Rapid determination of the sex of puberal salmonid fish by a technique of immunoagglutination Animal Physiology

The present study describes a rapid, simple and specific method to determine the sex of salmonid fish. This method is based on the detection by immunoagglutination of the serum vitellogenin of females in which oocytes are undergoing vitellogenesis. The application of this technique demonstrated the possibility of detecting vitellogenin in female brown trout as early as April and in all female Atlantic salmon captured in Brittany during the fishing season, between March and June.

Rhythms of reproduction in teleost fish Animal Physiology

Reproduction is a cyclic phenomenon which occurs periodically in fish , sometimes once every two or three years but more usually, once or several times a year. But if rhythm is defined as a succession of reproductive cycles, rhythmicity can hardly be an innate characteristic, since this cyclic reproductive activity is usually considered to result from adaptation to a changing environment mediated through the endocrine system. High temperatures inhibited goldfish (Carassius auratus) oogenesis at three different season(fall, winther, spring) in spite of an increase in plasma gonadotropin level. In winther, long photoperiod stimulated gonad growth. When fish were kept at a constant high temperature for 9 consecutive months from September to January, the gonads remained regressive during the first 4 months and then developed normally. Sexual differences in salmon gonadotropin Animal Physiology

Salmon (Oncorhynchus tschawytscha) GTH's were purified from a mixture of pituitaries of both sexes or from separate male and female glands. Some of their biochemical, immunological and biological properties have been compared. These hormones differed essentially in amino acid composition, especially lysine, histidine and half cystine residues, and by the presence of alpha-amino (or alpha-aminoiso) butyric acid in male and GTH II preparations. The same differences were found between GTH I-II and GTH male-female. Observed biochemical differences were sex-linked ; they did not influence immunological and qualitative biological effects, but could determine the affinity of the GTH of each sex or its specific receptors. Il existe une confusion importante dans la notion de spécificité d'action des hormones gonadotropes de Poissons. Cette notion a été précisée en utilisant deux méthodes différentes : d'une part la comparaison des activités biologiques de différents extraits hypophysaires de Poissons sur un même récepteur testiculaire, d'autre part la détermination de leur comportement immunologique vis-à-vis d'anticorps hautement spécifiques dirigés contre l'hormone gonadotrope de Carpe c-HG et o-LH. Stimulation de la sécrétion gonadotrope hypophysaire après castration chez la truite arc-en-ciel; variation de la réponse au cours du cycle reproducteur Animal Physiology

Chez la Truite arc-en-ciel ayant subi une castration bilatérale, le niveau de la gonadotropine circulante mesurée par radioimmunologie augmente, mais le type de réponse varie selon la période de l'année à laquelle l'opération est pratiquée. En mai, avant l'apparition du pic de sécrétion observé dans les conditions naturelles, l'augmentation des teneurs en gonadotropine après castration est lente et faible (8 ng/ml de plus que les témoins intacts après 30 jours). Elle est beaucoup plus rapide lorsque la castration est pratiquée fin mai, lors de l'apparition de ce pic (différence de 12 ng/ml après 13 jours). Ensuite, les différences par rapport aux témoins sont de 9 ng/ml après 12 jours en août (démarrage de la spermatogenèse), 10 ng/ml après 7 jours...

Stimulation hypophysaire de la sécrétion oestradiol-17beta chez la carpe commune, Cyprinus carpio L. [Hypophysial stimulation of oestradiol-17beta secretion in carp, Cyprinus carpio L.] Animal Physiology

[Using radioimmunoassays, plasma levels of carp gonadotropin (c-GTH) and oestradiol-17beta (E2) are measured every day in Carp, during induced spawning (water temperature = 15 °C). Ten Carps injected twice intraperitonealy with a total extract of Carp hypophysis (0,6 mg/kg BW at J0 and 5,4 mg/kg BW at J1) are compared to a lot receiving only saline solution. No evolution is detected in that last group when in the experimental one E2 level increases with exogenous c-GTH (see table). Even if E2 levels are low at the end of vitellogenesis, steroidogenic structures remain sensitive to an exogenous hypophysial stimulation. Secretion ability differs within individuals (correlation coefficient between total estimated c-GTH and E2 = 0,15).]

