Acclimatization of 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstock Scientia Agricola

The pre-acclimatization stage can be used to improve micropropagation protocols and increase the yield of produced plants. The influence of sucrose and photon flux density (PFD) levels on the acclimatization of in vitro-grown 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstocks was evaluated. Rooted shoots were obtained from 4-week-old in vitro shoots cultivated in QL (Quoirin and Lepoivre, 1977) culture medium supplemented with 15, 30 and 45 g L-1 of sucrose. The experiment was kept in a 25 ± 2ºC growth room, under 16-h photoperiod and PFD of 18 µmol m-2 s-1 or 43 µmol m-2 s-1. Using genotypes adapted to the target region as a transgenic platform accelerates the development of cultivars. Therefore, this study aimed to adapt an in vitro regeneration protocol for Brazilian wheat genotypes. For this purpose, the in vitro regeneration capacity of immature embryos from six Brazilian wheat genotypes using two protocols of regeneration of somatic embryos was analysed. ABSTRACT Guava (Psidium guajava L.) can be propagated by seed, layering, air layering, grafting (budding or grafting), cuttings (root or shoot) or tissue culture. Propagation by seed is used for rootstock production and for raising populations for screening at early phases in the breeding programs. Vegetative propagation methods are used to clone selected genotypes from these programs and commercial orchards because it perpetuates all characteristics of each cultivar. This review addresses different methods that can be used to propagate guava, the methods commercially adopted and the progress obtained in recent years. The propagation of many passion fruit species (Passiflora spp.) is done by seeds or sexual method, although asexual propagation, such stem cuttings, grafting or tissue culture is also possible and very useful in many cases. In this work, it was presented information on scientific and technological advances in sexual and asexual propagation of passion fruit, considering the different propagation methods and their applications. New demands for research and development activities are also related, given the wide range of passion fruit species and its potential for food, ornamental and medicinal use.

ABSTRACT Nursery trees of stone fruits (Prunus spp.) are traditionally produced by union of two distinct genotypes - the rootstock and the scion - which, by grafting, form a composite plant that will be maintained throughout of all plant life. In Brazil, the rootstocks are predominantly seed propagated and therefore usually results in heterogeneous trees for vigor and edaphic adaptation. Altered calcium currents in cultured sensory neurons of normal and trisomy 16 mouse fetuses, an animal model for human trisomy 21 (Down Syndrome) Biol. Res.

Down syndrome is determined by the presence of an extra copy of autosome 21 and is expressed by multiple abnormalities, with mental retardation being the most striking feature. The condition results in altered electrical membrane properties of fetal dorsal root ganglia (DRG) neurons, as in the trisomy 16 fetal mouse, an animal model of the human condition. Cultured trisomic DRG neurons from human and mouse fetuses present faster rates of depolarization and repolarization in the action potential compared to normal controls and a shorter spike duration. An efficient protocol for culturing meristems of sorghum hybrids Phyton

A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. ANATOMICAL STUDIES OF IN VITRO ORGANOGENESIS INDUCED IN LEAF-DERIVED EXPLANTS OF PASSIONFRUIT PAB

With the aim of studying the organogenesis in vitro in Passiflora edulis Sims f. flavicarpa Deg., the passionfruit, leaf-derived explants were cultured on media containing NAA or BAP and incubated either in continuous darkness or in light. The histological events leading to de novo organ formation were evaluated. Darkness induces rhizogenesis in the presence of NAA, whereas direct shoot regeneration is stimulated by light and BAP. This latter condition is recommended for passionfruit micropropagation as several adventitious shoot buds were formed from meristemoids of parenchymal origin.

Anatomical study of Racosperma (ex-Acacia) mangium tissues cultured in vitro BABT

In order to achieve adventitious shoot regeneration from young tissues of Racosperma mangium, several combinations of growth regulators were tested. Kinetin (KIN) and a -naphthalenacetic acid (NAA), thidiazuron (TDZ) and NAA or indole-3-butyric acid (IBA), 4-CPPU and 2,4-dichlorophenoxyacetic acid (2,4-D) were added to basic medium composed of MS salts, MS vitamins and 3% sucrose. Sections of hypocotyls and cotyledons of young seedlings were cultivated on these media. Calli developed on different media and, after two to three months, some of them regenerated organized structures. Avaliacao da capacidade de regeneracao in vitro em tomateiro industrial. Repositório Alice

Este trabalho teve por objetivo avaliar a capacidade de regeneracao das cultivares de tomateiro industrial (Lycopersicon esculentum Mill) IPA-5 e IPA-6, utilizando quatro composicoes de meio de cultura descritos na literatura e cinco variacoes de inoculacao. Foi testada uma nova variacao de inoculacao, denominada cotiledone fendido. A maior frequencia de formacao de gemas vegetativas foi 100% no caso de IPA-5, e 65% no caso de IPA-6. Para induzir o alongamento de brotos, foram necessarios tres subcultivos dos explantes apresentando gemas. No caso de IPA-5, o numero de brotos obtidos foi maior quando a inducao de gemas foi realizada em meio contendo BAP e AIA seguido de tres subcultivos, em meio como zeatina. Endophytic microorganisms live inside healthy plants and their isolation and identification may favor biotechnological applications. We isolated endophytic bacteria from Ilex paraguariensis callus cultivated in vitro for five months. Sequencing of 16S rRNA region to identify two isolates as Bacillus megaterium was used. The presence of bacterial cells in the intercellular spaces of callus cultures was detected by ultra structural analyses. The isolates were also evaluated for indole acetic acid (IAA) production and their potential in plant growth promotion of Phaseolus vulgaris L. Bauhinia forficata link shoot regeneration: histological analysis of organogenesis pathway BABT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological studies indicated that in vitro plant regeneration in B. forficata occurred through indirect organogenesis. Biochemical and morphological responses to abiotc elicitor chitin in suspension-cultured sugarcane cells BABT

Cells of Saccharum officinarum submitted to hydrolyzated chitin for 1 to 8h produced phenolic compounds. These alterations were observed through cytochemical methods using Toluidine Blue and Phloroglucinol/HCl. After 4 h, besides cell wall change, there was a change in nuclear pattern of chitin treated cells. There was a 96% increase in nuclear area in 6 h chitin treated material, as observed by Feulgen reaction. The treated cells showed chromatin compacted regions and a degeneration process of nucleoli. In the outer areas of cell wall, there was a polysaccharide desagregation, confirming results obtained for different plants with the use of other elicitors.
