Characterization of oligosaccharides in milk of a mink, Mustela vison OAK

Carbohydrates were extracted from a sample of milk from a mink, Mustela vison (Family Mustelidae). Free neutral and acidic oligosaccharides were isolated from the carbohydrate fraction and their chemical structures were compared with those of white-nosed coati (Nasua narica, Procyonidae) and harbour seal (Phoca vitulina, Phocidae) that we had studied previously. The ratio of free lactose to milk oligosaccharides was similar to that in milk of the white-nosed coati; in both species, this ratio was much lower than that in the milk of most eutherians. Characterization of two novel sialyl N-acetyllactosaminyl nucleotides separated from ovine colostrum OAK

The milk/colostrum of some mammalian species is known to contain sugar nucleotides including uridine diphosphate (UDP) oligosaccharides in addition to lactose and milk oligosaccharides, but the detailed structures of these UDP oligosaccharides have not so far been clarified. In this study we isolated two UDP-sialyl N-acetyllactosamines from ovine colostrum and characterized them using H-1-NMR and MALDITOFMS spectroscopies. Their structures were found to be Neu5Gc(alpha 2 - 3) Gal (beta 1 - 4)GlcNAc alpha 1-UDP and Neu5Gc(alpha 2-6) Gal(beta 1-4)GlcNAc alpha 1-UDP.

Chemical and Immunological Properties of Cell Wall Polysaccharide of Leuconostoc mesenteroides subsp. cremoris Isolated from the Commercial Fermented Milk,“Filmjolk" OAK

Cell wall polysaccharides were separated from Leuconostoc mesenteroides subsp. cremoris isolated from “Filmjolk",a fermented milk made commercially in Sweden. Two Components(CNPI and CNP2) were further purified by gelfiltration on HPLC and the molecu1ar weights were determined to be 850,000 and 81,000. CNP-2 was shown to contain two polysaccharides in major and minor by an lH-NMR. The major polysaccharide was shown to have a heptasaccharide repeating unit which contained five α and two β anomers,and also contained non reducing galactopyranose,O-2,3 disubstituted glucopyranose,O-3 substituted galactopyranose and O-3,6 disubstituted galactopyranose as units in a molar ratio of 3:1:1:2. Chemical characterization of milk oligosaccharides of the tiger quoll (Dasyurus maculatus), a marsupial OAK

Milk oligosaccharides were separated from the carbohydrate fraction of milk of the tiger quoll a species of marsupial that is closely related to the eastern quoll, Dasyurus viverrinus. They were characterized by H-1 - nuclear magnetic resonance spectroscopy and matrix - assisted laser desorption/ionization time-of-flight mass spectrometry. Effects of carbohydrate source on physicochemical properties of the exopolysaccharide produced by Lactobacillus fermentum TDS030603 in a chemically defined medium OAK

A thermophilic lactic acid bacterium, Lactobacillus fermentum TDS030603, produced about 100 mg/L of EPS in purified form when grew in de Man-Rogosa-Sharpe (MRS) broth. The 1% (w/v) solution of the purified EPS was highly viscous, exhibiting an apparent 22 viscosity (ηapp) of 0.88 Pa s at a shear rate of 10/s. To investigate the impact of carbohydrate source on the production yield and chemical structure of EPS and the viscosity of EPS solution, a chemically defined medium (CDM) has been developed. Immunoreactivity for Gonadotropin-Releasing Hormone in Microdialyzed Perfusates of Bovine Mature Follicles In Vitro OAK

The presence of gonadotropin-releasing hormone (GnRH)-like protein has been previously demonstrated in the bovine follicle. This GnRH-like protein was purified and concluded to be histone H2A. However, neither GnRH peptide nor specific GnRH-immunoreactivity (GnRH-IR) has been demonstrated in the bovine follicle so far. Sialyl Oligosaccharides in the Milk of Japanese Women: Changes in Concentration during the Course of Lactation OAK

