Isolation and characterization of microsatellite loci from the entomopathogenic hyphomycete, Paecilomyces fumosoroseus Inra

Nine microsatellite markers were isolated from the entomopathogenic haploid fungus Paecilomyces fumosoroseus. Genetic diversity was assessed in 26P. fumosoroseus isolates originated from the whitefly Bemisia tabaci collected in various geoclimatic areas. Eight loci were polymorphic with an observed number of alleles ranging from two to six. The loci differentiated some isolates and group of isolates according to their geographical location, showing promise for the study of gene flow. All loci failed to give clear amplifications in P. fumosoroseus isolates from hosts other than B. tabaci. These microsatellite markers provide powerful tools for ecological, epidemiological, and population genetic studies

Microsatellite variability in the entomopathogenic fungus, Paecilomyces fumosoroseus: genetic diversity and population structure Inra

The hyphomycete Paecilomyces fumosoroseus (Pfr) is a geographically widespread fungus capable of infecting various insect hosts. The fungus has been used for the biological control of several important insect pests of agriculture. However knowledge of the fungus' genetic diversity and population structure is required for its sustainable use as a biological control agent. We investigated length and sequence polymorphisms of nine microsatellite loci for 33 Pfr accessions sampled from various host species and geographical locations, and our results reveal complex mutational processes for these molecular markers. Only Pfr isolates from Bemisia tabaci were amplified successfully, indicating the existence of Pfr genotypes specifically associated with B. tabaci....

Twelve new polymorphic microsatellite loci and PCR multiplexing in the whitefly, Bemisia tabaci Inra

Twelve new dinucleotide microsatellite loci of the whitefly, Bemisia tabaci, were obtained from enriched genomic libraries. Three polymerase chain reaction multiplex sets comprising three, five and four loci were optimized and characterized across 133 B. tabaci females from Israeli rearings and natural populations collected in four Mediterranean countries (Tunisia, France, Spain and Morocco). There were three to 24 alleles per locus and the observed heterozygosity was from 0.084 to 0.420. Deviation from Hardy–Weinberg equilibrium was detected at four loci associated with significant heterozygote deficiencies due to null alleles and presence of subpopulations that were mostly in the Tunisian sample.
