Le projet GAMETOGENES vise à améliorer les connaissances des bases physiologiques et génétiques de la reproduction et des métabolismes associés chez un mollusque d'importance économique l'huître creuse, Crassostrea gigas. Il a pour objectifs (1) la définition de marqueurs clés de la gamétogenèse, de l'investissement reproducteur et du déterminisme du sexe chez une espèce hermaphrodite à protandrie irrégulière (2) la recherche de QTL de survie aux mortalités estivales ou à l'effort de reproduction dont le succès repose sur l'augmentation de la densité de marqueurs sur les cartes génétiques existantes (SNP en particulier) et sur les liens qui pourront être établis entre ces cartes de liaison et une carte physique (3) le développement d'outils pour valider la...

Regulation of a truncated isoform of AMP-activated protein kinase alpha (AMPKalpha) in response to hypoxia in the muscle of Pacific oyster Crassostrea gigas ArchiMer

AMP-activated protein kinase α (AMPKα) is a key regulator of energy balance in many model species during hypoxia. In a marine bivalve, the Pacific oyster Crassostrea gigas, we analyzed the protein content of adductor muscle in response to hypoxia during 6 h. In both smooth and striated muscles, the amount of full-length AMP-activated protein kinase α (AMPKα) remained unchanged during hypoxia. However, hypoxia induced a rapid and muscle-specific response concerning truncated isoforms of AMPKα. In the smooth muscle, a truncated isoform of AMPKα was increased from 1 to 6 h of hypoxia, and was linked with accumulation of AKT kinase, a key enzyme of the insulin signaling pathway which controls intracellular glucose metabolism. The hermaphrodite Pacific oyster Crassostrea gigas displays a high energy allocation to reproduction. We studied the expression of AMP-activated protein kinase (AMPK) during gametogenesis in the gonad and characterized the mRNA sequences of the AMPK subunits: the AMPK alpha mRNA sequence was previously characterized; we identified AMPK beta, AMPK gamma, and mRNAs of putative AMPK-related targets following bioinformatics mining on existing genomic resources. The Kinome of Pacific Oyster Crassostrea gigas, Its Expression during Development and in Response to Environmental Factors ArchiMer

Oysters play an important role in estuarine and coastal marine habitats, where the majority of humans live. In these ecosystems, environmental degradation is substantial, and oysters must cope with highly dynamic and stressful environmental constraints during their lives in the intertidal zone. The availability of the genome sequence of the Pacific oyster Crassostrea gigas represents a unique opportunity for a comprehensive assessment of the signal transduction pathways that the species has developed to deal with this unique habitat.
