A novel frameshift mutation of Chediak-Higashi syndrome and treatment in the accelerated phase BJMBR
Wu,X.L.; Zhao,X.Q.; Zhang,B.X.; Xuan,F.; Guo,H.M.; Ma,F.T..
Chediak-Higashi syndrome (CHS) is a rare autosomal recessive immunodeficiency disease characterized by frequent infections, hypopigmentation, progressive neurologic deterioration and hemophagocytic lymphohistiocytosis (HLH), known as the accelerated phase. There is little experience in the accelerated phase of CHS treatment worldwide. Here, we present a case of a 9-month-old boy with continuous high fever, hypopigmentation of the skin, enlarged lymph nodes, hepatosplenomegaly and lung infection. He was diagnosed with CHS by gene sequencing, and had entered the accelerated phase. After 8 weeks of therapy, the boy had remission and was prepared for allogenic stem cell transplantation.

A simple approach to cancer therapy afforded by multivalent pseudopeptides that target cell-surface nucleoproteins Inra
Destouches, D.; Page; Hamma-Kourbali, Y.; Machi, V.; Chaloin, O.; Frechault, S.; Birmpas, C.; Katsoris, P.; Beyrath, J.; Albanese, P.; Maurer, M.; Carpentier, G.; Strub, J.M.; Van Dorsselaer, A.; Muller, S.; Bagnard, D.; Briand, J.P.; Courty, J.
Recent studies have implicated the involvement of cell surface forms of nucleolin in tumor growth. In thisstudy, we investigated whether a synthetic ligand of cell-surface nucleolin known as N6L could exert antitumoractivity. We found that N6L inhibits the anchorage-dependent and independent growth of tumor cell lines andthat it also hampers angiogenesis. Additionally, we found that N6L is a proapoptotic molecule that increasesAnnexin V staining and caspase-3/7 activity in vitro and DNA fragmentation in vivo. Through affinity isolationexperiments and mass-spectrometry analysis, we also identified nucleophosmin as a new N6L target. Notably, inmouse xenograft models, N6L administration inhibited human tumor growth. Biodistribution studies carriedout in...

Antileishmanial and antitrypanosomal activity of the cutaneous secretion of Siphonops annulatus J. Venom. Anim. Toxins incl. Trop. Dis.
Pinto,Erika Gracielle; Antoniazzi,Marta Maria; Jared,Carlos; Tempone,and Andre Gustavo.
Background Among the tropical parasitic diseases, those caused by protozoans are considered a challenge to public health, being represented by leishmaniasis and Chagas disease. In view of the low effectiveness and toxicity of the current therapy, animal venoms such as amphibian secretions have been used as a promising source of new drug prototypes. The present work aimed to achieve bioguided fractionation of metabolites present in a cutaneous secretion of the caecilian Siphonops annulatus (Amphibia: Gymnophiona: Siphonopidae) with antileishmanial and antitrypanosomal activity. Methods Through liquid-liquid partition and chromatographic techniques, the secretion was fractionated using bioguided assays. The 50% inhibitory concentration (IC50) of the main...

de Marco, Viviani; do Amaral, Ricardo Coutinho; Jericó, Márcia Marques; Silva, Ricardo Duarte; Simões, Denise Maria.
Twenty-four dogs, of different ages and breed (mongrels or not) were studied. The objective of the study was to evaluate the efficacy of determining glycosilated hemoglobin for the diagnosis and future monitoring of diabetic dogs. Animals were divided in a control group (12 healthy animals presenting glycemia levels ranging from 80 to 120 mg/dl, during fasting), and a diabetic group (12 animals presenting glycosuria and glycemia levels up to 220 mg/dl during fasting). Glycosilated hemoglobin concentration was determined for all the animals in the beginning of the study and again after three months of insulin therapy. In the beginning of the trial, normal values for 100% of the control group. For the not treated diabetic group high levels were found. After...