The concentrations of the six major sialyl oligosaccharides found in the milk of 24 Japanese women were determined using reverse phase high performance liquid chromatography of the 3-methyl-1-phenyl-5- pyrazolone derivatives of the oligosaccharides. The milk was collected at 4, 10, 30 and 100 days postpartum. The concentrations of all six oligosaccharides decreased during the course of lactation. The concentrations of 6'-sialyllactose, sialyllacto-N-tetraose b and disialyllacto-N-tetraose decreased only after 10 days postpartum, whereas those of sialyllacto-N-tetraose a and sialyllacto-N-neotetraose c as well as 3'-sialyllactose decreased from 4 days postpartum. ペンシルバニア紀(>3億1000万年前)に,有羊膜類であるSynapsidは他の系統から分化し,腺を発達させた。乳腺は毛包と会合した祖先型アポクリン腺から発達した。乳腺と毛の関係は単孔類の乳腺に保持されている。また,有袋類における乳腺毛の初期個体発生時の存在とその後の退化に,その証拠が残されている。単孔類の乳頭のない乳腺区分にあるmammo-ピロ-脂腺単位の密集した集団は,透水性を示す卵への水分や他の成分の補給のための構造的進化を反映しているかもしれない。ラクトースなどの成分は,初期Synapsidの祖先アポクリン腺から分泌されていたかもしれない。三畳紀のTherapsidは,複雑な栄養成分を含む乳を分泌し,卵サイズを小さくし,孵化時の新生子の未熟化を引き起こした。乳腺区分に生じた乳頭は,乳腺の毛を退化させた。 乳腺分泌は,羊皮状の卵殻をもつ卵を産むSynapsidで開始されたと考えられる。羊皮状の卵殻をもつ卵は,低湿度の空気にふれた際,急速に水分損失を被る。Synapsidは抱卵嚢の中で抱卵することで卵の乾燥を防いだが,これは母親の行動を制限した。羊皮状の卵殻をもつ卵は,殻を通して液体水分を補給することができる。乳腺分泌は,本来卵に水分を供給する手段として開始され,Cynodont (キノドン類)において内温性獲得の要因となった。Synapsidの卵は,ナトリウムやカルシウムイオンなどの栄養素を摂取した可能性がある。孔のある卵殻と2層性の卵黄膜は,子宮内での分泌物と抱卵時での乳腺分泌物の摂取を促進したと考えられる。

Microflora of lactic acid bacteria and yeasts of Laban which had been made with bovine or ovine milk by traditional natural fermentation in Yemen,were studied. The mean of viable cell counts of microorganisms in Laban was 6.4 x10⁸ cfu/ml. The 60 strains of lactic acid bacteria and 24 strains of yeasts were isolated and identified. The percentages of strains on lactic acid bacteria flora were as follows: Lactococcus lactis subsp.lactis① was dominant in 88%. The activity of α3-D-mannoside-β-1,2-N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was detected in bovine follicular fluid (bFF). The GnT I activity in bFF had a pH optimum of 5.8 and an absolute requirement for either Co2+, Mn2+, or Mg2+, the activity being stimulated by these cations in the above order. The apparent Km value for α1-3α1-6 mannopentaose of GnT I in bFF was 2.17 mM. The substrate specificity for the GnT I activity decreased in the following order: α1-3α1-6 mannopentaose> α1-3α1-6 mannotriose>α1-3 mannobiose. Previous structural characterizations of marsupial milk oligosaccharides have been performed in the tammar wallaby, red kangaroo, koala, common brushtail possum and the eastern quoll. To clarify the homology and heterogeneity of milk oligosaccharides among marsupial species, which could provide information on their evolution, the oligosaccharides of wombat milk carbohydrate were characterized in this study. Neutral and acidic oligosaccharides were isolated from the carbohydrate fractions of two samples of milk of the common wombat and characterized by H-1-nuclear magnetic resonance spectroscopy. It is believed that human milk oligosaccharides (HMO) act as receptor analogues that inhibit the attachment of pathogenic microorganisms to the infant colon, but the direct involvement of HMO in anti infection has not as yet been demonstrated. The present study was conducted to clarify whether HMOs as well as commercially available galacto oligosaccharides (GO), from which lactose has been removed, affect the expression of toll-like receptor (TLR) 2 and 4, and of MD2 mRNA in the HT-29, human colonic cell line.
