The development of a new sex determination test of Siberian sturgeon based on vitellogenin (VTG) detection in plasma led to identify this complex factor in plasma from male, female and immature fish of this species. The hypothesis of an estrogenic contamination from an environmental origin was confirmed by a test based on the feeding of fish on different diets. In control fish fed with a casein-based diet free of any estrogenic compound, VTG disappeared from the plasma within 15 weeks. With a fish meal-based diet as well as with a soy-based diet, the plasma VTG levels raised significantly. Later, an estrogenic effect of soy orally administrated was demonstrated in vivo in the rainbow trout although it is lower than that observed in sturgeon. Soya is known...

Generation of a large scale repertoire of Expressed Sequence Tags (ESTs) from normalised rainbow trout cDNA libraries Animal Physiology

Background: Within the framework of a genomics project on livestock species (AGENAE), we initiated a high-throughput DNA sequencing program of Expressed Sequence Tags (ESTs) in rainbow trout, Oncorhynchus mykiss. Results: We constructed three cDNA libraries including one highly complex pooled-tissue library. These libraries were normalized and subtracted to reduce clone redundancy. ESTs sequences were produced, and 96 472 ESTs corresponding to high quality sequence reads were released on the international database, currently representing 42.5% of the overall sequence knowledge in this species. Transcriptional analysis of testis maturation using trout cDNA macroarrays Animal Physiology

The project seeks to identify genes involved in key stages of trout spermatogenesis and their regulation. Within the framework of the French project of farm animal genomics (AGENAE) we produced an original normalised trout testis cDNA library and obtained 1152 trout ESTs corresponding to 967 potential genes. To study the expression of those genes throughout first stages of spermatogenesis, we used nylon macroarray. Gonads in stage of immaturity (stage I), or at initiation of spermatogonial proliferation (stage II), meiosis (stage III) or spermiogenesis were selected by histological analysis.