Double-blind trial of the efficacy of pentoxifylline vs thalidomide for the treatment of type II reaction in leprosy BJMBR
Sales,A.M.; Matos,H.J.de; Nery,J.A.C.; Duppre,N.C.; Sampaio,E.P.; Sarno,E.N..
Type II reaction in leprosy, or erythema nodosum leprosum (ENL), is often characterized by severe clinical symptoms together with nerve function impairment leading to permanent disabilities. Thalidomide has been shown to be a highly effective drug for the treatment of ENL. It is, however, contraindicated for women of childbearing age due to its teratogenicity. On the other hand, pentoxifylline, used to treat hypercoagulable states, is not teratogenic and, like thalidomide, can inhibit the synthesis of tumor necrosis factor-a and other cytokines. In the present randomized double-blind clinical study we compared the effectiveness of orally administered pentoxifylline vs thalidomide in treating type II reaction in 44 patients. Daily doses of 300 mg...

Efficacy of tannins from Mimosa pudica and tannic acid in neutralizing cobra (Naja kaouthia) venom J. Venom. Anim. Toxins incl. Trop. Dis.
Sia,FY; Vejayan,J; Jamuna,A; Ambu,S.
In the present study, the effectiveness of Mimosa pudica tannins (MPT) in neutralizing the lethality of Naja kaouthia venom was compared with commercially derived tannins. Preincubation of MPT with N. kaouthia venom maintained 100% survival of mice after 24 hours. The mouse group in which there was no preincubation, no protection against the effects of the venom was observed. M. pudica tannin was found to be more effective in neutralizing the lethality of N. kaouthia venom when compared to commercial tannic acid. Two protein spots were missing in the two-dimensional gel electrophoresis (2-DE) of the MPT treated mouse indicating the down-regulation of venom proteins. The results from this study indicated that tannins obtained from M. pudica are better than...

BACKGROUND: Urinary tract infections (UTIs) in elderly patients can be a complex problem in terms of approach to diagnosis, treatment, and prevention, because the patients often present nonspecific symptoms. The epidemiological and clinical characteristics of UTI in elderly women were studied, in order to make early diagnosis and prevent serious clinical complications secondary to UTI. METHODS: This was a prospective population-based study, with elderly women, during their first medical office visit. Medical records were obtained by clinical history and physical examination in order to detect signs and symptoms of UTI and the presence of comorbidities. Clean-catch midstream urine specimens for urinary dipstick test, sediment, and culture were collected;...

How to Evaluate and Interpret a Scientific Article About Therapy Or Therapeutic Procedures? International Journal of Morphology
Otzen,Tamara; Manterola,Carlos; García-Domínguez,Maricela.
Regardless of the subject area and design used, it has been verified that between 40% and 60% of the studies published in biomedical journals are articles about therapy or therapeutic procedures (TP). Anyone writing a manuscript related to therapy or TP or reading an article of this type must demand at the very least a clear, precise and concise objective with respect to the research conducted, explicit mention of the design used with the respective inherent methodological details, and the mention and execution of statistical tools and the measures of association, or at least the numbers needed to calculate these values. The aim of this manuscript is to present a synthesis of the fundamental elements for the correct writing, reading and assessment of such...

Imaging and therapy with radionuclide labeled magnetic nanoparticles BABT
Ünak,Perihan.
Magnetic nanoparticles offer exciting new opportunities including the improvement of the quality of magnetic resonance imaging (MRI), hyperthermic treatment for malignant cells, site-specific drug delivery and also the recent research interest of manipulating cell membranes. The biological applications of these nanomaterials require these nanoparticles to have high magnetization values, size smaller than 20 nm, narrow particle size distribution and a special surface coating for both avoiding toxicity and allowing the coupling of biomolecules. In this review, we focus on the feasibility of radionuclide labeled magnetic nanoparticles, as drug carriers, and summarize recent advances in this field.

Instructions for the Use of MInCir Scale to Assess Methodological Quality in Therapy Studies International Journal of Morphology
Manterola,Carlos; Cartes-Velasquez,Ricardo; Otzen,Tamara.
The assessment of methodological quality (MQ) in biomedical research is an area of dynamic development over recent years globally. Understood as a complex and multidimensional construct, various groups have proposed tools for its assessment. Our team has designed and validated scales to assess MQ of therapy, diagnosis, and prognosis studies. However, as with other instruments, it is necessary to specify in detail how it is applied, in order to standardize the measurements made with this instrument. The aim of this article is to provide a guideline for the standardized application of the MInCir MQ scale for therapy or therapeutic procedure studies. A detailed description of the 3 domains and 6 items comprising the scale, specifying for each of them how to...
