Three selective enrichment broths - selenite cystine (SC), Muller-Kauffmann tetrathionate (MKT) and Rappaport-Vassiliadis (RV) - were compared, for Salmonella Typhimurium isolation from rectal swabs of a calf experimentally infected. The bacteriological procedure involved pre-enrichment in Hajna-GN broth (only for the samples inoculated in RV broth), selective enrichment (SC, MKT and RV broths), culture in modified brilliant green agar (BGA), presumptive biochemistry tests (using triple-sugar-iron agar and lysine-agar) and slide agglutination test with poli-O and poli-H Salmonella antisera. Construction and expression of aspartic protease from Onchocerca volvulus* as ompA fusion protein in a mutant strain of Salmonella typhimurium Genet. Mol. Biol.

Two constructions in pHS164 vector were designed to permit expression of OV7A and OV4A inserts encoding the N-terminal and C-terminal portion of an aspartic protease from Onchocerca volvulus, respectively. A novel 39 kD protein ompA-OV7A fusion protein was stably expressed as ompA fusion in a modified strain of Salmonella typhimurium strain SL5000 and E.coli strain JM109. Expression of the fusion protein in bacterial strains harboring the constructs were evaluated by western blotting. E.coli and Salmonella lysates were fractionated by 10% SDS-PAGE gel and then immobilized to nitrocellulose membrane by electroblotting. Depuration Times of Donax trunculus and Tapes decussatus BJM

The present study was performed to determine the depuration time and ability of Donax trunculus (Wedge Clam) and Tapes decussatus (Carpet Shell) contaminated with Escherichia coli, Salmonella enterica subsp. enterica serovar Typhimurium and Vibrio parahaemolyticus. Clams were contaminated with each bacterium at the level of 7.0 - 8.0 Log10 cfu/g. After contamination, clams were analyzed every 3 h in the first 24 h time period and every 6 h until the 72nd hour. During the depuration process of both clams, the level of bacteria decreased quickly to 40% of initial load in the first 12 h. El estudio tuvo como objetivo estandarizar y validar la técnica de Reacción en Cadena de la Polimerasa (PCR) para la detección de Salmonella typhimurium (St) en carne de cerdo, res y pollo, optimizando el tiempo y la sensibilidad con relación al método microbiológico; además se realizó el diagnóstico en estos tipos de carnes para evaluar su calidad microbiológica en la Ciudad de Texcoco. Un total de 60 muestras de carne (20 de cerdo, 20 de res y 20 de pollo) de 250 g fueron recolectadas en puntos de venta de la Ciudad de Texcoco. Infections with Salmonella serotypes are a major cause of food-borne diseases worldwide. Animal models other than the mouse have been employed for the study of nontyphoidal Salmonella infections because the murine model is not suitable for the study of Salmonella-induced diarrhea. The microbe has developed mechanisms to exploit the host cell machinery to its own purpose. Bacterial proteins delivered directly into the host cell cytosol cause cytoskeletal changes and interfere with host cell signaling pathways, which ultimately enhance disease manifestation. Role of previous stimulation on the neutrophil migration protects against the lethal effect of Salmonella typhimurium Braz. J. Vet. Res. Anim. Sci.

The intraperitoneal inoculation of 3.8 x 10(9) CFU of alive Salmonella typhimurium caused 100% mortality in rats. In these animals a small number of neutrophils migrated to the peritoneal cavity. The intraperitoneal inoculation of the animals with thioglycollate (Tg, 90 mg), carrageenin (Cg, 500 mu g), or killed S. typhimurium (KS, 1.9 x 10(9) CFU) 24 hours before challenge with alive S. typhimurium caused a significant neutrophil migration to the peritoneal cavity (determined 6 hours after challenge) and protected the animals against the lethal effect of the bacteria at a rate of 50%, 83% and 80%, respectively. However, similar treatment of the animals with brain heart infusion broth (BHI, S. typhimurium grout medium) protected the animals by only 23%....

銀耳多醣產品毒理及致突變性評估 Taiwan Agricultural Research Institute

[[abstract]]古籍記載,銀耳性味甘、平具有滋陰潤肺、養胃生津之功效。我們的研究顯示,從 銀耳子實體萃取而得之銀耳多醣,可以有效改善便秘症狀,為確保長期大量食用銀耳多 醣的安全性,進而執行本次安全性實驗。安全性試驗乃依據衛生署食品安全性評估規 範。首先進行銀耳多醣對大鼠 (SD 品系) 之口服急毒性試驗,依體重經口單次餵食大鼠 最高測試劑量5 g/kg 體重,之後連續觀察14 天。分析血液學、白血球分類及體內重要 臟器變化結果顯示,全部供試大鼠均無中毒症狀或死亡,且均無出現與試驗物質有關之 肉眼及組織病理變化,顯示銀耳多醣體對大鼠之口服急毒性LD50 值為大於5 g/kg 體重。 銀耳多醣並進行Ames 試驗,對五株組胺酸缺乏之沙門氏菌 TA98、TA100、TA102、 TA1535 及TA1537 進行回復突變之致變異性實驗。試驗顯示銀耳多醣不論未經或經S9 作用後與各菌株共同培養,各濃度銀耳多醣的細菌回復突變菌數與陰性對照組相比均無 顯著差異,顯示銀耳多醣體對供試沙門菌株並不具有致變異性,對細菌基因回復突變測 試為陰性反應。顯示銀耳多醣為一安全性的物質。
