An antithrombin-dependent sulfated polysaccharide isolated from the green alga Caulerpa cupressoides has in vivo anti- and prothrombotic effects Ciência Rural
Rodrigues,José Ariévilo Gurgel; Queiroz,Ismael Nilo Lino de; Quinderé,Ana Luíza Gomes; Vairo,Bruno Cunha; Mourão,Paulo Antônio de Souza; Benevides,Norma Maria Barros.
Red algae sulfated polysaccharides (SPs) have been widely described as anticoagulant and antithrombotic agents; however no description of antithrombotic activity regarding green algae SPs has been reported. Caulerpa cupressoides (Chlorophyta) has three different SPs fractions (SP1, SP2 and SP3). We investigated the effects of SP2 on thrombin activity by antithrombin and in an experimental model of venous thrombosis in rats. The inhibition of thrombin assay was evaluated using antithrombin (AT) in the presence of SP2 and the antithrombotic activity was investigated in rats with thromboplastin as the thrombogenic stimulus. The anticoagulant effects of SP2 are suggested be due to the potentiation of thrombin inhibition by antithrombin (IC50 ~ 10.0µg mL-1) and...

Antithrombotic effect of Lonomia obliqua caterpillar bristle extract on experimental venous thrombosis BJMBR
Prezoto,B.C.; Maffei,F.H.A.; Mattar,L.; Chudzinski-Tavassi,A.M.; Curi,P.R..
The venom of Lonomia obliqua caterpillar may induce a hemorrhagic syndrome in humans, and blood incoagulability by afibrinogenemia when intravenously injected in laboratory animals. The possible antithrombotic and thrombolytic activities of L. obliqua caterpillar bristle extract (LOCBE) were evaluated in this study. The minimal intravenous dose of the extract necessary to induce afibrinogenemia and anticoagulation was 3.0 and 10.0 µg protein/kg body weight for rabbits and rats, respectively. In rabbits, this dose induced total blood incoagulability for at least 10 h and did not reduce the weight of preformed venous thrombi, in contrast to streptokinase (30,000 IU/kg). In rats, pretreatment with 5.0 and 10.0 µg/kg LOCBE prevented the formation of thrombi...

Caudal vena cava thrombosis in a dairy cow (Bos taurus) in Argentina Ciência Rural
Schild,Carlos; Armendano,Joaquín Ignacio; Liboreiro,Matías; Bresky,Florencia; Morrell,Eleonora; Odriozola,Ernesto; Cantón,Germán.
ABSTRACT: Caudal vena cava thrombosis (CVCT) is an acute disease secondary to ruminal lactic acidosis and rumenitis with fatal outcome in cattle fed with a high-grain diet. The aim of this paper is to describe CVCT in an adult Holstein cow from a herd in Balcarce, Buenos Aires, Argentina. Clinical signs included dyspnea, weakness, recumbence, and hemoptysis, followed by death in 3.5 days. Important necropsy findings included multiple hepatic abscesses, a septic thrombus (4.0cm x 2.0cm) in the vena cava adjacent to the liver, and a 15cm hematoma in the right lung. Histological lesions observed in the pulmonary parenchyma included suppurative pneumonia with embolic bacterial colonies and severe interstitial fibroplasia. Dissociation and degeneration of...

Effect of Low Molecular Weight Fucoidan and Low Molecular Weight Heparin in a Rabbit Model of Arterial Thrombosis ArchiMer
Durand, E; Helley, D; Zen, A; Dujols, C; Bruneval, P; Colliec-jouault, Sylvia; Fischer, Aurelie; Lafont, A.
Background: Therapeutic use of unfractionated heparin and low molecular weight heparins (LMWHs) is limited by hemorrhagic adverse effects. We compared the antithrombotic effect of LMW fucoidan (LMWF) and LMWH in an experimental model. Methods: Thrombosis was induced in femoral arteries of male New Zealand White rabbits by in situ induction of endothelial apoptosis with staurosporine (10(-5) M for 30 min). Starting the day before apoptosis induction, the animals received subcutaneous LMWF (15 mg/kg), LMWH (enoxaparin 2.5 mg/kg) or saline solution (control group) twice a day for 4 days. Results: The degrees of apoptosis and endothelial denudation were similar in the 3 groups. The thrombotic score was significantly lower in the LMWF group than in the LMWH and...

