An ELISA technique for rapid diagnosis of vibriosis in sea bass Dicentrarchus labrax ArchiMer

Three batches of rabbits were immunized against 2 strains of Vibrio anguillarum (Va V 62 and Va V 408), which cause vibriosis in sea bass Dicentrarchus labrax (L. 1758) Serranidae. Homologous and heterologous reactions were obtained against these Vibrio strains and other bacteria using an improved indirect ELISA technique. Antibody and antigen titrations revealed high immunogenecity (high antibody titers for the antisera) of the 2 Vibrio strains. They correspond to serotype I and show close affinity. The specificity and sensitivity of 1 selected antiserum were tested on brain extracts from healthy sea bass and those infected artificially with V. anguillarum serotypes I and III. The effect of density on sea bass (Dicentrarchus labrax) performance in a tank-based recirculating system ArchiMer

Sea bass (Dicentrarchus tabrax) (135 +/- 4 g) were reared under tank-based recirculating aquaculture system for a 63-day period at four densities: 10, 40, 70, 100 kg m(-3). Fish performance, stress indicators (plasma cortisol, proteonemia plus other blood parameters-Na+, K+, glucose, pH, total CO2- and water quality were monitored. At the end of the 63-day period, resistance to infection was also studied by a nodavirus challenge. A 25-day test was performed on fish from two extreme densities (10 and 100 kg m(3)) and one intermediate density (40 kg m(3)). The occurrence of maternal immunity transfer in Sea bass (Dicentrarchus labrax) was investigated using ELISA and Western blot technics. In order to determine the possible presence of immunoglobulin (Ig), egg proteins obtained from fertilized eggs were analyzed. After several extraction protocol assays (precipitation, chromatography), monoclonal and polyclonal antibody probes specific to Sea bass Ig allowed to confirm the presence of an immunoglobulin-like protein in eggs with very low level. Biochemical characterization was carried out using SDS-PAGE. Results indicated that egg Ig had a molecular weight of 206 kDa. They were smaller than seric IgM (800 kDa). Since 1986, an unknown disease causing high mortalities appeared in Ifremer's Pacific OceanologicaI Center (Tahiti) nursery of sea bass (Lates calcarifer). opathology of lesions involving both centraI nervous system (CNS) and eye retina, and the tuation exists in South-East Asian countries (Indonesia, Philippines, Malaysia and Singapore), and in northem Australia. This paper describes the histmorphology of a new fish virus, which is believed to be the aetiological agent of high mortalities among sea bass larvae.

Effects of rearing density on sea bass (Dicentrarchus labrax) biological performance, blood parameters and disease resistance in a flow through system ArchiMer

During 84 days, the effects of density on juvenile sea bass (Dicentrarchus labrax) (76 +/- 16 g) were studied in an experimental tank-based flow through system. Performance, stress response and resistance to virus infection were analysed under five stabilized rearing densities: 10, 20, 40, 70 and 100 kg m(-3). Water quality parameters (CO2, total ammonia nitrogen and O-2) were measured and maintained close to the recommended values for farmed sea bass by adjusting water renewal exchange. Reproduction of nodavirus disease was performed by experimental infection of sea bass eggs during fertilization or at larval stage 4 with 2 genetically distinguishable nodavirus strains (Sb1 and Sb2) isolated from sea bass collected along the Atlantic and Mediterranean French coast. The pathogenicity of the virus strains was assigned after detection of the virus by ELISA and immunohistochemistry (IHC). The Atlantic (Sb1) strain was more pathogenic than the Mediterranean (Sb2) strain during the fertilization step whilst both strains were pathogenic following experimental exposure of 4 d old larvae. In aquaculture management it is important to establish objective criteria to assess health and welfare of the fish. Here we show that European sea bass (Dicentrarchus labrax) confronted with husbandry-associated stress (tank cleaning, i.e. scrubbing, and water temperature variation) during early life stages show poorer survival and disease resistance as juveniles. We evaluated several parameters for stress (plasma cortisol, glucose and lactate, hydromineral status), growth performance, the immune response (plasma IgM levels) and the effects of a nodavirus challenge. A field report on vibrio disease of seabass (Dicentrarchus labrax ) in the South of France ArchiMer

Vibrio anguillarum was isolated from diseased seabass reared in aquatic farms (hatcheries and grow-out farms) located along the French Mediterranean coast. A representative strain (V62) differs from the French V. anguillarum strain (V408) by its ability to grow on a 0% salinity medium. The pathogenicity of the representative strain for sea bass was confirmed by injections and bath challenge. The V62 strain reacted with V. anguillarum antiserum (V408) and 84% of the isolated strains from diseased sea bass agglutinated with anti V62 serum.

Feed demand behavior in sea bass juveniles: Effects on individual specific growth rate variation and health (inter-individual and inter-group variation) ArchiMer

Feeding motivation is one major indicator of fish welfare and an investigation on the link between feed demand, growth and physiological variables in sea bass juveniles was developed. A computerized on-demand feeding system coupled with a PIT tag monitoring device was used to continuously record for 219 days the triggering activity of 150 individuals (initial average body weight 131.6 +/- 1.80 g and coefficient of variation 16.8%). Each group was held in 400 1 tanks at 22.2 +/- 1.5 degrees C and light regime was 16:8 LD. In all the tanks, 89% of the fish actuated the trigger, but only two or three fish accounted for 45% of the total triggering activity.
