Dans le testicule d'un certain nombre de Poissons Téléostéens les cellules somatiques intra-lobulaires peuvent être considérées comme des cellules de Sertoli qui interviennent dans la contention et la nutrition des cellules germinales et dans leur phagocytose si elles dégénèrent. Les cellules de Sertoli assurent en outre, à la fin de la spermatogenèse, un rôle dans l'accumulation et l'excrétion de lipides qui participent vraisemblablement à la formation du liquide séminal. Malgré la présence passagère de réticulum agranulaire, cette étude morphologique ne permet pas de conclure définitivement au caractère stéroïdogène de ces cellules de Sertoli. Les phosphatases alcalines du testicule de Lebistes reticulatus : mise au point d'une microméthode de détermination quantitative Animal Physiology

L'utilisation de substrats chromogènes appropriés permet l'étude qualitative de l'équipement enzymatique des microorganismes. Nous avons appliqué cette méthode à la détermination quantitative des phosphatases alcalines du testicule de Lebistes reticulatus (poids moyen de l'organe : 2,5 mg). Cette microméthode de dosage enzymatique pourrait être généralisée à toute enzyme développant une coloration spontanée en présence d'un substrat approprié.

Les préparations hypophysaires en pisciculture. Obtention et emploi Animal Physiology

Le cycle reproducteur des poissons est sous la dépendance de l'hypophyse qui élabore et secrète les hormones gonadotropes. Ces hormones interviennent lors du déroulement de la gamétogenèse et sont produites activement pendant la maturation ovocytaire puis l'ovulation. La technique d'induction de ponte dite technique d'hypophysation consiste en un apport exogène d'hormone gonadotrope par injection d'hypophyses pour provoquer le déroulement de phases du cycle sexuel qui ne se produisent pas spontanément en condition de captivité comme l'ovulation.

LH secretion and ovulation following exposure of Arctic charr to different temperature and photoperiod regimes : Responsiveness of females to a gonadotropin releasing hormone analogue and a dopamine antagonist Inra

The timing of ovulation and LH plasma levels were investigated in Arctic charr reared at 5 °C and 10 °C, exposed to the ambient photoperiod, or short or long-day photoperiod regimes during the prespawning period. The effectiveness of sGnRHa alone, or sGnRHa combined with a dopamine antagonist, in stimulating LH secretion and inducing ovulation was also investigated. With the natural photoperiod, ovulation occurred spontaneously at 5 °C, but was inhibited at 10 °C. A transition from 10 to 5 °C soon resulted in suppression of the inhibition. Des expériences ont été conduites dans le but de définir la nature et la molarité du tampon à utiliser avec les deux dilueurs d'insémination et de conservation définis précédemment. Les effets des différents tampons ont été testés d'une part sur la stabilité du pH du dilueur après introduction de quantités croissantes de NaOH et HCl, de liquide cœlomique et d'ovules et d'autre part sur le pourcentage de fécondation établi sur de petits lots de 200 ovules. Les conclusions sont les suivantes : Le dilueur d'insémination (solution de NaCl à 250 mosm) peut être avantageusement tamponné avec un tampon carbonate-bicarbonate 0,02 M, pH de 9,2 à 9,5 Ce tampon se révèle plus stable après introduction de grande quantité d'ovules que les autres tampons testés...

Le méthallibure administré en bains à la dose de 2 p.p.m. entraîne chez le poisson rouge (Carassius auratus) une inhibition de la spermatogenèse qui intéresse toutes les cellules germinales à l'exception des spermatogonies A. Lorsque les mâles soumis à ce traitement reçoivent en même temps des injections de poudre hypophysaire brute de Carpe, la spermatogenèse est maintenue et même fortement stimulée. Maturation et ovulation in vitro des ovocytes de la truite arc-en-ciel Salmo gairdnerii Animal Physiology

Des extraits hypophysaires de Truite, Carpe, Saumon, provoquent la maturation (reprise de la méiose) mais non l'ovulation d'ovocytes incubés inclus dans du tissu ovarien. Divers progestagènes provoquent aussi la maturation, mais non l'ovulation d'ovocytes soit inclus dans le tissu ovarien, soit dissociés et extraits de ce tissu. L'incubation dans du liquide cœlomique, prélevé sur des femelles ayant spontanément ovulé, permet d'obtenir séparément, soit le processus de maturation sans ovulation, soit l'ovulation sans maturation. Les résultats tendent à montrer que la maturation ovocytaire et l'ovulation chez la Truite sont sous le contrôle direct de médiateurs différents agissant successivement.

