A new view on aphid resistance in melon: the role of Aphis gossypii variability National Institute of Agronomic Research

Resistance to Aphis gossypii in Cucumis melo has been largely studied but A. gossypii variability has never been considered. Resistance to colonization by A. gossypii, clone NM1, and to non persistent virus transmission by this clone is conferred by a NBS-LRR gene called Vat, isolated in the PI 161375 accession. We investigated resistance to A. gossypii with four clones of A. gossypii that belong to two very distinct genotypes, NM1 and C9. The Vat gene conferred a high level of resistance to a NM1 clone and partial resistance to a C9 clone. La résistance génétiquement déterminée des plantes est un moyen efficace et respectueux de l’environnement de lutter contre les pucerons. Cependant, des biotypes de pucerons, capables de contourner les résistances, sont apparus dans plusieurs systèmes plantes–pucerons. Marker assisted selection of genes and QTLs for resistance combinations to Aphis gossypii in melon National Institute of Agronomic Research

The Vat gene in melon confers resistance to both aphid colonization and aphid transmission of several viruses. QTLs that provide resistance to aphid were localized on the melon genome. We built genetic allelic combinations with/without the resistant allele at the Vat locus and QTLs. These combinations (homozygous for the resistance loci) had a homogeneous genetic background for 90 % of the genome. Plants with different allelic combinations were infested by two Aphis gossypii genotypes NM1 and C9, and observed for acceptance and biotic potential. The Vat gene reduced the acceptance by both aphid genotypes, but only reduced the biotic potential of NM1. QTLs had a complementary effect on acceptance. Molecular diversity at the Vat/Pm-W resistance locus in melon National Institute of Agronomic Research

The resistance Vat gene of melon was isolated by map-based cloning. It encodes a protein with a coiled coil domain (CC), a nucleotide binding site (NBS) and leucine–rich repeats (LRR). Whereas most resistance genes confer resistance to a single pathogen, two alleles with different resistance specificities were identified at the locus. The Vat allele confers resistance to plant colonization by the aphid Aphis gossypii and to transmission of unrelated viruses by this vector specifically; the Pm-W allele confers resistance to the powdery mildew Podosphaera xanthii.
