Floral biology and characterization of seed germination in physic nut (Jatropha curcas L.) Rev. bras. sementes

The objective of this study was to characterize morphologically the seed germination and floral biology of Jatropha curcas grown in Viçosa, Minas Gerais state. The floral biology study was made on fresh inflorescences of 20 plants. For the post-seminal development study, the seeds were submitted to laboratory and greenhouse germination test. J. curcas has flowers of both sexes within the same inflorescence, with each inflorescence having an average of 131 flowers, being 120 male and 10.5 female flowers. Low numbers of hermaphrodite flowers were also found, ranging from 0 to 6 flowers per inflorescence. The germination of J. curcas begins on the third day with radicle protrusion in the hilum region. The last few years have seen a significant increase in the number of large-scale sequencing projects generating whole genome databases. These sequence databases can be surveyed (genome sequence survey) for tandem repeats as an alternative means to develop microsatellites for monitoring and selecting natural populations and cultivars of Jatropha curcas. A total of 100 tandem repeats were revealed from mining 368 genomic surveyed sequences available in the Kazusa DNA Research Institute database. Twenty microsatellite sequences were successfully amplified, resulting in repeatable and scorable polymerase chain reaction products. There is currently great interest in the cultivation of the Jatropha curcas L., known locally as physic nut, for the production of biodiesel, leading to an increase in the demand for seeds and precise information as to their physiological quality, especially vigour. The objective was to evaluate the efficiency of the accelerated ageing test, with variations, in verifying the vigour of Jatropha seeds. J. curcas (physic nut) is one of the most promising oilseeds for biofuel production and the knowledge about genetic parameter estimates is important for its breeding. Here, we evaluated 121 families, from 48 to 60 months of age, aiming genetic evaluation and selection. The trial was set up in an 11 x 11 lattice design, with three replications and 4-plant plot. The traits evaluated were fruit yield (FY), grain (GY) and oil yield (OY), fruit husk percentage (FH) and seed oil content (OC). Individual heritabilities were low (0.19 to 0.23), requiring accurate selection and improvement methods. Physiological quality and seed respiration of primed Jatropha curcas seeds Acta Botanica

ABSTRACT Seed deterioration is a natural and irreversible process. Nevertheless, seed priming with water and antioxidants can minimize oxidative damage in oilseeds, resulting in attenuation of seed deterioration. The objective of this assay was to evaluate seed priming on respiratory activity of Jatropha curcas submitted to accelerated aging. Seeds from two provenances (Janauba and Pedro J. Caballero) were submitted to three priming treatments (control, immersion in deionized water, and with 750 µmol L-1 of ascorbic acid) and treated for accelerated aging at 41 °C for 72 h.  The objective of the present work was to investigate the antioxidant enzyme activities and the occurrence of lipid peroxidation during storage of J. curcas seeds obtained from fruits at different maturity stages. Seeds extracted from fruits collected at three stages of maturity were used, i.e., yellow, brownish-yellow, and brown (dry fruits). The seeds were subsequently stored for 18 months in Kraft paper bags in a laboratory environment. ABSTRACT Latex from Jatropha curcas L. exhibits biotechnological potential for the development of biodiesel and drugs. Little is known about the collection methods and latex productivity of this species. In order to garner information on the use of J. curcas latex, factors that influence production by this species were assessed. As a result, no significant difference was found between the volume of latex collected in the stem and the branches. With respect to environmental characteristics, climatic conditions, such as low temperatures and rainfall, affected production. Abstract Jatropha is an important genus of Euphorbiaceae, with species largely used for various purposes, including the manufacturing of soaps and pharmaceutical products and applications in the bioenergetic industry. Although there have been several studies focusing J. curcas in various aspects, the karyotype features of Jatropha species are poorly known. Therefore, we analyzed six Jatropha species through fluorochrome staining (CMA/DAPI), fluorescent in situ hybridization (FISH) with 5S and 45S rDNA probes and genome size estimation by flow cytometry.