Mise en évidence de quelques enzymes dans le sperme de la carpe (Cyprinus carpio L.) et de la truite (Salmo gairdnerii Richardson) et dans le liquide coelomique de la truite Animal Physiology

L'utilisation de la méthode Auxotab a permis la mise en évidence semi-quantitative de 16 enzymes dans les spermes de la Carpe et de la Truite, ainsi que dans le liquide cœlomique de la Truite. Il existe des différences d'équipement enzymatique sensibles entre le sperme total, le liquide séminal et les spermatozoïdes isolés ; elles portent essentiellement sur les phosphatases. Deux liquides cœlomiques dont l'activité biologique sur l'ovulation in vitro était connue, ont pu être différenciés par l'apparition d'une activité trypsique.

L'augmentation de la sécrétion gonadotrope (GtH) après castration chez la Truite arc-en-ciel est partiellement inhibée par du liquide séminal traité au charbon. Un extrait protéique testiculaire est inactif. Ce résultat laisse supposer l'existence dans le liquide séminal d'un facteur inhibiteur de la sécrétion gonadotrope. [The postcastration increase of gonadotropin secretion in the rainbow trout, was partially inhibited by charcoal treated seminal liquid. Protein testicular extract did not have any effect. This result demonstrates the probable existence of an inhibitory factor of gonadotropin secretion in the seminal plasma of the Trout.]

On the inhibitory effect of some steroids on spermatogenesis in adult rainbow trout (Salmo gairdneri) Animal Physiology

To test the antifertility effect of various steroids, fish were fed a diet containing steroids at two stages of the reproductive cycle: during spermatogenesis (June to November, experiment A) and during spermiation (November to February, experiment B). The fish in experiment A were given testosterone (T), methyltestosterone (MT), estradiol-17beta (E2), hexestrol (H), progesterone (Pg), and FGA (Fluorogestone acetate) at doses of 0.05 and (or) 0.5 mg/kg of food. In experiment B, T, MT, and E2 were given at a dose of 0.5 mg/kg of food. The fish received a daily ration amounting to 1% of their body weight. The use of hormonal factors in the induction of artificial egg laying was introduced in fish farming following the first works by Houssay in 1929. Since then, this technique has not evolved much. Pickford and Atz in 1957 have reviewed, exhaustively, most of the attempts carried out to date. This approach is still widely used, and in this way numerous hormonal concoctions have been put into use: mammalian hormones LH, FSH, HCG, PMS... and some fish hypophyseal extracts (mostly carp) often in a raw state. The hypophysation technique can be considered as finalized in the carp, through the use of homologous concoctions. However, in other species, success is more unpredictable and variable, depending on the nature of the hormones used.

Pituitary and plasma gonadotrophin levels and spermatogenesis in the goldfish Carassius auratus after methallibure treatment Animal Physiology

The level of gonadotrophin measured by radioimmunoassay in fully mature male goldfish was about 15 ng carp gonadotrophic hormone (c-HG)/ ml plasma and 30 microg c-HG/g pituitary (wet weight). After treatment for 35 days with methallibure (added to the water at the rate of 2 p.p.m./day), the level of gonadotrophin in the blood was very low and remained at the pretreatment level in the pituitary. This suggested that releasing factor might control either synthesis or discharge of gonadotrophin from the pituitary. These results have also shown that type B spermatogonia and meiosis were gonadotrophin dependent but that the development of spermatocytes into spermatids did not require high concentrations of gonadotrophin. Pituitary response to LH-RH has been studied in the male and female rainbow trout (Salmo gairdneri) at different stages of gametogenesis. The male was hardly sensitive at the onset of spermatogenesis, but a response occured at the spermatid stage and continued during spermatogenesis and spermiation. We particularly studied the end of the female cycle. Response was low in immature fishes and in the early stages of oocyte maturation, but became stronger at vitelline maturation. The relationships of these responses to plasma and pituitary t-GTH levels are discussed. Plasma estradiol-17beta and gonadotropin during ovulation in rainbow trout (Salmo gairdneri R.) Animal Physiology

