Effects of Ginseng on Rice Anther Culture Taiwan Agricultural Research Institute

[[abstract]]The effects of ginseng powder on anther browining, callus formation and organ regeneration of rice anher cultutre were studied. Results indicated that ginseng powder at 500-1,000 ppm reduced the rate of browning of anther culture and increased the callus-forming ability and breeding effeciency. 本試驗目的在探討培養基內添加人參粉末對水稻花藥培養之花藥褐化、癒傷組織形成及植株分化的影響。試驗結果發現,培養基內添加 500-1,000ppm 濃度的人參粉末能夠延緩花藥褐化速率,進而提高癒傷組織的形成能力及花藥培養之育種效率。

不同形式及濃度鐵源對水稻花藥培養之影響 Taiwan Agricultural Research Institute

[[abstract]]將水稻花藥培養於含不同形式及濃度鐵源的培養基,探討對水稻花藥癒合組織形成及分化能力的影響,及瞭解葉綠素含量與鐵源濃度及移值成活率的關係。試驗結果顯示添加0.1 mM 之Na2FeEDTA(二價鐵)或NaFeEDTA(三價鐵)於誘導癒合組織的培養基,有助於花藥癒合組織的形成,但提高濃度至0.2mM,反而產生毒害。 癒合組織於含0.05mM Na2FeEDTA或0.lmM NaFeEDTA或0.05mM Na2FeEDTA混合0.05mM NaFeEDTA 之培養基形成後,移至含相同鐵源之分化培養基,能分化較多的綠株;提高Na2FeEDTA的濃度至0.1 mM或0.2mM時,綠株分化率顯著降低。NaFeEDTA 在受測的三種濃度中,對綠株分化影響不大。在本試驗受測的濃度與形式範圍內,白苗的分化,不受鐵源影響。 分化綠株葉片之葉綠素含量與培養基內鐵源的形式及濃度有很大的關係,此種關係受到植株根系大小影響。約有85%之分化綠株,移植前具有發育中等之根系,其植株葉綠素含量與培養基內鐵源濃度呈正相關,同時葉綠素含量高的植株,具有較高的移植成活率。 Experiments were conducted to identify iron requirement in rice anther culture for callus induction, plant regeneration, chlorophyll content of anther-derived green plants and its survival ability. 低溫處理對水稻花藥接種適期之影響 Taiwan Agricultural Research Institute

[[abstract]]水稻稻穗經不同時間之低溫(10°C)處理後,選取含四分子期至晚單核期小孢子之花藥進行培養,探討低溫處理對花藥接種適期之影響。結果發現,各時期小孢子對低溫處理的反應差異極大。經七天之低溫處理,以中單核期小孢子之花藥最易形成癒合組織,且其誘導率較經低溫處理一天者提高一倍左右。低溫處理14天,可提高晚單核期小孢子之花藥形成癒合組織之能力,但對中單核期之花藥癒合組織誘導率則無提高之效果,過長之低溫處理(超過21天)反會抑制癒合組織之產生。 七天之低溫處理,會導致由四分子期小孢子之花藥所誘導的癒合組織喪失分化能力,短期(1─7天)之低溫處理,由中、晚單核期小孢子之花藥所形成的癒合組織,分化率有提高之現象,但較容易分化成白苗。超過14天之低溫處理,綠苗分化能力則顯著降低。 就育種效率而言,低溫處理七天以內,接種中單核期小孢子之花藥可得較高之育種效率;低溫處理14天則以接種晚單核期小孢子之花藥為佳,但其所分化綠株多為單倍體。超過21天之低溫處理,任何時期小孢子之花藥育種效率皆低於3%,而不適合進行培養。本試驗同時以二種貯存方式進行低溫處理,結果顯示,定期更換包裹於葉鞘基部之衛生紙,有助於花藥在長期低溫處理下維持較佳的活力。 The base of rice panicles with microspores at different developmental stages were wrapped with moist tissue paper and kept at 10°C for various durations to test the effect of cold shock on callus formation, organ regeneration and chromosome...