Estrogen replacement therapy and cardioprotection: mechanisms and controversies BJMBR
Subbiah,M.T.R..
Epidemiological and case-controlled studies suggest that estrogen replacement therapy might be beneficial in terms of primary prevention of coronary heart disease (CHD). This beneficial effect of estrogens was initially considered to be due to the reduction of low density lipoproteins (LDL) and to increases in high density lipoproteins (HDL). Recent studies have shown that estrogens protect against oxidative stress and decrease LDL oxidation. Estrogens have direct effects on the arterial tissue and modulate vascular reactivity through nitric oxide and prostaglandin synthesis. While many of the effects of estrogen on vascular tissue are believed to be mediated by estrogen receptors alpha and ß, there is evidence for `immediate non-genomic' effects. The role...

Increased expression of tissue factor and protease-activated receptor-1 does not correlate with thrombosis in human lung adenocarcinoma BJMBR
de Meis,E.; Azambuja,D.; Ayres-Silva,J.P.; Zamboni,M.; Pinheiro,V.R.; Levy,R.A.; Monteiro,R.Q..
A correlation between cancer and hypercoagulability has been described for more than a century. Patients with cancer are at increased risk for thrombotic complications and the clotting initiator protein, tissue factor (TF), is possibly involved in this process. Moreover, TF may promote angiogenesis and tumor growth. In addition to TF, thrombin seems to play a relevant role in tumor biology, mainly through activation of protease-activated receptor-1 (PAR-1). In the present study, we prospectively studied 39 lung adenocarcinoma patients in relation to the tumor expression levels of TF and PAR-1 and their correlation with thrombosis outcome and survival. Immunohistochemical analysis showed TF positivity in 22 patients (56%), most of them in advanced stages...

Plasmatic ADAMTS-13 metalloprotease and von Willebrand factor in children with cyanotic congenital heart disease BJMBR
Soares,R.P.S.; Bydlowski,S.P.; Nascimento,N.M.; Thomaz,A.M.; Bastos,E.N.M.; Lopes,A.A..
Changes in plasma von Willebrand factor concentration (VWF:Ag) and ADAMTS-13 activity (the metalloprotease that cleaves VWF physiologically) have been reported in several cardiovascular disorders with prognostic implications. We therefore determined the level of these proteins in the plasma of children with cyanotic congenital heart disease (CCHD) undergoing surgical treatment. Forty-eight children were enrolled (age 0.83 to 7.58 years). Measurements were performed at baseline and 48 h after surgery. ELISA, collagen-binding assays and Western blotting were used to estimate antigenic and biological activities, and proteolysis of VWF multimers. Preoperatively, VWF:Ag and ADAMTS-13 activity were decreased (65 and 71% of normal levels considered as 113...

Structural basis for the pathophysiology of lipoprotein(a) in the athero-thrombotic process BJMBR
Anglés-Cano,E..
Lipoprotein Lp(a) is a major and independent genetic risk factor for atherosclerosis and cardiovascular disease. The essential difference between Lp(a) and low density lipoproteins (LDL) is apolipoprotein apo(a), a glycoprotein structurally similar to plasminogen, the precursor of plasmin, the fibrinolytic enzyme. This structural homology endows Lp(a) with the capacity to bind to fibrin and to membrane proteins of endothelial cells and monocytes, and thereby to inhibit plasminogen binding and plasmin generation. The inhibition of plasmin generation and the accumulation of Lp(a) on the surface of fibrin and cell membranes favor fibrin and cholesterol deposition at sites of vascular injury. Moreover, insufficient activation of TGF-ß due to low plasmin...