A radioimmunoassay (RIA) for estradiol-17beta (E2-17beta) with a chromatographic step has been tested on rainbow trout plasma. Overloading and dilution tests gave satisfactory results. Intracardiac injection of E2-17beta (10 microg/kg) in July gave a peak in the first 30 min after injection ; return to the basic level occurred within the next 24 hrs. Using RIA, trout-gonadotropin (t-GTH) and E2-17beta were followed in trout plasma during the very last stages of the reproductive cycle. Postovulatory changes in the theca folliculi of the trout Animal Physiology

At time of ovulation the cells composing the theca folliculi of trout are morphologically smooth muscle like. The physiological behaviour of this tissue confirms its smooth muscle nature. Following ovulation a further cellular transformation and differentiation takes place : within 12-24 hrs after ovulation the theca cells start phagocytozing the adjacent collagen. Bundles of collagen are partially surrounded at first by cellular processes of the thecal cells while finally small bundles of collagen are interiorized. The number of collagen fibers per phagocytotic vesicle varies from one to many. Precocious induction of oocyte maturation and ovulation in Coho Salmon, Oncorhynchus kisutch Animal Physiology

Coho salmon (Oncorhynchus kisutch) were induced to ovulate 3-5 wk ahead of the normal time using partially purified salmon gonadotropin (SG-G100) alone or in combination with 17alpha hydroxy 20beta dihydroprogesterone (17alpha-20betaP). Treatment with 17alpha-20betaP alone induced final oocyte maturation but not ovulation and was associated with a negative feedback upon endogenous gonadotropin. The most successful treatments consisted of a "priming" injection of 0.1 mg/kg SG-G100 followed within 2-3 d by an injection of either 1 mg/kg SG-G100 or 3 mg/kg 17alpha-20betaP. Precocious induction of oocyte maturation and ovulation in rainbow trout (Salmo gairdneri) : problems when using 17 alpha-hydroxy-20 beta-dihydroprogesterone Animal Physiology

The efficiency of 17 alpha-hydroxy-20 beta-dihydroprogesterone (17 alpha-20 beta P) administered alone or after pituitary priming, was investigated in vivo before peripheral migration of the germinal vesicle 4 to 8 weeks prior to natural ovulation. Treatment with 17 alpha-20 beta P alone (2 injections of 3 mg/kg at a 2-day interval) induced oocyte maturation in 94 p. 100 of the fish, but only 25 p. 100 ovulated. Treatment with 17 alpha-20 beta P (3 mg/kg once) 2 days after pituitary priming (1 ml/kg of trout pituitary extract, TPE, containing 3.25 x 10exp-3 mg/ml of trout gonadotropin, t-GTH) induced maturation in all fish, 59 p. 100 of which ovulated. Purification of gonadotropin from rainbow trout (Salmo gairdnerii Richardson) pituitary glands Animal Physiology

A method of purifying trout gonadotropic hormone is discussed. The technique consists of the following stages : alcoholic extraction, two successive gel filtrations on Sephadex G-50 and G-100, chromatography on DEAE-cellulose and ultrogel ACA54. The biological activity of fractions obtained is measured using as assay the maturation in vitro of trout oocytes. The preparation obtained has a molecular weight of about 27500, determined by behavior on Sephadex-G 100 and analytical ultracentrifugation for a KS20w of 2.45 Polyacrylamide gel electrophoresis shows two strips of electrophoretic mobility, 0.50 and 0.58.
