Genetic structure and functioning of alien ship rat populations from a Corsican micro-insular complex Inra

The detrimental effects of the introduced ship rat on bird species on the Lavezzi Mediterranean archipelago has led to the decision to eradicate the rodent from the main island, Lavezzu (73 ha), as well as from several neighbouring islets. A genetic study using eight microsatellite markers has revealed some of the dynamics of this rat population. First, it has been shown that the rat population was genetically stable (no departure from Hardy-Weinberg equilibrium and no linkage disequilibrium) and as suggested by paleontological data, established for a long time. Characterization and PCR multiplexing of polymorphic microsatellite loci for the invasive ant Wasmannia auropunctata Inra

Highly polymorphic genetic markers provide a useful tool for estimating genetic parametersin studies of the evolution of sociality in insects. We isolated and characterized 12 polymorphicmicrosatellite loci in the invasive ant, Wasmannia auropunctata, and described experimental conditions for PCR (polymerase chain reaction) multiplexing and simultaneously genotyping these loci in two sets of five and seven markers. The number of alleles per locus ranged from two to 14 and the observed heterozygosity ranged from 0.233 to 0.967. Moreover, results of cross-species amplification tests are reported in three other species of Wasmannia and in two species of the genus Allomerus

Summary : Although the European rabbit (Oryctolagus cuniculus) is used both in agronomics and in research, genomic resources for this species are still limited and no microsatellite-based genetic map has been reported. Our aim was to construct a rabbit genetic map with cytogenetically mapped microsatellites so as to build an integrated genetic and cytogenetic map. A reference population of 187 rabbits comprising eight three-generation families with 10–25 offspring per family was produced. La variabilité génétique et la différenciation entre populations a été étudiée pour 11 locus microsatellites dans 15 populations de l’abeille ouest européenne. Les deux sous-espèces qui constituent ce rameau (Apis mellifera mellifera et A. m. iberica) ont une variabilité génétique plus réduite que la plupart des autres populations étudiées avec les mêmes outils moléculaires et plusieurs tests statistiques sont révélateurs d’une réduction récente de leur effectif. Olive cultivar origins are unknown. We attempted to use multivariate analyses and based Bayesian methods to look for sites of olive cultivar geographic origins. In this study, to establish the method, forty cultivars were chosen with different chlorotypes that represent the olive RAPD-based diversity in the Mediterranean basin. We constructed a dendrogram, as reference, using molecular SSR data. Cultivar origins in the oleaster were examined by using Bayesian clustering methods. Eight polymorphic microsatellite loci for the Australian plague locust, Chortoicetes terminifera Inra

Few population genetics studies have been carried out on major locust species. In particular, an understanding of the population genetic structure of the Australian plague locust, Chortoicetes terminifera, is lacking. We isolated and characterized eight polymorphic microsatellite loci in C. terminifera, and described experimental conditions for polymerase chain reaction multiplexing and genotyping these loci. Genome-wide effects of postglacial colonization in Arabidopsis lyrata Inra

The perennial outcrossing Arabidopsis lyrata is becoming a plant model species for molecular ecology and evolution. However, its evolutionary history, and especially the impact of the climatic oscillations of the Pleistocene on its genetic diversity and population structure, is not well known. We analyzed the broad-scale population structure of the species based on microsatellite variation at 22 loci. A wide sample in Europe revealed that glaciations and postglacial colonization have caused high divergence and high variation in variability between populations. Isolation and characterization of microsatellite loci from the entomopathogenic hyphomycete, Paecilomyces fumosoroseus Inra

Nine microsatellite markers were isolated from the entomopathogenic haploid fungus Paecilomyces fumosoroseus. Genetic diversity was assessed in 26P. fumosoroseus isolates originated from the whitefly Bemisia tabaci collected in various geoclimatic areas. Eight loci were polymorphic with an observed number of alleles ranging from two to six. The loci differentiated some isolates and group of isolates according to their geographical location, showing promise for the study of gene flow. All loci failed to give clear amplifications in P. fumosoroseus isolates from hosts other than B. tabaci. Characterization of 41 microsatellite loci developed from the genome sequence of the poplar rust fungus, Melampsora larici-populina Inra

Forty-one microsatellite markers were developed for the poplar rust fungus Melampsora larici-populina. The genome sequence of M. larici-populina strain 98AG31 was screened in silico for microsatellite loci using Magellan software. Thirty-nine loci, evaluated on a sexual population of 35 M. larici-populina isolates sampled on larch trees in the French Alps, were identified to be highly polymorphic. These markers revealed two to 34 alleles, with an average of 7.23 alleles per locus. The estimated heterozygosity ranged from 0.029 to 0.958 across the 41 microsatellite loci. These markers will be highly useful for population genetics studies of M. larici-populina.

