Acetylsalicylic acid and labeling of blood constituents with technetium-99m BABT

Acetylsalicylic acid is the drug most used an anti-inflammatory agent and for secondary prevention of thrombotic phenomenon. Drugs can modify the labeling of blood constituents with technetium-99m (99mTc). The aim of this work was to evaluate the effect of in vitro or in vivo assays with acetylsalicylic acid on the labeling of the blood constituents with 99mTc. In vitro assay was performed with samples of whole blood from Wistar rats incubated with acetylsalicylic acid (1.0 mg/ml) for one hour before the 99mTc-labeling process. Knowledge of the radiochemical purity of radiopharmaceuticals is mandatory and can be evaluated by several methods and techniques. Planar chromatography is the technique normally employed in nuclear medicine since it is simple, rapid and usually of low cost. There is no standard system for the chromatographic technique, but price, separation efficiency and short time for execution must be considered. We have studied an alternative system using common chromatographic stationary phase and alcohol or alcohol:chloroform mixtures as the mobile phase, using the lipophilic radiopharmaceutical [99mTc(MIBI)6]+ as a model. Vitex agnus castus (VAC) is utilized in popular medicine and some actions have been attributed to its extract. Buzhong Yi Qi Wan (Buzhong) is a medicinal herb widely used in Traditional Chinese Medicine to treat the digestive and circulatory systems. Red blood cell and plasma proteins labeled with technetium-99m (99mTc) are used in nuclear medicine. The aim of this work was to investigate the effects of an aqueous Buzhong extract on the labeling of blood constituents with 99mTc. Heparinized blood (Wistar rats) was incubated in vitro with different Buzhong extract concentrations and 99mTc-labeling was performed. Sources of magnetic field, radiofrequency and audible sonic waves and pulsed light have been used in physiotherapy to treat different disorders. In nuclear medicine, blood constituents(Bl-Co) are labeled with technetium-99m (99mTc) are used. This study evaluated the consequences of magnetic field, radiofrequency and audible sonic waves and intense pulsed light sources on the labeling of Bl-Co with 99mTc. This study evaluates the effects of an acute treatment with a Centella asiatica (CA) extract on the biodistribution of the radiopharmaceutical Na99mTcO4 and on the fixation of technetium-99m on blood constituents. Wistar rats were treated with CA extract and, 1 hour after, Na99mTcO4 was administered; organs/tissues were withdrawn and weighted. Artemisia vulgaris (AV) is an antihelmintic and antimalarial drug; Aloe vera(babosa) acts as antidiabetic, laxative and anti-inflammatory;Benznidazole (BZ) is a trypanocidal of Trypanosoma cruzi (TC). Technetium-99m (99mTc) has been used in nuclear medicine to obtain diagnostic images. This study evaluated the plant effects in TC parasitemia and on the biodistribution of 99mTc in mice. The aim of this study was to evaluate the influence of the proteic extract of R. communis on the cell physiology by the osmotic fragility, labeling of the blood elements with the 99mTc and cell morphology. To evaluate the osmotic fragility, the blood samples of the Wistar rats were incubated with the concentrations of R. communis and with the solutions of NaCl (0.4; 0.7; 0.9%). The radiopharmaceutical sodium pertechnetate (Na99mTcO4) is used to obtain diagnostic images of the stomach, thyroid, parathyroids, salivary glands, brain and in the study of esophageal reflux and blood flow. An anti-carcinoembryonic antigen (CEA) monoclonal antibody (mAb 6D1.1) was evaluated in vitro and in vivo to determine its suitability as a tracer for immunoscintigraphy of colorectal carcinomas. Determination of mAb affinity for CEA showed a constant of association of 0.63 ± 0.11 x 109 M-1. Binding of technetium-99m (99mTc)-6D1.1, labeled by a direct method, to human cultured lineages was highly specific. Binding to only CEA-positive LS-174T cells resulted in a saturable curve inhibited by pre-incubation with unlabeled mAb. No binding at all was observed for the human lineages MeWo (melanoma) or ZR75-30 (breast carcinoma), neither of them expressing CEA cells. Evaluation of possible failure of the mononuclear phagocyte system after total splenectomy in rats BABT

Young and adult Wistar rats were submitted to total splenectomy and compared to animals not submitted to any surgical manipulation in order to evaluate the phagocytic function of spleen. The animals were infected with Escherichia coli labeled with technetium-99m and killed 20 minutes later. Liver, lung, spleen and a blood clot sample were taken. No significant differences were found in the percentage of bacterial radioactivity uptake in mononuclear phagocyte system (MPS) organs in young and adult splenectomized rats. However, phagocytosis index by macrophages of MPS organs was smaller in splenectomized animals than in control group. Sambucus australis (sabugueiro) has been used to treat inflammatory and rheumatologic disorders. Blood constituents labeled with technetium-99m (99mTc) have been used in nuclear medicine to obtain diagnostic images. The aim of this work was to evaluate the effect of a sabugueiro extract on the labeling of blood cells with 99mTc. Blood samples from Wistar rats were incubated with sabugueiro extract and the radiolabeling assay of blood constituents was carried out. After centrifugation, samples of plasma and blood cells were separated. Aliquots of plasma and blood cells were precipitated with trichloroacetic acid and centrifuged to isolate soluble and insoluble fractions.
