background: Since the finishing of the sequencing of the Arabidopsis thaliana genome, the Arabidopsis community and the annotator centers have been working on the improvement of gene annotation at the structural and functional levels. In this context, we have used the large CATMA resource on the Arabidopsis transcriptome to search for genes missed by different annotation processes. Probes on the CATMA microarrays are specific gene sequence tags (GSTs) based on the CDS models predicted by the Eugene software. Among the 24 576 CATMA v2 GSTs, 677 are in regions considered as intergenic by the TAIR annotation. Background: Invasion of the mosquito salivary glands by Plasmodium is a critical step for malaria transmission. From a SAGE analysis, we previously identified several genes whose expression in salivary glands was regulated coincident with sporozoite invasion of salivary glands. To get insights into the consequences of these salivary gland responses, here we have studied one of the genes, PRS1 (Plasmodium responsive salivary 1), whose expression was upregulated in infected glands, using immunolocalization and functional inactivation approaches. Bacterioplankton production determined by DNA synthesis, protein synthesis, and frequency of dividing cells in Tuamotu atoll lagoons and surrounding ocean IRD

This study compares three independent methods used for estimating bacterioplankton production in waters from the lagoon (mesotrophic) and the surrounding ocean (oligotrophic) of two atolls from the Tuamotu archipelago (French Polynesia). Thymidine and leucine incorporation were calibrated in dilution cultures and gave consistent results when the first was calibrated against cell multiplication and the second against protein synthesis. This study demonstrates that determining conversion factors strongly depends on the selected calculation method (modified derivative, integrative, and cumulative).