L’organisation de la diversité génétique a été décrite dans le cas d’un peuplement mélangé de chêne sessile et pédonculé constitué de 355 arbres adultes répartis sur une surface de 6 ha. La diversité a été appréhendée à l’aide de 6 loci microsatellites manifestant un polymorphisme très élevé (de 15 à 32 allèles par locus). New polymorphic microsatellites loci, cross-species amplification and PCR multiplexing in the black Aphid, Aphis fabae Scopoli Inra

Aphis fabae includes four morphological cryptic subspecies, which are mostly identified by their partially distinct secondary host range. To determine the extent of gene flow and isolation between these four taxa, we isolated and characterized 12 microsatellite loci from Aphis fabae fabae and tested cross-species amplification of eight loci from the closely related species Aphis gossypii . Using eight previously described microsatellite loci, we have developed the polymerase chain reaction (PCR) multiplexing of 24 loci, which were separated in tree sets and five PCRs. Une grande partie des forêts européennes est dominée par les chênes (genre Quercus). Ces espèces ont longtemps été étudiées d’un point de vue taxonomique et écologique. Récemment, l’étude du polymorphisme de l’ADN chloroplastique a permis de mieux comprendre la taxonomie du genre ainsi que l’histoire de la recolonisation postglaciaire de ces espèces. Des flux de gènes intenses entre espèces ont été mis en évidence grâce au développement de marqueurs nucléaires très polymorphes (microsatellites). L’hybridation est un processus aux conséquences diverses sur l’évolution des espèces qui est difficile à étudier lorsque les espèces se distinguent mal au niveau morphologique. Isolation and characterization of microsatellites in the harlequin ladybird Harmonia axyridis (Coleoptera, Coccinellidae) and cross-species amplification within the family Coccinellidae Inra

A total of 18 microsatellite DNA loci were isolated and characterized from the harlequin ladybird, Harmonia axyridis (Coleoptera: Coccinellidae). We optimized a multiplex panel consisting of two polymerase chain reactions, allowing the genotyping of all loci. The number of alleles and heterozygosity observed at each locus ranged from 1 to 12 and from 0 to 100%, respectively. After Bonferroni correction for multiple tests, none of the loci deviated significantly from Hardy-Weinberg equilibrium and there was no indication of significant linkage disequilibrium among pairs of loci. Successful cross-species amplification was obtained for only three of the seven tested species of Coccinellidae

Twelve new polymorphic microsatellite loci and PCR multiplexing in the whitefly, Bemisia tabaci Inra

Twelve new dinucleotide microsatellite loci of the whitefly, Bemisia tabaci, were obtained from enriched genomic libraries. Three polymerase chain reaction multiplex sets comprising three, five and four loci were optimized and characterized across 133 B. tabaci females from Israeli rearings and natural populations collected in four Mediterranean countries (Tunisia, France, Spain and Morocco). There were three to 24 alleles per locus and the observed heterozygosity was from 0.084 to 0.420. Nothofagus is the main component of southern South American temperate forests. Overexploitation in the past has led to the loss of 40% of the original distribution range. Genetic diversity as well as biological processes shaping the distribution of the genetic variation (e.g. gene flow) constitutes basic knowledge for the implementation of conservation measures and for the definition of Evolutionary Significant Units. Nuclear microsatellites are the marker of choice for gene flow and fine-scale genetic structure studies. We enlarged a previous set of microsatellites (SSRs) for South American Nothofagus species, with special concern to Nothofagus nervosa (Phil.) Dim. et Mil. Five new SSRs are presented with allele numbers up to 12 in a single population....
