O desenvolvimento de linhagens homozigotas é um desafio nos programas de melhoramento de meloeiro. A ginogênese in vitro apresenta- -se como alternativa para produção de linhagens haploides e di-haploides. O objetivo deste trabalho foi a indução de calos e organogênese em óvulos de meloeiro. Óvulos das variedades inodorus, cantalupensis e reticulatus foram inoculados em meio de cultivo MS contendo 1,0 µM de BAP e 2,0 µM de ANA. Foram efetuados estudos histológicos nos ovários e óvulos, antes do cultivo in vitro, e nas fases de calogênese e organogênese. |

Abstract Dormancy in grape seeds slows the progress of many breeding programs of the crop, culminating in low uniformity and low germination percentages. Conventional methods used to overcome dormancy are time-consuming. Thus, in vitro seed germination emerges as a promising alternative to ensure the germination of grape seeds. In this study, we examined the in vitro germination and vigor of seedlings originating from seeds of ‘Red Globe’, ‘Italia’ and ‘Niagara Rosada’ grapes in growth media supplemented with five concentrations of gibberellic acid (GA3) (0, 1.41, 2.83, 4.24 and 5.66 µmol L-1) for 47 days. ABSTRACT In vitro rhizome production, encapsulation and cold storage of Acorus calamus were attempted for its propagation and ‘true-to-type’ conservation. Shoot cultures were initiated using underground rhizome buds, on 6-benzyladenine (BA) containing Murashige and Skoog (MS) medium. Maximum microrhizome production was observed in presence of 33.3 µM BA, on modified MS medium containing 6% sucrose, 100 mg/L citric acid and 1 g/L polyvinyl pyrrolidone-40. Synthetic seeds were produced from regenerated microtubers by encapsulation in calcium alginate beads. These synthetic seeds were stored in complete darkness at 100C temperature for different durations for mid-term conservation.