Stimulation of gonadotropin secretion after castration in rainbow trout Animal Physiology

The effects of bilateral castration on plasma gonadotropin (t-GtH), as measured by radioimmunoassay, were investigated in male rainbow trout at several stages in the reproductive cycle: at the start of spermatogenesis at the end of May, after its completion in September, and before and after the beginning of spermiation in October and December. Castration resulted in increased level of circulating immunoreactive t-GtH but the response varied with the season. The rise after castration was four times the initial value in May and September, and increased twofold in October and sevenfold in December. The duration of the response following castration was 4 months in May, 2 months in September, and less than 1 month in October and December.

Stimulation of milk synthesis in the rabbit by fish pituitary extract Animal Physiology

The lactogenic properties of extracts of the pituitary glands of salmon and trout were evaluated by using the organ culture technique with rabbit mammary explants. Crude extracts and fractions obtained after chromatography on Ultrogel and selected for their capacity to compete with ovine prolactin in a rabbit mammary gland radioreceptor assay were added to the culture medium. The criteria of lactogenesis were lactose synthetase activity, casein synthesis, measurements of the concentration of casein messenger RNA and the histology of mammary glands. All these tests led to the conclusion that salmon and trout pituitary glands contain a prolactin-like principle capable of initiating milk synthesis in the rabbit mammary cell.

During a 9-month period (corresponding to a sexual cycle), the adult male tench, Tinca tinca, was kept in fish farm ponds receiving heated water from a power plant. In 1974, the males were submitted to three different temperature regimes : group I : ambiant temperature ; group II : ambiant temperature + 3°C ; group III : ambiant temperature + 6°C. The experiment was repeated in 1975, but only using groups I and III. Female tench were bred for two consecutive years under different thermoperiods ; the effect of the thermoperiods on vitellogenesis, spawning, plasma and pituitary gonadotropin (GTH) levels was studied. The temperature did not affect development during the preparatory period. We found no ovarian development below 10°C. The temperature accelerated vitellogenesis only if the mean daily average was more than 10°C and the rate of increase of that parameter from 10 to 23°C (spawning temperature) determined female fertility. The date of first spawning may be predicted by the summation of degree days higher than 10°C. GTH secretion increased during vitellogenesis to reach maximal values during spawning. The annual reproduction cycle in adult carp in Poland: ovarian state and serum gonadotropin level Animal Physiology

In environmental conditions in Poland, new vitellogenesis began in carp just after the spawning season. This phase was correlated with the maintenance of a high serum, gonadotropin level, which decreased and remained low when the temperature was also low ; oogenesis remained blocked at the stage attained before the drop in temperature and gonadotropin level. The rise of temperature in May increased GTH and ovarian responsiveness to hypophysation treatment. Although pituitary function seemed normal in some fish, there was no spawning even after hypophysation treatment. Resorbing oocytes were present during the whole year, and a minimum rate of GTH secretion would be necessary to prevent massive resorption. Animals were ready for a new reproduction cycle...

Ultrastructural studies on experimentally induced vitellogenesis in juvenile rainbow trout (Salmo Gairdbneri R.) Animal Physiology

Juvenile rainbow trout (weighing 10 or 20 g) were treated thrice weekly for 10 weeks with salmon-gonadotropin (S-GTH), salmon pituitary extract (S-PE), S-GTH plus estradiol-17beta, and estradiol-17beta alone. The effects of these treatments on the oocytes were studied at ultrastructural level. Saline-injected control fish contained oocytes at previtellogenic stage of development. S-GTH. (0.1 or 0.5 microg/g) induced a synthesis of endogenous yolk in the oocyte cytoplasm but failed to initiate incorporation of exogenous yolk or vitellogenin into the oocytes. A somatostatin-like antigen has been localized in the brain and the digestive tract of the rainbow trout.
